    Increased collagen synthesis rate during wound healing in muscle

    Wound healing in muscle involves the deposition of collagen, but it is not known whether this is achieved by changes in the synthesis or the degradation of collagen. We have used a reliable flooding dose method to measure collagen synthesis rate in vivo in rat abdominal muscle following a surgical incision. Collagen synthesis rate was increased by 480% and 860% on days 2 and 7 respectively after surgery in the wounded muscle compared with an undamaged area of the same muscle. Collagen content was increased by approximately 100% at both day 2 and day 7. These results demonstrate that collagen deposition during wound healing in muscle is achieved entirely by an increase in the rate of collagen synthesis

    HIV testing history and preferences for future tests among gay men, bisexual men and other MSM in England: results from a cross-sectional study.

    OBJECTIVES: The British HIV Association's (BHIVA) testing guidelines recommend men who have sex with men (MSM) test annually or more frequently if ongoing risk is present. We identify which groups of MSM in England are less likely to have tested for HIV and their preferences for future tests by testing model, in order to inform health promotion programmes. METHODS: Data come from the Gay Men's Sex Survey 2014, a cross-sectional survey of MSM, aged 16 years or older and living in the UK. Only men who did not have diagnosed HIV and were living in England were included in this analysis. We used logistic regression models to understand how social determinants of health were associated with not testing for HIV in the past 12 months, and never having tested. We then cross-tabulated preferred testing location by demographic characteristics. RESULTS: Younger men, older men and men who were not gay identified were least likely to have tested for HIV. Higher educational attainment, migrancy, Black ethnicity and being at higher of risk were associated with greater levels of HIV testing. Men who were less likely to have tested for HIV preferred a wider range of options for future HIV testing. CONCLUSIONS: If the BHIVA's HIV testing policy of 2008 was used to guide testing priorities among MSM focus would be on increasing the rate of annual testing among MSM at less risk of HIV (ie, younger men, older men and non-gay identified MSM). Instead the promotion of more frequent testing among the groups most at risk of infection should be prioritised in order to reduce the time between infection and diagnosis

    Effects of co-treatment of Rauwolfia vomitoria and Gongronema latifolium on neurobehaviour and the neurohistology of the cerebral cortex in mice

    Rauwolfia vomitoria and Gongronema latifolium are medicinal plants with antioxidant, antidiabetic and analgesic properties among others. R. vomitoria is reported to possess adverse neural effects, which G. latifolium has shown the potential to address. This study therefore investigated the effects of co-treatment of R. vomitoria and G. latifolium on the neurobehaviour and histology of the cerebral cortex of female mice. Twenty female Wistar mice were divided into 4 groups (A, B, C and D). Group A designated as the control received 0.4 mL of 20 % Tween, while groups B, C and D received oral doses of 150 mg/kg of R. vomitoria (RV), 200 mg/kg of G. latifolium (GL) and a combination of 150 mg/kg of R. vomitoria and 200 mg/kg of G. latifolium (RV+GL), respectively for seven days. Light and dark field behaviour test was carried out on day 8 and the animals were immediately sacrificed. Their brains were excised and routinely processed by haematoxylin and eosin method. There was no difference in body and brain weights, and the behavioural parameters. Cellular cyto-architecture showed higher glial population with no apparent histopathology. The cellular population was higher (p<0.0001) in the RV and RV+GL groups, while the GL group was less (p<0.0001) populated all compared to the control.In conclusion, the reported treatment regimes, RV administered singly and in combination with GL may not affect some neurobehavioural activities, but may result in cellular increase in the cerebral cortex.KEY WORDS: Rauwolfia vomitoria; Gongronema latifolium; Cerebral cortex; Mic

    Predictors of crystal methamphetamine use in a community-based sample of UK men who have sex with men.

    Background Crystal methamphetamine (‘crystal meth’) use by men who have sex with men is an ongoing public health issue in the UK. We conducted a descriptive epidemiological study to characterise demographic and socio-sexual risk factors for crystal meth use in a national sample of UK MSM recruited in late 2014. Methods We used data from the 2014 Gay Men's Sex Survey (n = 16,565), an online community-based survey in the UK. We used logistic regression to relate risk factors to last-year use of crystal meth. Results In univariate models, crystal meth use was significantly associated with being between the ages of 30 and 49 (30–39, OR 2.24; 40–49, OR 2.21), living in London, having received a positive HIV test result (OR 7.37, 95% CI [6.28, 8.65]), and with higher education qualifications (1.40, [1.13, 1.75]), as well as with having multiple steady (2.15, [1.73, 2.68]) and non-steady (13.83, [10.30, 18.58]) partners with condomless anal intercourse. Relationships were similar in multivariate models, but education was no longer associated with last-year crystal meth use and lack of full-time employment was. Conclusions This analysis confirms and updates previous findings from the UK. Crystal meth use may now be more concentrated in London since previous surveys. This analysis presents novel findings regarding the association between number and sexual risk with partners and last-year meth use

    On the potential application of polar and temperate marine microalgae for EPA and DHA production

    Long chain polyunsaturated fatty acids (LC-PUFAs) such as eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) are considered essential omega-3 fatty acids in human nutrition. In marine microalgae EPA and/or DHA are allegedly involved in the regulation of membrane fluidity and thylakoid membrane functioning. The cellular content of EPA and DHA may therefore be enhanced at low temperature and irradiance conditions. As a result, polar and cold temperate marine microalgal species might potentially be suitable candidates for commercial EPA and DHA production, given their adaptation to low temperature and irradiance habitats. In the present study we investigated inter- and intraspecific EPA and DHA variability in five polar and (cold) temperate microalgae. Intraspecific EPA and DHA content did not vary significantly in an Antarctic (Chaetoceros brevis) and a temperate (Thalassiosira weissflogii) centric diatom after acclimation to a range of irradiance levels at two temperatures. Interspecific variability was investigated for two Antarctic (Chaetoceros brevis and Pyramimonas sp. (Prasinophyceae)) and three cold-temperate species (Thalassiosira weissflogii, Emiliania huxleyi (Prymnesiophyceae) and Fibrocapsa japonica (Raphidophyceae)) during exponential growth. Interspecific variability was shown to be much more important than intraspecific variability. Highest relative and absolute levels of DHA were measured in the prymnesiophyte E. huxleyi and the prasinophyte Pyramimonas sp., while levels of EPA were high in the raphidophyte F. japonica and the diatoms C. brevis and T. weissflogii. Yet, no significant differences in LC-PUFA content were found between polar and cold-temperate species. Also, EPA and DHA production rates varied strongly between species. Highest EPA production rate (174 μg L(-1) day(-1)) was found in the Antarctic diatom Chaetoceros brevis, while DHA production was highest in the cold-temperate prymnesiophyte Emiliania huxleyi (164 μg L(-1) day(-1)). We show that, following careful species selection, effective mass cultivation of marine microalgae for EPA and DHA production may be possible under low temperature and irradiance conditions

    Spatially and genetically distinct African trypanosome virulence variants defined by host interferon-g response

    We describe 2 spatially distinct foci of human African trypansomiasis in eastern Uganda. The Tororo and Soroti foci of <i>Trypanosoma brucei rhodesiense</i> infection were genetically distinct as characterized by 6 microsatellite and 1 minisatellite polymorphic markers and were characterized by differences in disease progression and host-immune response. In particular, infections with the Tororo genotype exhibited an increased frequency of progression to and severity of the meningoencephalitic stage and higher plasma interferon (IFN)–γ concentration, compared with those with the Soroti genotype. We propose that the magnitude of the systemic IFN-γ response determines the time at which infected individuals develop central nervous system infection and that this is consistent with the recently described role of IFN-γ in facilitating blood-brain barrier transmigration of trypanosomes in an experimental model of infection. The identification of trypanosome isolates with differing disease progression phenotypes provides the first field-based genetic evidence for virulence variants in T. <i>brucei rhodesiense</i>

    A two-stage mechanism of viral RNA compaction revealed by single molecule fluorescence

    Long RNAs often exist as multiple conformers in equilibrium. For the genomes of single-stranded RNA viruses, one of these conformers must include a compacted state allowing the RNA to be confined within the virion. We have used single molecule fluorescence correlation spectroscopy to monitor the conformations of viral genomes and sub-fragments in the absence and presence of coat proteins. Cognate RNA-coat protein interactions in two model viruses cause a rapid collapse in the hydrodynamic radii of their respective RNAs. This is caused by protein binding at multiple sites on the RNA that facilitate additional protein-protein contacts. The collapsed species recruit further coat proteins to complete capsid assembly with great efficiency and fidelity. The specificity in RNA-coat protein interactions seen at single-molecule concentrations reflects the packaging selectivity seen for such viruses in vivo. This contrasts with many in vitro reassembly measurements performed at much higher concentrations. RNA compaction by coat protein or polycation binding are distinct processes, implying that defined RNA-coat protein contacts are required for assembly
