    Isotopic evidence for long term warmth in the Mesozoic.

    PMCID: PMC3595697Atmospheric CO2 concentrations appear to have been considerably higher than modern levels during much of the Phanerozoic and it has hence been proposed that surface temperatures were also higher. Some studies have, however, suggested that Earth's temperature (estimated from the isotopic composition of fossil shells) may have been independent of variations in atmospheric CO2 (e.g. in the Jurassic and Cretaceous). If large changes in atmospheric CO2 did not produce the expected climate responses in the past, predictions of future climate and the case for reducing current fossil-fuel emissions are potentially undermined. Here we evaluate the dataset upon which the Jurassic and Cretaceous assertions are based and present new temperature data, derived from the isotopic composition of fossil brachiopods. Our results are consistent with a warm climate mode for the Jurassic and Cretaceous and hence support the view that changes in atmospheric CO2 concentrations are linked with changes in global temperatures

    Antigenic and functional characterization of p57 produced by Renibacterium salmoninarum

    Renibacterium salmoninarum, the causative agent of bacterial kidney disease, produces large quantities of a 57-58 kDa protein (p57) during growth in broth culture and during infection of salmonid fish. Biological activities of secreted p57 include agglutination of salmonid leucocytes and rabbit erythrocytes. We define the location of epitopes on p57 recognized by agglutination-blocking monoclonal antibodies (MAbs) 4C11, 4H8 and 4D3, and demonstrate that the majority of secreted p57 is a monomer that retains salmonid leucocyte agglutinating activity. The 3 MAbs bound a recombinant, amino-terminal fragment of p57 (211 aa) but not a carboxy-terminal fragment (315 aa) demonstrating that the neutralizing epitopes are located within the amino-terminal portion of p57. When combinations of the MAbs were used in an antigen capture ELISA, the epitopes recognized by the 3 MAbs were shown to be sterically separate. However, when the same MAb was used as both the coating and detection MAb, binding of the biotinylated detection MAb was not observed. These data indicate that the epitopes recognized by the 3 agglutination-blocking antibodies are functionally available only once per molecule and that native p57 exists as a monomer. Similar ELISA results were obtained when kidney tissues from 3 naturally infected chinook salmon were assayed. Finally, a p57 monomer was purified using anion exchange and size exclusion chromatography that retained in vitro agglutinating activity. A model in which p57 is released from R. salmoninarum as a biologically active monomer during infection of salmonid fish is proposed

    Adverse drug reactions among HIV infected and uninfected adults receiving anti-tuberculous therapy at Kenyatta National Hospital, Nairobi, Kenya

    Background: Information about the prevalence of adverse drug reactions (ADRs) among HIV infected and HIV uninfected patients receiving anti-tuberculous therapy in Africa is limited due to unavailability of local data or publications and hence the basis of this study.Objective: To determine the prevalence of adverse drug reactions among HIV infected and HIV uninfected adult patients on anti-TB therapy.Design: A retrospective cohort study.Setting: Kenyatta National Hospital, Kenya.Subjects: HIV infected and HIV uninfected patients receiving anti-TB therapy between January 2006 to December 2007Main Outcome Measures:Documented adverse drug reactions.Results: Three hundred and fourteen records were reviewed, 157 for both HIV infected and HIV uninfected. Of the 314 patient files, 96 (30.5%) had ADRs; 70 (44.6%) verses 26 (16.6%) for HIV infected and HIV uninfected respectively. Overall, the most frequent ADR among the two groups was gastrointestinal disturbances (21.7%) verses (10.2%) for HIV infected and uninfected respectively, (RR=2.44 [1.28-4.63], P=0.006). This was followed by peripheral neuropathy (16.6%) verses (4.5%) for HIV infected and uninfected respectively, (RR=4.25 [1.79-10.12], P=0.005). 73(46.49%) of the HIV infected patients were also receiving anti-retroviral therapy, ofwhich 36(49.31 %) of them had ADRs documented. Twenty five (29.8%) of the HIV infected who were not taking anti-retroviral therapy, had ADRs documented.Conclusions: Gastrointestinal disturbances and peripheral neuropathy were the most common ADRs in both groups. Surveillance systems should be established in hospitals for ADRs monitoring and control

    Development of a single droplet freezing apparatus for studying crystallisation in cocoa butter droplets

    The single droplet freezing apparatus described by Pore et al. (J. Am. Oil. Chem. Soc., 86, 215-225), which allows crystallisation to be monitored in situ by X-ray diffraction, was modified to allow rapid switching of coolant gas and monitoring by video microscopy. The apparatus was used to study drops of cocoa butter undergoing simulated spray freezing at high cooling rates, e.g. 130 K/min. The transformation of an Ivory Coast cocoa butter to the Form V polymorph was significantly faster in drops (~40 h) than in static bulk samples (10 days) crystallised under isothermal conditions. Phase transformation was observed from Forms I/II → III → IV → melt → V, with Form V crystallising directly from the melt at 28.6°C. Numerical simulations of the temperature evolution within the droplet established that the drops are not isothermal, explaining why nucleation was initially observed in the lower (upstream) part of the droplet.The provision of an EPSRC studentship for AMT and project support from Nestlé PTC York is gratefully acknowledged.     Robust identification of dynamically distinct regions in stratified turbulence

    We present a new robust method for identifying three dynamically distinct regions in a stratified turbulent flow, which we characterise as quiescent flow, intermittent layers and turbulent patches. The method uses the cumulative filtered distribution function of the local density gradient to identify each region. We apply it to data from direct numerical simulations of homogeneous stratified turbulence, with unity Prandtl number, resolved on up to 8192×8192×40968192\times 8192\times 4096 grid points. In addition to classifying regions consistently with contour plots of potential enstrophy, our method identifies quiescent regions as regions where \unicode[STIX]{x1D716}/\unicode[STIX]{x1D708}N^{2}\sim O(1), layers as regions where \unicode[STIX]{x1D716}/\unicode[STIX]{x1D708}N^{2}\sim O(10) and patches as regions where \unicode[STIX]{x1D716}/\unicode[STIX]{x1D708}N^{2}\sim O(100). Here, \unicode[STIX]{x1D716} is the dissipation rate of turbulence kinetic energy, \unicode[STIX]{x1D708} is the kinematic viscosity and NN is the (overall) buoyancy frequency. By far the highest local dissipation and mixing rates, and the majority of dissipation and mixing, occur in patch regions, even when patch regions occupy only 5 % of the flow volume. We conjecture that treating stratified turbulence as an instantaneous assemblage of these different regions in varying proportions may explain some of the apparently highly scattered flow dynamics and statistics previously reported in the literature.The research activities of G.D.P. and S.dB.K. were funded by the US Office of Naval Research via grant N00014-15-1-2248.     On the Impact of Link Layer Retransmissions on TCP for Aeronautical Communications

    In this article, we evaluate the impact of link layer retransmissions on the performance of TCP in the context of aeronautical communications.We present the architecture of aeronautical networks, which is manly driven by an important channel access delay, and the various retransmission strategies that can be implemented at both link and transport layers. We consider a worst case scenario to illustrate the benefits provided by the ARQ scheme at the link layer in terms of transmission delay.We evaluate the trade-off between allowing a fast data transmission and a low usage of satellite capacity by adjusting link layer parameters

    Role of the mesoamygdaloid dopamine projection in emotional learning

    Amygdala dopamine is crucially involved in the acquisition of Pavlovian associations, as measured via conditioned approach to the location of the unconditioned stimulus (US). However, learning begins before skeletomotor output, so this study assessed whether amygdala dopamine is also involved in earlier 'emotional' learning. A variant of the conditioned reinforcement (CR) procedure was validated where training was restricted to curtail the development of selective conditioned approach to the US location, and effects of amygdala dopamine manipulations before training or later CR testing assessed. Experiment 1a presented a light paired (CS+ group) or unpaired (CS- group) with a US. There were 1, 2 or 10 sessions, 4 trials per session. Then, the US was removed, and two novel levers presented. One lever (CR+) presented the light, and lever pressing was recorded. Experiment 1b also included a tone stimulus. Experiment 2 applied intra-amygdala R(+) 7-OH-DPAT (10 nmol/1.0 A mu l/side) before two training sessions (Experiment 2a) or a CR session (Experiment 2b). For Experiments 1a and 1b, the CS+ group preferred the CR+ lever across all sessions. Conditioned alcove approach during 1 or 2 training sessions or associated CR tests was low and nonspecific. In Experiment 2a, R(+) 7-OH-DPAT before training greatly diminished lever pressing during a subsequent CR test, preferentially on the CR+ lever. For Experiment 2b, R(+) 7-OH-DPAT infusions before the CR test also reduced lever pressing. Manipulations of amygdala dopamine impact the earliest stage of learning in which emotional reactions may be most prevalent

    Fiber Acoustic Waveguide : A Sensor Candidate

    Sensor development plays a key role in the field of nondestructive evaluation and process control. The annual fiber optic sensor market alone is a multimillion dollar business (1). Acoustic waves are about five orders of magnitude slower than optical waves and can also be guided in cladded glass fibers, similar to optical fibers, with low loss and low dispersion (2–7). Fiber acoustic waveguides are believed to be a very attractive and basic component for further sensor development (8). In this paper a brief theoretical description of a weakly guiding acoustic fiber (7) is given. The material selection criteria for the core and the cladding of the fiber guide, the properties of single-mode operation, and some sensing mechanisms for temperature and pressure variations are discussed. The acoustic waveguide with a liquid core is also considered
