Urbanicity and lifestyle risk factors for cardiometabolic diseases in rural Uganda: a cross-sectional study
Urban living is associated with unhealthy lifestyles that can increase the risk of cardiometabolic diseases. In sub-Saharan Africa (SSA), where the majority of people live in rural areas, it is still unclear if there is a corresponding increase in unhealthy lifestyles as rural areas adopt urban characteristics. Trauma Immediately Preceding REM-Behavior Disorder:A Valuable Prognostic Marker?
Background: The definition of rapid eye movement (REM) sleep behavior disorder (RBD) has varied over the years. Rapid eye movement sleep behavior disorder can be considered isolated or idiopathic or can occur in the context of other disorders, including trauma-associated sleep disorder (TSD) and overlap parasomnia. However, whether trauma in RBD carries any prognostic specificity is currently unknown.
Study Objectives: To test the hypothesis that RBD secondary to trauma is less likely to result in the development of neurodegeneration compared to idiopathic RBD (iRBD) without trauma in the general population.
Methods: A retrospective cohort study of 122 consecutive RBD patients (103 males) at two tertiary sleep clinics in Europe between 2005 and 2020 was studied. Patients were diagnosed as having iRBD by video polysomnography (vPSG) and had a semi-structured interview at presentation, including specifically eliciting any history of trauma. Patients with secondary RBD to recognized causes were excluded from the study. Patients with iRBD were categorized into three groups according to reported trauma history: (1) No history of trauma, (2) traumatic experience at least 12 months prior to RBD symptom onset, and (3) traumatic experience within 12 months of RBD symptom onset. Idiopathic RBD duration was defined as the interval between estimated onset of RBD symptoms and last hospital visit or death. Follow-up duration was defined as the interval between iRBD diagnosis and last hospital visit or death.
Results: In a follow-up period of up to 18 years, no patient who experienced trauma within 12 months preceding their iRBD diagnosis received a diagnosis of a neurodegenerative disorder (n = 35), whereas 38% of patients without trauma within the 12 months of symptom onset developed a neurodegenerative illness. These patients were also significantly more likely to have a family history of α-synucleinopathy or tauopathy.
Conclusions: The development of RBD within 12 months of experiencing a traumatic life event, indistinguishable clinically from iRBD, did not lead to phenoconversion to a neurodegenerative disorder even after 18 years (mean follow up 6 years). We suggest that a sub-type of RBD be established and classified as secondary RBD due to trauma. Additionally, we advocate that a thorough psychological and trauma history be undertaken in all patients presenting with dream enactment behaviors (DEB)

Moving Beyond Pump and Treat Toward Enhanced Attenuation and Combined Remedies T-Area, Savannah River Site
Groundwater beneath T-Area, a former laboratory and semiworks operation at the Department of Energy (DOE) Savannah River Site, is contaminated by chlorinated solvents (cVOCs). Since the contamination was detected in the 1980s, the cVOCs at T-Area have been treated by a combination of soil vapor extraction and groundwater pump and treat. The site has received approval to discontinue the active treatments and implement a full scale test of enhanced attenuation--an engineering and regulatory strategy that has recently been developed by DOE and the Interstate Technology and Regulatory Council. Enhanced attenuation uses active engineering solutions to alter the target site in such a way that the contaminant plume will passively stabilize and shrink and to document that the action will be effective, timely, and sustainable. The paradigm recognizes that attenuation remedies are fundamentally based on a mass balance. Thus, long-term plume dynamics can be altered either by reducing the contaminant loading from the source or by increasing the rate of natural attenuation processes within all, or part of, the plume volume. The combination of technologies that emerged for T-Area included: (1) neat (pure) vegetable oil deployment in the deep vadose zone in the former source area, (2) emulsified vegetable oil deployment within the footprint of the groundwater plume, and (3) identification of attenuation mechanisms and rates for the distal portion of the plume. In the first part, neat oil spreads laterally forming a thin layer on the water table to intercept and reduce future cVOC loading (via partitioning) and reduce oxygen inputs (via biostimulation). In the second and third parts, emulsified oil forms active bioremediation reactor zones within the plume footprint to degrade existing groundwater contamination (via reductive dechlorination) and stimulates long-term attenuation capacity in the distal plume (via cometabolism). For T-Area, the enhanced attenuation development process proved to be a powerful tool in developing a defensible strategy that provides a high degree of performance while minimizing adverse collateral impacts of remediation (e.g., energy use and wetland damage) and minimizing life-cycle costs
LABORATORY INVESTIGATIONS IN SUPPORT OF FLUID BED FLUORIDE VOLATILITY PROCESSES. PART IV. THE FLUID BED FLUORINATION OF UO
In one of the processes under development, the uranium and plutonium content of a spent reactor fuel of the Dresden type would be fluorinated in a fluid-bed reactor to produce the volatile uranium and plutonium hexafluorides. The study of the fluorination of U/sub 3/O/sub 8/ was undertaken because it is the major product obtained in an oxidative decladding step that is being proposed for the removal of uranium and plutonium from stainless steel-clad and Zircaloy- clad fuel elements. The study Will also provide data needed for the development of apparatus and procedures for future work with mixtures of uranosic oxide and plutoniam dioxide. Experiments were performed to determine the optimum reaction conditions for the conversion of U/sub 3/O/sub 8/ to uranium hexafluoride and for minimizing the elutriation of unreacted U/sub 3/O/sub 8/ from the fluid bed. Elutriation of U/sub 3/O/sub 8/ from the fluid bed was minimized when a fluid bed height of 8 in. of 120 mesh alumina, a fluorinating gas phase containing 20 vol% fluorine, and a reaction temperature of 500 deg C were employed. Conversion of greater than 99% of the U/sub 3/O/sub 8/ to uranium hexafluoride was obtained when the feeding-fluorination period, in w,hich the U/sub 3/O/sub 8/ powder was fed into the fluid bed and the major part of the fluorination was accomplished by reacting the oxide with 20 vol % fluorine, was followed by a recycle-fluorination period of 5 hr at 500 deg C with 100% fluorine. Kinetic data for the fluorination of U/sub 3/O/sub 8/, obtained by means of a thermobalance, are reported for the temperature range from 300 to 400 deg C. siRNA–Mediated Methylation of Arabidopsis Telomeres
Chromosome termini form a specialized type of heterochromatin that is important for chromosome stability. The recent discovery of telomeric RNA transcripts in yeast and vertebrates raised the question of whether RNA–based mechanisms are involved in the formation of telomeric heterochromatin. In this study, we performed detailed analysis of chromatin structure and RNA transcription at chromosome termini in Arabidopsis. Arabidopsis telomeres display features of intermediate heterochromatin that does not extensively spread to subtelomeric regions which encode transcriptionally active genes. We also found telomeric repeat–containing transcripts arising from telomeres and centromeric loci, a portion of which are processed into small interfering RNAs. These telomeric siRNAs contribute to the maintenance of telomeric chromatin through promoting methylation of asymmetric cytosines in telomeric (CCCTAAA)n repeats. The formation of telomeric siRNAs and methylation of telomeres relies on the RNA–dependent DNA methylation pathway. The loss of telomeric DNA methylation in rdr2 mutants is accompanied by only a modest effect on histone heterochromatic marks, indicating that maintenance of telomeric heterochromatin in Arabidopsis is reinforced by several independent mechanisms. Comparison of different methods for delayed post-mortem diagnosis of falciparum malaria
<p>Abstract</p> <p>Background</p> <p>Between 10,000 and 12,000 cases of imported malaria are notified in the European Union each year. Despite an excellent health care system, fatalities do occur. In case of advanced autolysis, the post-mortem diagnostic is impaired. Quicker diagnosis could be achieved by using rapid diagnostic malaria tests.</p> <p>Methods</p> <p>In order to evaluate different methods for the post-mortem diagnosis of <it>Plasmodium falciparum </it>malaria in non-immunes, a study was performed on the basis of forensic autopsies of corpses examined at variable intervals after death in five cases of fatal malaria (with an interval of four hours to five days), and in 20 cases of deaths unrelated to malaria. Detection of parasite DNA by PCR and an immunochromatographic test (ICT) based upon the detection of <it>P. falciparum </it>histidine-rich protein 2 (PfHRP2) were compared with the results of microscopic examination of smears from cadaveric blood, histopathological findings, and autopsy results.</p> <p>Results</p> <p>In all cases of fatal malaria, post-mortem findings were unsuspicious for the final diagnosis, and autoptic investigations, including histopathology, were only performed because of additional information by police officers and neighbours. Macroscopic findings during autopsy were unspecific. Histopathology confirmed sequestration of erythrocytes and pigment in macrophages in most organs in four patients (not evaluable in one patient due to autolysis). Microscopy of cadaveric blood smears revealed remnants of intraerythrocytic parasites, and was compromised or impossible due to autolysis in two cases. PCR and ICT performed with cadaveric blood were positive in all malaria patients and negative in all controls.</p> <p>Conclusion</p> <p>In non-immune fatalities with unclear anamnesis, ICT can be recommended as a sensitive and specific tool for post-mortem malaria diagnosis, which is easier and faster than microscopy, and also applicable when microscopic examination is impossible due to autolysis. PCR is more expensive and time-consuming, but may be used as confirmatory test. In highly endemic areas where asymptomatic parasitaemia is common, confirmation of the diagnosis of malaria as the cause of death has to rely on histopathological findings.</p

POTENTIAL ENHANCEMENTS TO NATURAL ATTENUATION: LINES OF INQUIRY SUPPORTING ENHANCED PASSIVE REMEDIATION OF CHLORINATED SOLVENTS
The Department of Energy (DOE) is sponsoring an initiative to facilitate efficient, effective and responsible use of Monitored Natural Attenuation (MNA) and Enhanced Passive Remediation (EPR) for chlorinated solvents. This Office of Environmental Management (EM) ''Alternative Project,'' focuses on providing scientific and policy support for MNA/EPR. A broadly representative working group of scientists supports the project along with partnerships with regulatory organizations such as the Interstate Technology and Regulatory Council and the U.S. Environmental Protection Agency (EPA). The initial product of the technical working group was a summary report that articulated the conceptual approach and central scientific tenants of the project, and that identified a prioritized listing of technical targets for field research. This report documented the process in which: (1) scientific ground rules were developed, (2) lines of inquiry were identified and then critically evaluated, (3) promising applied research topics were highlighted in the various lines of inquiry, and (4) these were discussed and prioritized. The summary report will serve as a resource to guide management and decision-making throughout the period of the subject MNA/EPR Alternative Project. To support and more fully document the information presented in the summary report, we are publishing a series of supplemental documents that present the full texts from the technical analyses within the various lines of inquiry (see listing). The following report - documenting our evaluation of the state of the science of the characterization and monitoring process and tools-- is one of those supplemental documents
The Candida albicans Ku70 Modulates Telomere Length and Structure by Regulating Both Telomerase and Recombination
The heterodimeric Ku complex has been shown to participate in DNA repair and telomere regulation in a variety of organisms. Here we report a detailed characterization of the function of Ku70 in the diploid fungal pathogen Candida albicans. Both ku70 heterozygous and homozygous deletion mutants have a wild-type colony and cellular morphology, and are not sensitive to MMS or UV light. Interestingly, we observed complex effects of KU70 gene dosage on telomere lengths, with the KU70/ku70 heterozygotes exhibiting slightly shorter telomeres, and the ku70 null strain exhibiting long and heterogeneous telomeres. Analysis of combination mutants suggests that the telomere elongation in the ku70 null mutant is due mostly to unregulated telomerase action. In addition, elevated levels of extrachromosomal telomeric circles were detected in the null mutant, consistent with activation of aberrant telomeric recombination. Altogether, our observations point to multiple mechanisms of the Ku complex in telomerase regulation and telomere protection in C. albicans, and reveal interesting similarities and differences in the mechanisms of the Ku complex in disparate systems
