    THE EFFECT OF SOME PLANT GROWTH REGULATORS AND THEIR COMBINATION WITH METHYL JASMONATE ON ANTHOCYANIN FORMATION IN ROOTS OF KALANCHOE BLOSSFELDIANA

    ABSTRACT In this study, we investigated the effect of plant growth regulators (PGRs) -auxins, gibberellin, cytokinin, abscisic acid, brassinosteroid, ethylene and their interaction with methyl jasmonate (JA-Me) applied to roots of the whole plants Kalanchoe blossfeldiana on the accumulation of anthocyanins in roots. The highest stimulation of anthocyanins synthesis was stated with application of JA-Me alone. In response to treatments with the other tested PGRs, the content of anthocyanins in roots of a whole plant was different depending on the concentration of the PGR when being applied alone or together with JA-Me. Auxin, indole-3-acetic acid (IAA) at a concentration of 50 mg·L -1 , indole-3-butyric acid (IBA) at 5 mg·L -1 and abscisic acid (ABA) at 10 mg·L -1 induced anthocyanin accumulation with approximately 60-115% compared to the control while 24-epibrassinolid (epiBL), gibberellic acid (GA3) and 6-benzylaminopurine (BAP) had no effect on the anthocyanin accumulation. The simultaneous administration of the PGRs with JA-Me usually resulted in the accumulation of anthocyanins in roots in a manner similar to that caused by JA-Me. PGRs applied to isolated roots did not stimulate anthocyanin accumulation, except for the combination of JA-Me with 50 mg·L -1 IAA. The results indicate that in K. blossfeldiana, the aboveground parts of the plant play an important role in the biosynthesis of anthocyanins in roots

    Differential effects of auxin polar transport inhibitors on rooting in some Crassulaceae species

    Effects of auxin polar transport inhibitors, 2,3,5-triio-dobenzoic acid (TIBA), 1-N-naphthylphthalamic acid (NPA) and methyl 2-chloro-9-hydroxyfluorene-9-carboxylate (morphactin IT 3456), as a lanolin paste, on root formation in cuttings of some species of Crassulaceae, such as Bryophyllum daigremontianum, B. calycinum, Kalanchoe blossfeldiana and K. tubiflora, were studied. Cuttings of these plants were easily rooted in water without any treatment. TIBA and morphactin IT 3456 completely inhibited root formation in the cuttings of these plants but NPA did not when these inhibitors were applied around the stem below the leaves. When TIBA and morphactin were applied around the stem near the top, but leaves were present below the treatment, the root formation was observed in B. calycinum and K. blossfeldiana but in a smaller degree than in control cuttings. These results strongly suggest that endogenous auxin is required for root formation in cuttings of Crassulaceae plants. The differential mode of action of NPA is discussed together with its effect on auxin polar transport

    The effects of N-1-naphthylphthalamic acid (NPA) and 2,3,5-triiodobenzoic acid (TIBA) on the swelling of the stem in intact and decapitated plants of Bryophyllum calycinum in relation to the interaction with auxin, indole-3-acetic acid (IAA), are described. NPA induced conspicuous local internode swelling only in the area of its application in intact plants and in the decapitated internode in the case of simultaneous application of IAA on the top of the internode. By contrast, TIBA applied to an internode of intact plants induced swelling along the entire internode above the treatment area, and similar results were obtained in the decapitated internode when TIBA was applied in the middle of the internode and IAA was applied onto the top of the internode. The differential effect of NPA and TIBA on stem swelling in B. calycinum is discussed in relation to their differential mode of action on auxin transport

    Auxin effectively induces the formation of the secondary abscission zone in Bryophyllum calycinum Salisb. (Crassulaceae)

    We have found that auxin, indole-3-acetic acid (IAA) substantially induces the formation of the secondary abscission zone in stem and petiole explants and in decapitated stem and petiole after excision of blade in intact plants of Bryophyllum calycinum when IAA at a concentration of 0.1% as lanolin paste was applied in the middle of these organs. The secondary abscission zone was formed at a few mm above of the treatment with IAA, and senescence of the part above abscission zone was observed. IAA additionally applied on the top of explants or top of the dacapitated stem or the debladed petiole totally prevented the secondary abscission zone formation and senescence induced by IAA applied in the middle of these organs. Possible mechanisms of the formation of the secondary abscission zone are discussed in terms of the interaction of auxin and ethylene

    Effect of benzyladenine (BA) on auxin-induced stem elongation and thickening in tulip (Tulipa gesneriana L.)

    It is well known that stem elongation in tulip is induced by the auxin produced in the leaves and gynoecium. Excision of the flower bud and all the leaves in an early stage of tulip growth resulted in almost total inhibition of stem growth, but the inhibition was completely recovered by the exogenous application of auxin to the place from which the flower bud had been removed. Hormonal control of stem thickening in tulip is much less known. Additional application of benzyladenine (BA) to the tulip stem by soaking a cotton wick wrapped around all the internodes only slightly inhibited stem growth induced by IAA at a concentration of 0.1 and 2.0%, but substantially stimulated the thickening of all the internodes. The treatment of the tulip stem with benzyladenine enabled direct contact of the cytokinin with the epidermis, which is an important factor in stem elongation. The experiment conducted in field conditions also showed that BA only slightly inhibited the elongation of the fourth internode induced by IAA, but stimulated the thickening of that internode. IAA applied at a concentration of 2.0% stimulated ethylene production to a much higher extent than IAA at a concentration of 0.1%, and BA affected the auxin-induced ethylene production only to a small extent. Metabolic significance of these findings is discussed

    Effect of fluridone on some physiological and qualitative features of ripening tomato fruit

    In tomato fruits, chlorophyll, lycopene and ß-carotene are mostly responsible for the color. During ripening of tomato fruits, the color of the pericarp changes from green to red as chlorophyll is degraded and carotenoids accumulate. These changes are associated with an increase in respiration and ethylene production. Carotenoid biosynthesis pathway in plants can be disturbed by herbicide fluridone (1-methyl-3-phenyl-5-[3-trifluoromethyl(phenyl)]- 4(1H)-pyridinone), which inhibits the activity of phytoene desaturase, an enzyme responsible for conversion of phytoene to phytofluene. Fluridone is also used as an inhibitor of biosynthesis of abscisic acid (ABA) and strigolactones, and it reduces chlorophyll production in plants. In our research we studied the effect of fluridone on some physiological parameters, such as color, firmness, ethylene production, lycopene and chlorophyll content during ripening of the tomato fruit. Tomato plants cv. Altadena (Syngenta) were cultivated in a greenhouse in controlled temperature and both immature and mature fruits were used for the experiments, performed between August and November 2016. Fluridone at concentrations of 0.1% and 1.0% in lanolin paste was applied as a 2-3 mm stripe from the top to the base of tomato fruits, and as a control a stripe of lanolin was applied in the same way on the opposite side of the fruits. Fluridone at a concentration of 1.0% greatly inhibited lycopene accumulation in the pericarp of tomato fruits from the treated side. The measurements of fruit firmness have shown no significant differences between firmness of the part of the tomato fruits treated with fluridone, and the non-treated ones. Tomato fruits treated with fluridone produced amounts of ethylene similar to those found in control tissues on the opposite side of the same fruit. Fluridone delayed chlorophyll degradation in tomato fruits. The metabolic significance of these findings is discussed with the role of carotenogenesis inhibition in tomato fruit ripening

    Hormonal regulation of the growth of leaves and inflorescence stalk in Muscari armeniacum Leichtl.

    It is known that chilling of Muscari bulbs is necessary for the growth of the inflorescence stalk and flowering, but not for the growth of leaves. Gibberellic acid (GA) accelerated stem growth and flowering in chilled Muscari bulbs. In the present experiment it was shown that in unchilled derooted Muscari bulbs the growth of leaves, but not the growth of the inflorescence stalk, was observed when bulbs were stored in water, GA at a concentration of 50 and 100 mg/L, benzyladenine (BA) at a concentration of 25 and 50 mg/L, or a mixture of GA+BA (50+25 mg/L), but abscisic acid (ABA) at a concentration of 10 mg/L greatly inhibited the growth of leaves. In chilled derooted Muscari bulbs the growth of leaves and inflorescence stalk was observed when bulbs were stored in water or GA, but BA and GA+BA treatments totally inhibited the growth of the inflorescence stalk without an effect on the growth of leaves. These results clearly showed that the growth of leaves and inflorescence stalk in Muscari bulbs are controlled by plant growth regulators in different ways. ABA totally inhibited the growth of leaves and inflorescence stalk in chilled derooted Muscari bulbs. It was shown that after the excision of the inflorescence bud in cultivated chilled Muscari bulbs, the inflorescence stalk died, but application of indole-3-acetic acid (IAA) 0.5% in the place of the removed inflorescence bud induced the growth of the inflorescence stalk. IAA applied under the inflorescence bud inhibited the development of flowers (flower-bud blasting) and induced the growth of the inflorescence stalk below the treatment site. These results are discussed with reference to hormonal regulation of stem (stalk) growth in tulip, narcissus, hyacinth, and Hippeastrum

    Effect of Methyl Jasmonate on the Terpene Trilactones, Flavonoids, and Phenolic Acids in Ginkgo biloba L. Leaves: Relevance to Leaf Senescence

    The present study compared the effects of natural senescence and methyl jasmonate (JA-Me) treatment on the levels of terpene trilactones (TTLs; ginkgolides and bilobalide), phenolic acids, and flavonoids in the primary organs of Ginkgo biloba leaves, leaf blades, and petioles. Levels of the major TTLs, ginkgolides B and C, were significantly higher in the leaf blades of naturally senesced yellow leaves harvested on 20 October compared with green leaves harvested on 9 September. In petioles, a similar effect was found, although the levels of these compounds were almost half as high. These facts indicate the importance of the senescence process on TTL accumulation. Some flavonoids and phenolic acids also showed changes in content related to maturation or senescence. Generally, the application of JA-Me slightly but substantially increased the levels of TTLs in leaf blades irrespective of the difference in its application side on the leaves. Of the flavonoids analyzed, levels of quercetin, rutin, quercetin-4-glucoside, apigenin, and luteolin were dependent on the JA-Me application site, whereas levels of (+) catechin and (−) epicatechin were not. Application of JA-Me increased ferulic acid and p-coumaric acid esters in the petiole but decreased the levels of these compounds in the leaf blade. The content of p-coumaric acid glycosides and caffeic acid esters was only slightly modified by JA-Me. In general, JA-Me application affected leaf senescence by modifying the accumulation of ginkogolides, flavonoids, and phenolic acids. These effects were also found to be different in leaf blades and petioles. Based on JA-Me- and aging-related metabolic changes in endogenous levels of the secondary metabolites in G. biloba leaves, we discussed the results of study in the context of basic research and possible practical application

    Changes in Polar Metabolites Content during Natural and Methyl-Jasmonate-Promoted Senescence of <i>Ginkgo biloba</i> Leaves

    The present study clarified changes in the contents of polar metabolites (amino acids, organic acids, saccharides, cyclitols, and phosphoric acid) in leaf senescence in Ginkgo biloba with or without the application of methyl jasmonate (JA-Me) in comparison with those in naturally senescent leaf blades and petioles. The contents of most amino acids and citric and malic acids were significantly higher in abaxially, and that of myo-inositol was lower in abaxially JA-Me-treated leaves than in adaxially JA-Me-treated and naturally senescent leaves. The levels of succinic and fumaric acids in leaves treated adaxially substantially high, but not in naturally senescent leaves. In contrast, sucrose, glucose, and fructose contents were much lower in leaf blades and petioles treated abaxially with JA-Me than those treated adaxially. The levels of these saccharides were also lower compared with those in naturally senescent leaves. Shikimic acid and quinic acid were present at high levels in leaf blades and petioles of G. biloba. In leaves naturally senescent, their levels were higher compared to green leaves. The shikimic acid content was also higher in the organs of naturally yellow leaves than in those treated with JA-Me. These results strongly suggest that JA-Me applied abaxially significantly enhanced processes of primary metabolism during senescence of G. biloba compared with those applied adaxially.
