Obesity-associated variants within FTO form long-range functional connections with IRX3
PMCID: PMC4113484.-- et al.Genome-wide association studies (GWAS) have reproducibly associated variants within introns of FTO with increased risk for obesity and type 2 diabetes (T2D). Although the molecular mechanisms linking these noncoding variants with obesity are not immediately obvious, subsequent studies in mice demonstrated that FTO expression levels influence body mass and composition phenotypes. However, no direct connection between the obesity-associated variants and FTO expression or function has been made. Here we show that the obesity-associated noncoding sequences within FTO are functionally connected, at megabase distances, with the homeobox gene IRX3. The obesity-associated FTO region directly interacts with the promoters of IRX3 as well as FTO in the human, mouse and zebrafish genomes. Furthermore, long-range enhancers within this region recapitulate aspects of IRX3 expression, suggesting that the obesity-associated interval belongs to the regulatory landscape of IRX3. Consistent with this, obesity-associated single nucleotide polymorphisms are associated with expression of IRX3, but not FTO, in human brains. A direct link between IRX3 expression and regulation of body mass and composition is demonstrated by a reduction in body weight of 25 to 30% in Irx3-deficient mice, primarily through the loss of fat mass and increase in basal metabolic rate with browning of white adipose tissue. Finally, hypothalamic expression of a dominant-negative form of Irx3 reproduces the metabolic phenotypes of Irx3-deficient mice. movilidad IP, balanceo de carga y alta disponibilidad por medio de la herramienta de virtualización de código abierto VNUML. Para mostrar estos conceptos hemos añadido diversas utilidades al sistema de ficheros utilizado en VNUML. Gracias a estas herramientas, los alumnos han podido mejorar

A framework for supporting network continuity in vehicular IPv6 communications
The appearance of recent standards about cooperative ITS architectures towards a reference communication stack has been an inflection point in the research about vehicular networks. The ISO Communication Access for Land Mobiles (CALM) and the ETSI European ITS communication architecture have paved the way towards real and interoperable vehicular cooperative systems. Within these convergent proposals, IPv6 communications are recognized as a key component to enable traffic efficiency and infotainment applications. The proper operation of these applications and the achievement of value-added ITS services require an uninterrupted network connectivity. This paper addresses this problem by proposing a novel communication stack to support the provision of continuous and secure IPv6 vehicular communications. The solution follows the ISO/ETSI guidelines for the development of cooperative ITS systems and is based on standardized technologies such as Network Mobility (NEMO) protocol to provide an integral management of IPv6mobility. The solution integrates IEEE 802.21 media independent handover services for optimizing the handover process. Ultrasonography detection of pregnancy in outdoor iberian sows
In a flock of pure Iberian sows, with outdoor breeding and total freedom, ultrasonography detection of pregnancy length is studied (11.1±0.6 seconds), according to pregnancy or not pregnancy (9.7 vs 20.4 seconds). Also, possible influence of cycle number is analysed. By other side permanence of ear tags after a year is valued (99.5%). Results show that indoor and intensive production is not justified in order to attain more reproductive control.En un rebaño de cerdas Ibéricas puras, criadas en régimen de total libertad (sin empleo de jaulas), se estudia el tiempo empleado en el diagnóstico de gestación con ecógrafo (11,1±0,6 segundos por cerda), se analiza la duración del diagnóstico dependiendo de si hay preñez o no (9,7 vs 20,4 segundos) y la posible influencia del número de ciclo. Por otra parte se valora la permanencia de los crotales de identificación individual tras un año (99,5%). Los resultados indican que la intensificación de la producción en esta raza no está justificada por la consecución de un mayor control reproductivo
Bisphenol A Induces Accelerated Cell Aging in Murine Endothelium.
Bisphenol A (BPA) is a widespread endocrine disruptor affecting many organs and systems. Previous work in our laboratory demonstrated that BPA could induce death due to necroptosis in murine aortic endothelial cells (MAECs). This work aims to evaluate the possible involvement of BPA-induced senescence mechanisms in endothelial cells. The β-Gal assays showed interesting differences in cell senescence at relatively low doses (100 nM and 5 µM). Western blots confirmed that proteins involved in senescence mechanisms, p16 and p21, were overexpressed in the presence of BPA. In addition, the UPR (unfolding protein response) system, which is part of the senescent phenotype, was also explored by Western blot and qPCR, confirming the involvement of the PERK-ATF4-CHOP pathway (related to pathological processes). The endothelium of mice treated with BPA showed an evident increase in the expression of the proteins p16, p21, and CHOP, confirming the results observed in cells. Our results demonstrate that oxidative stress induced by BPA leads to UPR activation and senescence since pretreatment with N-acetylcysteine (NAC) in BPA-treated cells reduced the percentage of senescent cells prevented the overexpression of proteins related to BPA-induced senescence and reduced the activation of the UPR system. The results suggest that BPA participates actively in accelerated cell aging mechanisms, affecting the vascular endothelium and promoting cardiovascular diseases.post-print3206 K
Environmental concern as a moderator of information processing: A fMRI study
The psychological processes that predict behaviour can be influenced by the approaches taken in environmental awareness messages. The persuasiveness generated by different approaches depends on the environmental concern of viewers. The study identifies brain regions active while processing positive advertisements. It also studies brain activity in subjects with high (vs low) environmental concern while processing positive ads. In addition, it relates the brain activity evoked in response to positive ads which predicts more significant attitudes toward the ads, in subjects with great environmental concern. The results indicate that positive messages activate regions linked to self-value and societal benefit in those subjects who are more concerned. We found a stronger effect in regions linked to an emotional response in viewers with greater environmental concern. The identified emotional response may precede higher attitudinal ratings
A monounsaturated fatty acid rich diet reduces macrophage uptake of plasma oxidized LDL in healthy young men
During atherogenesis, a pathological accumulation of lipids occurs within aortic intimal macrophages through uptake of plasma oxidised LDL (oxLDL). The aim of the present study was to determine whether macrophage uptake of plasma oxLDL and LDL susceptibility to oxidation may be determined by quantity and quality of dietary fat. Twenty healthy young men were subjected to three dietary periods, each lasting 4 weeks. The first was an SFA-enriched diet (38 % fat, 20 % SFA), which was followed by a carbohydrate (CHO)-rich diet (30 % fat, < 10 % SFA, 55 % CHO) or a MUFA olive oil-rich diet (38 % fat, 22 % MUFA) following a randomised cross-over design. After each diet period, LDL particles were oxidised with Cu ions to determine LDL susceptibility to oxidation and subsequently incubated with the U937-macrophage cell line to determine the percentage of uptake of plasma oxLDL. The shift from the MUFA diet to the SFA- or CHO-rich diets reduced the resistance of LDL particles to oxidation, decreasing lag time (P = 0·038) and increasing the propagation rate (P = 0·001). Furthermore, the MUFA-rich diet demonstrated reduced macrophage uptake of plasma oxLDL (P = 0·031) as compared with the SFA-rich diet. Finally, macrophage uptake of plasma oxLDL was correlated (r 0·45; P = 0·040) with total amount of conjugated dienes after LDL oxidation. Our data suggest that a MUFA-rich diet may have favourable effects on cardiovascular risk since it prevents the oxidative modifications of LDL and reduces macrophage uptake of plasma oxLDL
