    Cancer incidence and nuclear facilities in Ukraine: a community-based stydy

    The study goal was to investigate malignant tumors incidence in 5 Ukrainian cities with nuclear hazardous enterprises: extractive, processing enterprises of uranium ore (Zhovti Wody and Dniprodzerzhynsk of Dnipropetrovsk region) and nuclear power stations (Energodar of Zaporizhska region, Pivdennoukrainsk of Mykolayivska region, Netishyn of Khmelnytska region). Materials and methods: average annual population of the cities under study in 2003–2008 was 439 600 persons. Total and specific cancer incidence was investigated. Site specific incidence was analyzed for malignancies proved to be radiosensitive in previous studies: trachea, bronchus and lung, breast, kidney, thyroid cancer and leukemia. Data on cancer cases were received in National Cancer Registry of Ukraine (National Cancer Institute). There was used the data of the State Statistics Committee of Ukraine on the size of the studied population by gender — age groups. Standardized incidence ratio of cancer at a whole and for each of five specific forms of malignancies were calculated for the population of each city and group of cities depending on the nature of industrial activity. Results: During the observed period there were registered 9 381 cancer cases in inhabitants of Ukrainian cities with radiation hazardous facilities. There was stated that cancer incidence rate in population of 5 cities significantly exceeded national and regional levels. Among specific forms of malignancy there were observed excess of lung, trachea, bronchus, breast, kidney cancer and leukemia in population of extractive, processing uranium ore cities. No excess of thyroid cancer was identified. In cities with nuclear power station there were registered excess of kidney cancer. Conclusion: Results of the study suggest the necessity to explore the role of various factors in forming the identified cancer incidence features in the Ukrainian population living near the nuclear power facilities

    Virion Structure and In Vitro Genome Release Mechanism of Dicistrovirus Kashmir Bee Virus

    Infections with Kashmir bee virus (KBV) are lethal for honeybees and have been associated with colony collapse disorder. KBV and closely related viruses contribute to the ongoing decline in the number of honeybee colonies in North America, Europe, Australia, and other parts of the world. Despite the economic and ecological impact of KBV, its structure and infection process remain unknown. Here, we present the structure of the virion of KBV determined to a resolution of 2.8 angstrom. We show that the exposure of KBV to acidic pH induces a reduction in interpentamer contacts within capsids and the reorganization of its RNA genome from a uniform distribution to regions of high and low density. Capsids of KBV crack into pieces at acidic pH, resulting in the formation of open particles lacking pentamers of capsid proteins. The large openings of capsids enable the rapid release of genomes and thus limit the probability of their degradation by RNases. The opening of capsids may be a shared mechanism for the genome release of viruses from the family Dicistroviridae. IMPORTANCE The western honeybee (Apis mellifera) is indispensable for maintaining agricultural productivity as well as the abundance and diversity of wild flowering plants. However, bees suffer from environmental pollution, parasites, and pathogens, including viruses. Outbreaks of virus infections cause the deaths of individual honeybees as well as collapses of whole colonies. Kashmir bee virus has been associated with colony collapse disorder in the United States, and no cure for the disease is currently available. Here, we report the structure of an infectious particle of Kashmir bee virus and show how its protein capsid opens to release the genome. Our structural characterization of the infection process determined that therapeutic compounds stabilizing contacts between pentamers of capsid proteins could prevent the genome release of the virus.We gratefully acknowledge the Cryoelectron Microscopy and Tomography core facility of CEITEC supported by MEYS CR (LM2018127) .     Adult trkB signaling in parvalbumin interneurons is essential to prefrontal network dynamics

    Inhibitory interneurons expressing parvalbumin (PV) are central to cortical network dynamics, generation of c oscillations, and cognition. Dysfunction of PV interneurons disrupts cortical information processing and cognitive behavior. Brain-derived neurotrophic factor (BDNF)/tyrosine receptor kinase B (trkB) signaling regulates the maturation of cortical PV interneurons but is also implicated in their adult multidimensional functions. Using a novel viral strategy for cell-type-specific and spatially restricted expression of a dominant-negative trkB (trkB.DN), we show that BDNF/trkB signaling is essential to the integrity and maintenance of prefrontal PV interneurons in adult male and female mice. Reduced BDNF/trkB signaling in PV interneurons in the medial prefrontal cortex (mPFC) resulted in deficient PV inhibition and increased baseline local field potential (LFP) activity in a broad frequency band. The altered network activity was particularly pronounced during increased activation of the prefrontal network and was associated with changed dynamics of local excitatory neurons, as well as decreased modulation of the LFP, abnormalities that appeared to generalize across stimuli and brain states. In addition, our findings link reduced BDNF/trkB signaling in prefrontal PV interneurons to increased aggression. Together our investigations demonstrate that BDNF/trkB signaling in PV interneurons in the adult mPFC is essential to local network dynamics and cognitive behavior.     Capturing the state-of-knowledge in EURAD knowledge management

    Knowledge about a wide variety of aspects is fundamental for the safe management and disposal of radioactive waste. This importance of Knowledge Management (KM) is also recognised by EURAD, the European Joint Programme on Radioactive Waste Management (RWM), which brings together over 100 organisations from different countries and backgrounds (Waste Management Organisations, Technical Support Organisations, and Research Entities). This vast resource of expertise and experience feeds into several dedicated EURAD KM programme activities. One of these activities, led by Work Package 11 State-of-Knowledge (WP11 SoK), is capturing experts’ views on the current State-of-Knowledge on topics relevant to RWM and making this knowledge accessible through dedicated documents. For this, EURAD has developed the “Goals Breakdown Structure” (GBS), which provides a framework in which topics are structured thematically, as well as a hierarchy of documents that allows accessing knowledge on different levels of detail (see EURAD Roadmap). To make this knowledge available, EURAD is currently developing a Wiki (i.e., a web-based resource that allows access to knowledge and collaborative interactions) and is drafting a sustainable Knowledge Management System and other supportive KM-IT tools while already feeding the tools with content. This article gives an insight into the general EURAD KM concept, the approaches used, and the results obtained until EURAD’s mid-term, after 2.5 years

    Development of guidance documents in the EURAD and PREDIS projects

    Particular emphasis is dedicated to Knowledge Management activities within the EURAD (European Joint Programme on Radioactive Waste Management) and PREDIS (Pre-disposal management of radioactive waste) projects to ensure the capture of existing knowledge, transfer of knowledge between Members States and management of the knowledge for future generations. The EURAD project has three work packages dedicated to knowledge management. One of them, the EURAD Guidance work package (WP12) is developing a comprehensive suite of specific guidance documents that can be used by Members States with radioactive waste management (RWM) programmes that are at an early stage of development but can be beneficial also to more advanced programmes. The PREDIS project does not have a specifically allocated work package for guidance development. Rather, such activities are integrated within deliverables produced as part of the Strategic Implementation and State of Knowledge actions of the Roadmap contributions on predisposal waste management. The EURAD guidance work is based on the existing PLANDIS guide on RD&D planning, developed by the Implementing Geological Disposal of Radioactive Waste Technology Platform (IGD-TP). The guidance documents complement other documents prepared in parallel knowledge management activities inside EURAD project: the State of Knowledge documents. The differentiation is that guidance documents explain in more detail how the process can be established and performed, including illustrative examples. The guides are self-standing documents and integrated with the EURAD Roadmap. The target end users of the guidance are primarily programme owners and managers (i.e., governments/administrations, Waste Management Organisations, Research Entities and Technical Support Organisations) responsible for planning and implementing the RWM programme and the supporting RD&D activities at a national level, even though they might also be of use and interest to other interested stakeholders, such as representatives of civil society. To produce a first list of prioritised topics for guidance documents with the aim to select a topic for a pilot guide, the Guidance WP has developed a screening process that includes review by experts and end users. Based on the priority list, the first pilot guide was developed with the title “Cost Assessment and Financing Schemes of Radioactive Waste Management Programmes”. Experience gained during the selection of topics for the pilot guide and during its production are being incorporated into the procedure for identification of new topics for which guides will be developed. First, the degree of coverage of the EURAD Roadmap themes by suitable guide documents will be analysed by the WP 12 team. The analysis will be combined with feedback from experts verifying the needs for missing guides. Finally, the potential end user community representatives will be given the opportunity to comment on the prioritisation of selected guidance documents and make additional suggestions. The potential end users stay involved also during the production of the guides. This procedure aims to optimise the scarce expert resources in relation to the identified needs of guidance documents.     Chernobyl fallout and cancer incidence in Finland.

    Twenty-five years have passed since the Chernobyl accident, but its health consequences remain to be well established. Finland was one of the most heavily affected countries by the radioactive fallout outside the former Soviet Union. We analyzed the relation of the estimated external radiation exposure from the fallout to cancer incidence in Finland in 1988–2007. The study cohort comprised all ∼3.8 million Finns who had lived in the same dwelling for 12 months following the accident (May 1986–April 1987). Radiation exposure was estimated using data from an extensive mobile dose rate survey. Cancer incidence data were obtained for the cohort divided into four exposure categories (the lowest with the first-year committed dose <0.1 mSv and the highest ≥0.5 mSv) allowing for a latency of 5 years for leukemia and thyroid cancer, and 10 years for other cancers. Of the eight predefined cancer sites regarded as radiation-related from earlier studies, only colon cancer among women showed an association with exposure from fallout [excess rate ratio per increment in exposure category 0.06, 95% confidence interval (CI) 0.02–0.11]. No such effect was observed for men, or other cancer sites. Our analysis of a large cohort over two decades did not reveal an increase in cancer incidence following the Chernobyl accident, with the possible exception of colon cancer among women. The largely null findings are consistent with extrapolation from previous studies suggesting that the effect is likely to remain too small to be empirically detectable and of little public health impact
