Quantitative regulation of FLC via coordinated transcriptional initiation and elongation
The basis of quantitative regulation of gene expression is still poorly understood. In Arabidopsis thaliana, quantitative variation in expression of FLOWERING LOCUS C (FLC) influences the timing of flowering. In ambient temperatures, FLC expression is quantitatively modulated by a chromatin silencing mechanism involving alternative polyadenylation of antisense transcripts. Investigation of this mechanism unexpectedly showed that RNA polymerase II (Pol II) occupancy changes at FLC did not reflect RNA fold changes. Mathematical modeling of these transcriptional dynamics predicted a tight coordination of transcriptional initiation and elongation. This prediction was validated by detailed measurements of total and chromatin-bound FLC intronic RNA, a methodology appropriate for analyzing elongation rate changes in a range of organisms. Transcription initiation was found to vary ∼25-fold with elongation rate varying ∼8- to 12-fold. Premature sense transcript termination contributed very little to expression differences. This quantitative variation in transcription was coincident with variation in H3K36me3 and H3K4me2 over the FLC gene body. We propose different chromatin states coordinately influence transcriptional initiation and elongation rates and that this coordination is likely to be a general feature of quantitative gene regulation in a chromatin context
Quantitative Proteomics Using Isobaric Labeling:A Practical Guide
In the past decade, relative proteomic quantification using isobaric labeling technology has developed into a key tool for comparing the expression of proteins in biological samples. Although its multiplexing capacity and flexibility make this a valuable technology for addressing various biological questions, its quantitative accuracy and precision still pose significant challenges to the reliability of its quantification results. Here, we give a detailed overview of the different kinds of isobaric mass tags and the advantages and disadvantages of the isobaric labeling method. We also discuss which precautions should be taken at each step of the isobaric labeling workflow, to obtain reliable quantification results in large-scale quantitative proteomics experiments. In the last section, we discuss the broad applications of the isobaric labeling technology in biological and clinical studies, with an emphasis on thermal proteome profiling and proteogenomics.</p
Suppression of FM-to-AM conversion in third-harmonic generation at the retracing point of a crystal
FM-to-AM conversion can cause many negative effects (e.g., reducing of margin
against damage to the optics, and etc.) on performances of third-harmonic
conversion system. In this letter, the FM-to-AM conversion effect in
third-harmonic generation is investigated both at and away from the retracing
point of type-II KDP crystal. Obtained results indicate that the FM-to-AM
conversion can be suppressed effectively when the crystal works at the

Accurate Guidance Method and App Development for Assigning Parking Spaces Based on Indoor Wi-Fi
Existing parking guidance systems only provide road guidance outside the parking lot but do not provide accurate guidance to specific parking spaces inside the parking lot. By using a Kalman filter, the Grubbs test, and a neural network algorithm to improve the RSSI-based location fingerprint identification technology, an accurate location method based on indoor Wi-Fi is obtained, which implements precise route guidance and a reverse search function for parking spaces. We utilize Beidou positioning to develop a Gaode map for outdoor navigation and use an integrated system of ultrasonic detector/indicators and ground locks to manage parking spaces. Through the secondary development of an Android system and the application of a MySql database, an app for precise parking guidance was developed. The system makes full use of the Internet and parking information, eliminates information asymmetry, improves the utilization ratio of the urban static traffic resources, allocates parking spaces in real-time, breaks information islands, provides parking search and recommendation functions for users, achieves parking information-sharing, and effectively improves parking efficiency and the parking utilization ratio
Passenger Satisfaction Analysis of an Urban Subway Based on Travel Behaviour Characteristics
The study explores the relationship between subway passenger satisfaction and passenger travel behaviour characteristics from the perspective of subway passengers. This study takes Qingdao as an example and designs a questionnaire that includes the basic personal information and travel behaviour characteristics of passengers and evaluates their subway satisfaction. A total of 6340 valid questionnaires were obtained through the combination of online surveys and on-site random surveys. By using the fuzzy synthetic evaluation, the overall score of passenger satisfaction with the Qingdao subway is determined. According to the relationship between passenger satisfaction and travel behaviour characteristics, the chi-square test is used to select the correlation variables group. The results show that there is no significant correlation among the satisfaction of subway passengers, the main means of transportation and the availability of private cars; the frequency of taking the subway is related to the satisfaction of subway passengers; and the purpose of travelling and the reasons for choosing the subway are significantly related to the satisfaction of subway passengers. Finally, based on the analysis of the differences in satisfaction under different conditions, some suggestions were proposed to improve the satisfaction of subway passengers
Discovery and identification of potential biomarkers of papillary thyroid carcinoma
<p>Abstract</p> <p>Background</p> <p>Thyroid carcinoma is the most common endocrine malignancy and a common cancer among the malignancies of head and neck. Noninvasive and convenient biomarkers for diagnosis of papillary thyroid carcinoma (PTC) as early as possible remain an urgent need. The aim of this study was to discover and identify potential protein biomarkers for PTC specifically.</p> <p>Methods</p> <p>Two hundred and twenty four (224) serum samples with 108 PTC and 116 controls were randomly divided into a training set and a blind testing set. Serum proteomic profiles were analyzed using SELDI-TOF-MS. Candidate biomarkers were purified by HPLC, identified by LC-MS/MS and validated using ProteinChip immunoassays.</p> <p>Results</p> <p>A total of 3 peaks (<it>m/z </it>with 9190, 6631 and 8697 Da) were screened out by support vector machine (SVM) to construct the classification model with high discriminatory power in the training set. The sensitivity and specificity of the model were 95.15% and 93.97% respectively in the blind testing set. The candidate biomarker with <it>m/z </it>of 9190 Da was found to be up-regulated in PTC patients, and was identified as haptoglobin alpha-1 chain. Another two candidate biomarkers (6631, 8697 Da) were found down-regulated in PTC and identified as apolipoprotein C-I and apolipoprotein C-III, respectively. In addition, the level of haptoglobin alpha-1 chain (9190 Da) progressively increased with the clinical stage I, II, III and IV, and the expression of apolipoprotein C-I and apolipoprotein C-III (6631, 8697 Da) gradually decreased in higher stages.</p> <p>Conclusion</p> <p>We have identified a set of biomarkers that could discriminate PTC from non-cancer controls. An efficient strategy, including SELDI-TOF-MS analysis, HPLC purification, MALDI-TOF-MS trace and LC-MS/MS identification, has been proved successful.</p
Impact of countdown signals on traffic safety and efficiency: a review and proposal
Countdown signals for motorized vehicles, which are intended to ensure safety on the road and regulate motor vehicle speed limits at road intersections, are still considered a relatively novel concept. These signals have been adopted by only a few countries, and the number of cities that use them is limited. This review aims to summarize the effects of countdown signals on traffic safety and efficiency and to determine the consistency and differences of existing research propositions on the matter. Based on the review, considerable research presents evidently different conclusions in the areas of driver red-light running and traffic safety. Particularly, some studies propose that countdown signals reinforce traffic safety, whereas others consider that such signals adversely affect traffic safety. Meanwhile, related literature provides varying conclusions on the aspect of traffic efficiency for vehicle headway. At present, the number of studies conducted regarding the driving behaviors of motorists toward countdown-signalized intersections is insufficient. Accordingly, such inadequate diversity in research causes difficulty in completely assessing the benefits and disadvantages of countdown signals.
