To achieve a sustainable blue future, progress assessments must include interdependencies between the sustainable development goals
The Sustainable Development Goals (SDGs) focus on providing society with a sustainable future. Progress toward the goals is being tracked by a series of indicators. These indicators show progress toward individual goals and targets but do not show how success or failure in relation to one goal might affect success or failure in another area. We show how interactions between the oceans and human poverty, hunger, and gender equity are hidden by indicator assessments and how this undermines the capacity of governments and organizations to maximize long-term moves toward sustainability. These findings are important for decision makers who work in the public and private sectors and wish to avoid unforeseen outcomes when implementing sustainability initiatives. Here, we suggest extensions to the current assessment framework to help counteract the identified issues, providing a research agenda for scientists working in all fields of sustainability science
From counting to construction of BPS states in N=4 SYM
We describe a universal element in the group algebra of symmetric groups,
whose characters provides the counting of quarter and eighth BPS states at weak
coupling in N=4 SYM, refined according to representations of the global
symmetry group. A related projector acting on the Hilbert space of the free
theory is used to construct the matrix of two-point functions of the states
annihilated by the one-loop dilatation operator, at finite N or in the large N
limit. The matrix is given simply in terms of Clebsch-Gordan coefficients of
symmetric groups and dimensions of U(N) representations. It is expected, by
non-renormalization theorems, to contain observables at strong coupling. Using
the stringy exclusion principle, we interpret a class of its eigenvalues and
eigenvectors in terms of giant gravitons. We also give a formula for the action
of the one-loop dilatation operator on the orthogonal basis of the free theory,
which is manifestly covariant under the global symmetry.Comment: 41 pages + Appendices, 4 figures; v2 - refs and acknowledgments adde
Single ventricle with persistent truncus arteriosus as two rare entities in an adult patient: a case report
Abstract Introduction Single ventricle and truncus arteriosus are both rare congenital cardiac syndromes with limited survival. Their occurrence together is extremely uncommon and prolonged survival is exceptionally rare. We present the case of a patient who had both of these defects with survival to age 45. Case presentation We describe the vase of a 45-year-old man with the unusual occurrence of two very rare congenital cardiac defects. He was found to have both truncus arteriosus and single ventricle with long survival. His history, clinical course, and anatomic findings are discussed along with the factors which may have contributed to his longevity, which is unique in the medical literature. His management reflected the state of medical knowledge at the time when he presented, and although alternate approaches may have been utilized if the patient presented today, this case does indicate the efficacy of the management options available at the time and place of the patient's contacts with the medical care system in Belarus. We discuss the findings, frequency, classification, and management of both of these congenital defects. Conclusion This case demonstrates that patients with very complex congenital cardiac disease may survive to adulthood, presenting challenges in both medical and surgical treatment. As the management of these patients is constantly evolving, and interventional techniques are improving, patients such as this with prolonged survival will be more common, with each case providing insights to future treatment. Challenges in management may include prior care provided in health care systems with limited resources.</p

Rapamycin-loaded nanoparticles for inhibition of neointimal hyperplasia in experimental vein grafts

Abstract

Background

Nanoparticles possess several advantages as a carrier system for intracellular delivery of therapeutic agents. Rapamycin is an immunosuppressive agent which also exhibits marked antiproliferative properties. We investigated whether rapamycin-loaded nanoparticles(NPs) can reduce neointima formation in a rat model of vein graft disease.

Methods

Poly(lactic-co-glycolic acid) (PLGA) NPs containing rapamycin was prepared using an oil/water solvent evaporation technique. Nanoparticle size and morphology were determined by dynamic light scattering methodology and electron microscopy. In vitro cytotoxicity of blank, rapamycin-loaded PLGA (RPLGA) NPs was studied using MTT Assay. Excised rat jugular vein was treated ex vivo with blank-NPs, or rapamycin-loaded NPs, then interposed back into the carotid artery position using a cuff technique. Grafts were harvested at 21 days and underwent morphometric analysis as well as immunohistochemical analysis.

Results

Rapamycin was efficiently loaded in PLGA nanoparticles with an encapsulation efficiency was 87.6%. The average diameter of NPs was 180.3 nm. The NPs-containing rapamycin at 1 ng/ml significantly inhibited vascular smooth muscular cells proliferation. Measurement of rapamycin levels in vein grafts shown that the concentration of rapamycin in vein grafts at 3 weeks after grafting were 0.9 ± 0.1 μg/g. In grafted veins without treatment intima-media thickness was 300.4 ±181.5 μm after grafting 21 days. Whereas, Veins treated with rapamycin-loaded NPs showed a reduction of intimal-media thickness of 150.2 ± 62.5 μm (p = 0.001). CD-31 staining was used to measure luminal endothelial coverage in grafts and indicated a high level of endothelialization in 21 days vein grafts with no significant effect of blank or rapamycin-loaded NPs group.

Conclusions

We conclude that sustained-release rapamycin from rapymycin loaded NPs inhibits vein graft thickening without affecting the reendothelialization in rat carotid vein-to-artery interposition grafts and this may be a promising therapy for the treatment of vein graft disease.</p
