    Identification of potential target genes for the tomato fruit-ripening regulator RIN by chromatin immunoprecipitation

    <p>Abstract</p> <p>Background</p> <p>During ripening, climacteric fruits increase their ethylene level and subsequently undergo various physiological changes, such as softening, pigmentation and development of aroma and flavor. These changes occur simultaneously and are caused by the highly synchronized expression of numerous genes at the onset of ripening. In tomatoes, the MADS-box transcription factor RIN has been regarded as a key regulator responsible for the onset of ripening by acting upstream of both ethylene- and non-ethylene-mediated controls. However, except for <it>LeACS2</it>, direct targets of RIN have not been clarified, and little is known about the transcriptional cascade for ripening.</p> <p>Results</p> <p>Using immunoprecipitated (IPed) DNA fragments recovered by chromatin immunoprecipitation (ChIP) with anti-RIN antibody from ripening tomato fruit, we analyzed potential binding sites for RIN (CArG-box sites) in the promoters of representative ripening-induced genes by quantitative PCR. Results revealed nearly a 5- to 20-fold enrichment of CArG boxes in the promoters of <it>LeACS2</it>, <it>LeACS4</it>, <it>PG</it>, <it>TBG4</it>, <it>LeEXP1</it>, and <it>LeMAN4 </it>and of <it>RIN </it>itself, indicating direct interaction of RIN with their promoters <it>in vivo</it>. Moreover, sequence analysis and genome mapping of 51 cloned IPed DNAs revealed potential RIN binding sites. Quantitative PCR revealed that four of the potential binding sites were enriched 4- to 17-fold in the IPed DNA pools compared with the controls, indicating direct interaction of RIN with these sites <it>in vivo</it>. Near one of the four CArG boxes we found a gene encoding a protein similar to thioredoxin y1. An increase in the transcript level of this gene was observed with ripening in normal fruit but not in the <it>rin </it>mutant, suggesting that RIN possibly induces its expression.</p> <p>Conclusions</p> <p>The presented results suggest that RIN controls fruit softening and ethylene production by the direct transcriptional regulation of cell-wall-modifying genes and ethylene biosynthesis genes during ripening. Moreover, the binding of RIN to its own promoter suggests the presence of autoregulation for <it>RIN </it>expression. ChIP-based analyses identified a novel RIN-binding CArG-box site that harbors a gene associated with <it>RIN </it>expression in its flanking region. These findings clarify the crucial role of RIN in the transcriptional regulation of ripening initiation and progression.</p

    Frequent Transmission of Streptococcus pneumoniae Serotype 35B and 35D, Clonal Complex 558 Lineage, across Continents and the Formation of Multiple Clades in Japan

    Streptococcus pneumoniae is a common bacterial pathogen that causes infections in children worldwide, even after administration of the pneumococcal conjugate vaccine. S. pneumoniae serotype 35B, especially the clonal complex 558 (CC558) lineage, has emerged globally following implementation of the 13-valent pneumococcal conjugate vaccine. Serotype 35B strains are also associated with multidrug resistance to both β-lactams and non-β-lactam drugs. In addition, a novel serotype, 35D, which is closely related to 35B and differs in polysaccharide structure, was recently reported. However, the genetic relationship among globally disseminating serotype 35B and D (35B/D) strains remains unknown. To investigate the molecular epidemiology of global serotype 35B/D strains, we conducted a genomic analysis of serotype 35B/D strains from various continents, including those from the Japanese national surveillance collection. A total of 87 isolates were identified as serotype 35B/D in the Japanese surveillance collection (n = 1, 358). All the isolates were assigned to either CC558 or CC2755. Serotype 35D isolates were interspersed with serotype 35B isolates. Phylogenetic analysis revealed the formation of multiple clusters by the Japanese serotype 35B/D-CC558 isolates among the foreign isolates, which suggested multiple events of introduction of the clone into Japan. The global 35B/D-CC558 strains were found to share specific penicillin-binding protein profiles, pbp1a-4, pbp2b-7, and pbp2x-7, associated with penicillin, cephalosporin, and carbapenem nonsusceptibility. Moreover, 88.5% of the Japanese 35B/D-CC558 and 35B/D-CC2755 isolates were found to harbor the Tn916-like integrative and conjugative elements Tn2009, Tn2010, and Tn6002, associated with multidrug resistance to macrolides and tetracyclines.     Spred2-deficiency enhances the proliferation of lung epithelial cells and alleviates pulmonary fibrosis induced by bleomycin

    The mitogen-activated protein kinase (MAPK) pathways are involved in many cellular processes, including the development of fibrosis. Here, we examined the role of Sprouty-related EVH-1-domain-containing protein (Spred) 2, a negative regulator of the MAPK-ERK pathway, in the development of bleomycin (BLM)-induced pulmonary fibrosis (PF). Compared to WT mice, Spred2−/− mice developed milder PF with increased proliferation of bronchial epithelial cells. Spred2−/− lung epithelial cells or MLE-12 cells treated with spred2 siRNA proliferated faster than control cells in vitro. Spred2−/− and WT macrophages produced similar levels of TNFα and MCP-1 in response to BLM or lipopolysaccharide and myeloid cell-specific deletion of Spred2 in mice had no effect. Spred2−/− fibroblasts proliferated faster and produced similar levels of MCP-1 compared to WT fibroblasts. Spred2 mRNA was almost exclusively detected in bronchial epithelial cells of naïve WT mice and it accumulated in approximately 50% of cells with a characteristic of Clara cells, 14 days after BLM treatment. These results suggest that Spred2 is involved in the regulation of tissue repair after BLM-induced lung injury and increased proliferation of lung bronchial cells in Spred2−/− mice may contribute to faster tissue repair.     Evolution of the Fe-3dd impurity band state as the origin of high Curie temperature in p-type ferromagnetic semiconductor (Ga,Fe)Sb

    (Ga1x_{1-x},Fex_x)Sb is one of the promising ferromagnetic semiconductors for spintronic device applications because its Curie temperature (TCT_{\rm C}) is above 300 K when the Fe concentration xx is equal to or higher than ~0.20. However, the origin of the high TCT_{\rm C} in (Ga,Fe)Sb remains to be elucidated. To address this issue, we use resonant photoemission spectroscopy (RPES) and first-principles calculations to investigate the xx dependence of the Fe 3dd states in (Ga1x_{1-x},Fex_x)Sb (xx = 0.05, 0.15, and 0.25) thin films. The observed Fe 2pp-3dd RPES spectra reveal that the Fe-3dd impurity band (IB) crossing the Fermi level becomes broader with increasing xx, which is qualitatively consistent with the picture of double-exchange interaction. Comparison between the obtained Fe-3dd partial density of states and the first-principles calculations suggests that the Fe-3dd IB originates from the minority-spin (\downarrow) ee states. The results indicate that enhancement of the interaction between ee_\downarrow electrons with increasing xx is the origin of the high TCT_{\rm C} in (Ga,Fe)Sb

    The internal behavior of fast ions interacting with magnetohydrodynamic bursts excited by energetic ions has been experimentally investigated in the compact helical system. The resonant convective oscillation of fast ions was identified inside the last closed-flux surface during an energetic-particle mode (EPM) burst.     Subaru Hyper Suprime-Cam Survey for An Optical Counterpart of GW170817

    We perform a zz-band survey for an optical counterpart of a binary neutron star coalescence GW170817 with Subaru/Hyper Suprime-Cam. Our untargeted transient search covers 23.623.6 deg2^2 corresponding to the 56.6%56.6\% credible region of GW170817 and reaches the 50%50\% completeness magnitude of 20.620.6 mag on average. As a result, we find 60 candidates of extragalactic transients, including J-GEM17btc (a.k.a. SSS17a/DLT17ck). While J-GEM17btc is associated with NGC 4993 that is firmly located inside the 3D skymap of GW170817, the other 59 candidates do not have distance information in the GLADE v2 catalog or NASA/IPAC Extragalactic Database (NED). Among 59 candidates, 58 are located at the center of extended objects in the Pan-STARRS1 catalog, while one candidate has an offset. We present location, zz-band apparent magnitude, and time variability of the candidates and evaluate the probabilities that they are located inside of the 3D skymap of GW170817. The probability for J-GEM17btc is 64%64\% being much higher than those for the other 59 candidates (9.3×1032.1×101%9.3\times10^{-3}-2.1\times10^{-1}\%). Furthermore, the possibility, that at least one of the other 59 candidates is located within the 3D skymap, is only 3.2%3.2\%. Therefore, we conclude that J-GEM17btc is the most-likely and distinguished candidate as the optical counterpart of GW170817.Comment: 14 pages, 9 figures.     Japanese guidelines for atopic dermatitis 2020.

    Atopic dermatitis (AD) is a disease characterized by relapsing eczema with pruritus as a primary lesion, which is frequently encountered in clinical practice. Skin barrier dysfunction leads to enhanced skin irritability to non-specific stimuli and epicutaneous sensitization. In the lesion site, a further inflammation-related reduction in skin barrier function, enhanced irritability and scratching-related stimuli deteriorate eczema, leading to vicious cycle of inflammation. The current strategies to treat AD in Japan from the perspective of evidence-based medicine consist of three primary measures: (i) the use of topical corticosteroids and tacrolimus ointment as the main treatment for the inflammation; (ii) topical application of emollients to treat the cutaneous barrier dysfunction; and (iii) avoidance of apparent exacerbating factors, psychological counseling and advice about daily life. The guidelines present recommendations to review clinical research articles, evaluate the balance between the advantages and disadvantages of medical activities, and optimize medical activity-related patient outcomes with respect to several important points requiring decision-making in clinical practice

    Long‐term follow up of nivolumab in previously untreated Japanese patients with advanced or recurrent malignant melanoma

    The immune checkpoint inhibitor nivolumab inhibits the programmed death 1 receptor and suppresses the immune resistance of cancer cells. This is a long-term follow up of a single-arm, open-label, multicenter, phase II study of nivolumab in untreated Japanese patients with stage III/IV or recurrent melanoma. In addition, a post-hoc subgroup analysis stratified by melanoma types was performed. Nivolumab was administered intravenously at a dose of 3 mg/kg every 2 weeks. The primary endpoint was the overall response rate (ORR), and secondary endpoints included overall survival (OS), progression-free survival (PFS), best overall response, the disease control rate and change in tumor diameter. Safety was assessed by recording treatment-related adverse events (TRAE), including select immune-related adverse events. Of the 24 patients initially included in the primary phase II study, 10 survived for over 3 years (41.7%). The ORR was 34.8% (90% confidence interval [CI]: 20.8, 51.9) for all patients. When analyzing by melanoma type, the ORR was 66.7% (90% CI: 34.7, 88.3) for superficial spreading, 33.3% (90% CI: 11.7, 65.3) for mucosal, and 28.6% (90% CI: 10.0, 59.1) for acral lentiginous tumors. The median OS was 32.9 months, the 3-year OS rate was 43.5%, and the 3-year PFS rate was 17.2%. A long-term response was observed in all the tumor types. The most common TRAE included skin toxicity (45.8%) and endocrine disorders (29.2%). This study demonstrated the long-term efficacy and tolerability of nivolumab in patients with advanced or recurrent melanoma, irrespective of melanoma type

    Investigations of the radial propagation of blob-like structure in a non-confined electron cyclotron resonance heated plasma on Q-shu University Experiment with a Steady-State Spherical Tokamak

    A study of radial propagation and electric fields induced by charge separation in blob-like structures has been performed in a non-confined cylindrical electron cyclotron resonance heating plasma on Q-shu University Experiment with a Steady-State Spherical Tokamak using a fast-speed camera and a Langmuir probe. The radial propagation of the blob-like structures is found to be driven by E × B drift. Moreover, these blob-like structures were found to have been accelerated, and the property of the measured radial velocities agrees with the previously proposed model [C. Theiler et al., Phys. Rev. Lett. 103, 065001 (2009)]. Although the dependence of the radial velocity on the connection length of the magnetic field appeared to be different, a plausible explanation based on enhanced short-circuiting of the current path can be proposed

     本学部では,現在,カリキュラム改正を含め教育全般の質改革を行っている. その一環として,平成26 年度から新入生全員がタブレット端末(iPad)を購入・持参し,授業・学修における活用を開始した.今回,タブレット端末の導入とともに,電子教科書導入,LMS(学習管理システム:Learning Management System)の使用,教職員のスキルアップ,授業デザインの見直しなど,ICT(情報通信技術:Information and Communication Technology)を中心とした授業の質改革への取り組みを報告する
