    Limb Idleness Index (LII): a novel measurement of pain in a rat model of osteoarthritis

    SummaryObjectivesMechanical allodynia during ambulation in osteoarthritis (OA) animal models can be assessed as decreased extent of loading or decreased duration of loading. We propose to measure gait adaptation to pain by both mechanisms with the development of Limb Idleness Index (LII) in a rat model of knee OA.MethodsRats were assigned to anterior cruciate ligament transection (ACLT), Sham, or Normal group (n = 6). Gait data were collected at pre-injury, 1, 2, 3 and 6 months post-injury. Ratios of target print intensity, anchor print intensity, and swing duration were combined to obtain LII. The association of gait changes with pain was assessed by buprenorphine treatment at 3 and 6 months post-injury. At 6 months, OA-related structural changes in knee joints were examined by μCT and results from histological scoring were correlated with LII.ResultsAs compared to pre-injury level (range 0.75–1.20), LII in ACLT group was increased at 6 months post-injury, which was significantly higher than that in Sham and Normal groups (P = 0.024). The increase in LII in ACLT group was effectively reversed by buprenorphine treatment (P = 0.004). ACLT group exhibited a significantly higher maximum Osteoarthritis Research Society International (OARSI) score as compared to Sham (P = 0.005) and Normal (P = 0.006) groups. Significant correlation was found between LII and side-to-side difference in OARSI score (r = 0.893, P < 0.001).ConclusionsLII presents a good measurement for OA-related knee pain in rat model

    Modeling Web Services by Iterative Reformulation of Functional and Non-Functional Requirements

    Abstract. We propose an approach for incremental modeling of composite Web services. The technique takes into consideration both the functional and nonfunctional requirements of the composition. While the functional requirements are described using symbolic transition systems—transition systems augmented with state variables, function invocations, and guards; non-functional requirements are quantified using thresholds. The approach allows users to specify an abstract and possibly incomplete specification of the desired service (goal) that can be realized by selecting and composing a set of pre-existing services. In the event that such a composition is unrealizable, i.e. the composition is not functionally equivalent to the goal or the non-functional requirements are violated, our system provides the user with the causes for the failure, that can be used to appropriately reformulate the functional and/or non-functional requirements of the goal specification.

    Adiabatic evolution under quantum control

    One of the difficulties in adiabatic quantum computation is the limit on the computation time. Here we propose two schemes to speed-up the adiabatic evolution. To apply this controlled adiabatic evolution to adiabatic quantum computation, we design one of the schemes without any prior knowledge of the instantaneous eigenstates of the final Hamiltonian. Whereas in another scheme, the control is constructed with the instantaneous eigenstate that is the target state of the control. As an illustration, we study a two-level system driven by a time-dependent magnetic field under the control. The physics behind the control scheme is explained.Comment: 5 pages, 3 figure

    2D Mathematical Modeling For Fluvial Processes Considering The Influence Of Vegetation And Bank Erosion

    A 2D mathematical model for fluvial processes capable of considering the influence of vegetation and non-cohesive bank erosion is established based on a body-fitted coordinate system in this paper. The authors have improved a previously developed simulation model by taking into account the impact of vegetation with a vegetation stress term in the momentum conservation equation. A simple simulation method is adopted in the bank erosion model. Simulation runs were performed for a conceptual alluvial channel, the results of channel plan-form and cross section changes suggest that the 2D model predictions agree acceptable with the classic theories of channel pattern formation considering the effect of vegetation

    Single photon emitters based on Ni/Si related defects in single crystalline diamond

    We present investigations on single Ni/Si related color centers produced via ion implantation into single crystalline type IIa CVD diamond. Testing different ion dose combinations we show that there is an upper limit for both the Ni and the Si dose 10^12/cm^2 and 10^10/cm^2 resp.) due to creation of excess fluorescent background. We demonstrate creation of Ni/Si related centers showing emission in the spectral range between 767nm and 775nm and narrow line-widths of 2nm FWHM at room temperature. Measurements of the intensity auto-correlation functions prove single-photon emission. The investigated color centers can be coarsely divided into two groups: Drawing from photon statistics and the degree of polarization in excitation and emission we find that some color centers behave as two-level, single-dipole systems whereas other centers exhibit three levels and contributions from two orthogonal dipoles. In addition, some color centers feature stable and bright emission with saturation count rates up to 78kcounts/s whereas others show fluctuating count rates and three-level blinking.Comment: 7 pages, submitted to Applied Physics B, revised versio

    Mechanical control of nuclear import by Importin-7 is regulated by its dominant cargo YAP

    Mechanical forces regulate multiple essential pathways in the cell. The nuclear translocation of mechanoresponsive transcriptional regulators is an essential step for mechanotransduction. However, how mechanical forces regulate the nuclear import process is not understood. Here, we identify a highly mechanoresponsive nuclear transport receptor (NTR), Importin-7 (Imp7), that drives the nuclear import of YAP, a key regulator of mechanotransduction pathways. Unexpectedly, YAP governs the mechanoresponse of Imp7 by forming a YAP/Imp7 complex that responds to mechanical cues through the Hippo kinases MST1/2. Furthermore, YAP behaves as a dominant cargo of Imp7, restricting the Imp7 binding and the nuclear translocation of other Imp7 cargoes such as Smad3 and Erk2. Thus, the nuclear import process is an additional regulatory layer indirectly regulated by mechanical cues, which activate a preferential Imp7 cargo, YAP, which competes out other cargoes, resulting in signaling crosstalk.We thank Miguel Sánchez for text editing.
