Phylogenetic analysis of strains of Orf virus isolated from two outbreaks of the disease in sheep in Greece
<p>Abstract</p> <p>Background</p> <p>Although orf is endemic around the world, there are few descriptions of <it>Orf virus </it>strains and comparisons of these strains. We report the sequence and phylogenetic analysis of the partial B2L gene of <it>Orf virus </it>from two outbreaks of the disease in Greece. The first was an outbreak of genital form of the disease in a flock imported from France, whilst the second was an outbreak of the disease in the udder skin of ewes and around the mouth of lambs in an indigenous flock.</p> <p>Results</p> <p>Phylogenetic analysis was performed on a part (498 bp) of the B2L gene of 35 <it>Parapoxvirus </it>isolates, including the two <it>Orf virus </it>isolates recovered from each of the two outbreaks in the present study. This analysis revealed that the maximum nucleotide and amino-acid variation amongst <it>Orf virus </it>strains worldwide (n = 33) was 8.1% and 9.6%, respectively. The homology of the nucleotide and amino-acid sequences between the two Greek isolates was 99.0% and 98.8%, respectively. The two Greek isolates clustered only with <it>Orf virus </it>strains.</p> <p>Conclusions</p> <p>We suggest that there can be differences between strains based on their geographical origin. However, differences in the origin of strains or in the clinical presentation of the disease may not be associated with their pathogenicity. More work is required to determine if differing clinical presentations are linked to viral strain differences or if other factors, e.g., flock immunity, method of exposure or genetic susceptibility, are more important to determine the clinical presentation of the infection.</p
Correspondence of Somatic Cell Counts in Bulk-Tank Milk to Prevalence of Subclinical Mastitis in Sheep Flocks
The objective of the present study was to propose thresholds of somatic cell counts in bulk-tank milk indicative of the prevalence of subclinical mastitis in a flock. A retrospective analysis was performed on data from a longitudinal survey of subclinical mastitis in Greece, in which the prevalence of subclinical mastitis in 12 flocks sampled four times throughout a milking period was evaluated by collecting milk samples from individual ewes for bacteriological and cytological testing; further, cell counts in the bulk-tanks of the farms were also measured during the visits. Four cohorts were created: A, with cell counts in the bulk-tank milk between 0.100 × 106 and 0.400 × 106 cells mL−1, B, with cell counts between 0.400 × 106 and 650 × 106 cells mL−1, C, with cell counts between 0.650 × 106 and 900 × 106 cells mL−1, and D, with SCC between 0.900 × 106 and 1.450 × 106 cells mL−1. There was a significant positive correlation between prevalence of the infection in the flocks and somatic cell counts in bulk-tank milk on the same sampling occasion (p p < 0.0001). Ninety-five percent confidence intervals of the prevalence of subclinical mastitis according to the somatic cell counts in the bulk-tank milk were calculated as follows: for cohort A, 8.7% to 12.1%, for B, 12.4% to 19.4%, for C, 22.5% to 30.8% and for D, 27.3% to 45.3%. The information that may be thus obtained can be useful in guiding the implementation of various health management procedures for mastitis control in sheep flocks, with no need to perform milk sample collection from ewes and subsequent laboratory examinations
Clinical Aspects of Bacterial Distribution and Antibiotic Resistance in the Reproductive System of Equids
Antibiotic administration is a standard therapeutic practice for the treatment of reproductive disorders of equids. This might lead to undesirable microbial imbalance and could favour the acquisition of antibiotic resistance. Therefore, it is imperative for clinicians to understand patterns of antibiotic resistance when considering and developing treatment regimes. Continued engagement of clinicians with novel alternative approaches to treat reproductive infections would be essential in order to address this rising threat within the One Health perspective. The objectives of the present review were to present the bacterial infections in the reproductive system of equids (horses, donkeys), to upraise the literature related to the issue of antibiotic resistance of bacteria causing these infections and to discuss the topic from a clinical perspective. Initially, the review summarised the various infections of the reproductive system of equids (genital system of females, genital system of males, mammary glands) and the causal bacteria, providing relevant information about horses and donkeys. Subsequently, the clinical therapeutics of these infections were presented, taking into account the significance of antibiotic resistance of bacteria as a limiting factor in treating the infections. Finally, approaches to circumvent antibiotic resistance in clinical settings were summarized. It was concluded that awareness regarding antibiotic resistance in equine reproductive medicine would increase, as we would recognise the multifaceted problem of resistance. Actions and initiatives within the One Health approach, minimizing the potential dissemination of resistant strains to humans and to the environment, with specific applications in medicine of equids should be appropriately instituted internationally
Associations of Socio-Demographic Characteristics of Dairy Goat Farmers in Greece with Biosecurity-Related Practices in the Farms
Given the significance of goat farming in Greece, our study aimed to explore biosecurity conditions on goat farms and refers to an investigation performed on 119 farms countrywide in Greece. The objective of the present work was to evaluate and report on potential associations between the socio-demographic characteristics of goat farmers and practices that play a role in biosecurity on farms. Data were obtained during a large cross-sectional study performed across Greece in 119 goat herds. The socio-demographic characteristics of farmers, as well as 16 variables related to biosecurity, were recorded; these were practices or events that could potentially enhance or compromise biosecurity on farms. A score based on all the biosecurity-related variables was devised by assigning a ‘1’ or ‘−1’ mark for each variable recorded on a farm that could enhance or compromise, respectively, biosecurity. Among variables potentially enhancing biosecurity on farms, the application of disinfections on the farm and the maintenance of isolation for sick animals were practiced more frequently; among variables potentially compromising biosecurity on farms, the presence of hunters in the area around the farm and grazing practices for animals were recorded more frequently. The median overall score for biosecurity-related practices for all farms in the study was 1 (interquartile range: 2.5; minimum: −4; maximum: 5). Among socio-demographic characteristics, the presence of working staff on a farm was associated with five individual biosecurity-related practices and the length of farming experience with three individual biosecurity-related practices. In the multivariable analysis, female farmers (p = 0.007) and the presence of working staff on the farm (p = 0.025) emerged as the two significant socio-demographic characteristics with an association with the overall biosecurity level on farms. This knowledge can be useful when developing biosecurity programs on goat farms. Recognition of locally applied farm-level practices enhancing biosecurity could form a basis for farmers to apply more rigorous and effective relevant plans
Scientometrics Study of Research Output on Sheep and Goats from Greece
The study is a scientometrics evaluation of published articles performed in Greece on sheep and goats during the last 25 years, a period coinciding with implementation of reforms to shape and consolidate tertiary education and research establishments in the country. Objectives were: evaluation of the relevant publications and presentation of quantitative characteristics regarding scientific content and bibliometric details. The Web of Science platform was used (search terms: [[sheep OR goat*] OR [Ovis aries OR Capra hircus]] (1997–2022)) and 1080 papers were considered in detail. Throughout the study period, there was a clear progressive increase in numbers of papers published. The papers originated from 39 different entities, most from Aristotle University of Thessaloniki (34.0%), University of Thessaly (28.0%), Agricultural University of Athens (21.2%) and Hellenic Agricultural Organization—Dimitra (13.6%). Papers were published in 318 different journals. Journals with more published papers were Small Ruminant Research and Journal of the Hellenic Veterinary Medical Society. The most frequent general topic of study in the papers was health and welfare (46.7% of papers); second most frequent topic was animal products (18.6%). The papers have received 16,558 citations, i.e., οn average 15.4 citations per paper; the h-index was 56, the i10-index was 518 and the yearly citations per paper were 1.71. Papers on goats had higher impact than papers on sheep. There were 1711 individual authors, of which 728 were first or last authors. In total, 24 authors have each co-authored ≥2.5% of all papers; five authors were each first or last in that proportion of all papers. The findings have indicated that relevant research has focused on milk production and diseases of the udder of small ruminants; moreover, there was accumulation of relevant research in Greece in some establishments only. The findings of the study can be employed to initiate improved relevant research approaches in the country
Dairy Sheep and Goat Farmers: Socio-Demographic Characteristics and Their Associations with Health Management and Performance on Farms
The objectives were to determine the socio-demographic profiles of small ruminant dairy farmers and to study associations with management practices, production outcomes and health parameters on their farms. In total, 325 sheep flocks and 119 goat herds across Greece were included in the study and visited for collection of information. Socio-demographic characteristics of the dairy farmers and details of management practices, production outcomes and health parameters on their farms were recorded. For the analysis of results, multivariable models were created using mixed-effects logistic regression, with farms as the random effect. Most dairy farmers were male (93.2%), most were full-time professionals in farming (89.4%) and most had a farming family tradition (86.9%). The mean age was 47.0 years and the mean farming experience was 24.3 years. For 17.3% of the farmers, the highest level of education received was primary education, for 54.3% it was secondary education, for 16.4% it was post-secondary vocational training and for 12.0% it was tertiary education. In 35.4% of dairy farms, external farm workers were employed. Of the socio-demographic characteristics, farming experience was associated with geographical location of farms, management system, breed of animals, application of quarantine measures, laboratory evaluation of feedstuffs, ultrasonographic examination for pregnancy diagnosis, application of vaccination against clostridial infections, means of calculation of bodyweight for drug administration to animals, maintenance of colostrum bank, number of annual veterinary visits, annual milk production per animal, number of newborns and somatic cell counts and total bacterial counts in milk. Further, the employment of external farm workers on the farm was associated with management system, machine-milking, number and breed of animals, application of quarantine measures, laboratory evaluation of feedstuffs, ultrasonographic examination for pregnancy diagnosis, number of annual veterinary visits, annual milk production per animal, protein content in milk and number of newborns. The findings indicate that dairy sheep/goat farming is still a family-driven business, but, nevertheless, there are now younger people among these farmers, many of them with post-secondary education. Socio-demographic characteristics may influence the management practices applied, which in turn can have consequences for production and health results of the farms
Use of Antibiotics against Bacterial Infections on Dairy Sheep and Goat Farms: Patterns of Usage and Associations with Health Management and Human Resources
The objectives of the study were (a) to describe the patterns of antibiotic usage against four major clinical problems and (b) to evaluate factors that were associated with their use on small ruminant farms. Sheep and goat farmers mostly administered the antibiotics to animals at the dose prescribed (80.4%) and observed the necessary withdrawal period (98.7%), but fewer farmers (22.3%) weighed the animals to calculate their bodyweight before antibiotic administration. For the treatment of clinical mastitis, oxytetracycline, penicillin and streptomycin were the antibiotics used more frequently; 2.03 different antibiotics were used per sheep flock and 2.06 per goat herd, most frequently administered in injectable forms (88.8% of farms). In cases of abortion, oxytetracycline was administered more frequently; 1.12 different antibiotics were used per sheep flock and 1.03 per goat herd. In 94 farms (21.2%), routine administration of antibiotics was performed to newborns; oxytetracycline and ampicillin were administered more often. For the treatment of pneumonia in newborns, oxytetracycline, penicillin and tulathromycin were used more frequently; 1.33 antibiotics were used per sheep flock and 1.29 per goat herd. For the treatment of diarrhoea in lambs and kids, oxytetracycline, amoxicillin and penicillin were the antibiotics used more frequently; 1.34 antibiotics were used per sheep flock and 1.59 per goat herd. Results of multivariable analyses indicated 16 variables associated with the various outcomes for usage of antibiotics for the treatment of the above clinical problems. Of these, 11 variables were associated with the farmers: education of farmers was significant for three outcomes; the age, the experience, the professional involvement and farming family tradition of farmers and the daily period spent at the farm were each significant for one outcome
