    Endosymbiont dominated bacterial communities in a dwarf spider

    The microbial community of spiders is little known, with previous studies focussing primarily on the medical importance of spiders as vectors of pathogenic bacteria and on the screening of known cytoplasmic endosymbiont bacteria. These screening studies have been performed by means of specific primers that only amplify a selective set of endosymbionts, hampering the detection of unreported species in spiders. In order to have a more complete overview of the bacterial species that can be present in spiders, we applied a combination of a cloning assay, DGGE profiling and high-throughput sequencing on multiple individuals of the dwarf spider Oedothorax gibbosus. This revealed a co-infection of at least three known (Wolbachia, Rickettsia and Cardinium) and the detection of a previously unreported endosymbiont bacterium (Rhabdochlamydia) in spiders. 16S rRNA gene sequences of Rhabdochlamydia matched closely with those of Candidatus R. porcellionis, which is currently only reported as a pathogen from a woodlouse and with Candidatus R. crassificans reported from a cockroach. Remarkably, this bacterium appears to present in very high proportions in one of the two populations only, with all investigated females being infected. We also recovered Acinetobacter in high abundance in one individual. In total, more than 99% of approximately 4.5M high-throughput sequencing reads were restricted to these five bacterial species. In contrast to previously reported screening studies of terrestrial arthropods, our results suggest that the bacterial communities in this spider species are dominated by, or even restricted to endosymbiont bacteria. Given the high prevalence of endosymbiont species in spiders, this bacterial community pattern could be widespread in the Araneae order

    Polygyny and strong genetic structuring within an isolated population of the wood ant Formica rufa

    Social structuring of populations within some Formica species exhibits considerable variation going from monodomous and monogynous populations to polydomous, polygynous populations. The wood ant species Formica rufa appears to be mainly monodomous and monogynous throughout most of its distribution area in central and northern Europe. Only occasionally it was mentioned that F. rufa can have both polygynous and monogynous colonies in the same geographical region. We studied an isolated polydomous F. rufa population in a deciduous mixed forest in the north-west of Belgium. The level of poly-domy within the colonies varied from monodomous to 11 nests per colony. Our genetic analysis of eight variable microsatellites suggest an oligo- to polygynous structure for at least the major part of the sampled nests. Relatedness amongst nest mate workers varies considerable within the population and colonies but confirms in general a polygynous structure. Additionally high genetic diversity (e.g. up to 8 out of 11 alleles per nest for the most variable locus) and high within nest genetic variance (93%) indicate that multiple queens contribute to the gene pool of workers of the same nest. Moreover significant genetic structuring among colonies indicates that gene flow between colonies is restricted and that exchange of workers between colonies is very limited. Finally we explain how possible factors as budding and the absence of Serviformica can explain the differences in genetic structure within this polygynous F. rufa population

    Loss of genetic diversity and increased genetic structuring in response to forest area reduction in a ground dwelling insect: a case study of the flightless carabid beetle Carabus problematicus (Coleoptera, Carabidae)

    1.Old growth temperate broadleaved forests are characterised by a large proportion of forest specialists with low dispersal capability. Hence, species bound to this habitat are expected to be highly susceptible to the effects of decreasing patch size and increasing isolation. 2. Here, we investigate the relative effect of both factors by genotyping individuals of a flightless and forest specialist beetle Carabus problematicus from 29 populations, sampled in 21 different forest fragments in Belgium, at eight microsatellite loci. 3. A high degree of genetic differentiation among fragments was observed, with populations from smaller forests being considerably more differentiated and characterised by a lower genetic diversity compared to those of larger forests. 4. A more detailed study on forest remnants of a former historic continuous woodland area revealed that population differentiation was high among, but not within remnants, irrespective of geographical distance. This suggests that patch fragmentation rather than geographical distance is the ultimate factor that hampers gene flow in this species. 5. he results indicate that gene flow among suitable habitat patches is primarily reduced by the inability of this specialised species to traverse the landscape matrix. This lack of dispersal may pose a serious threat for the persistence of C. problematicus and ecologically similar species, and suggests that present populations can best be protected by securing or increasing the size of existing habitat patches

    Presentamos la primera revisión exhaustiva de los taxones de hormigas del archipiélago de Galápagos. La revisión se basa en datos recopilados durante búsquedas bibliográficas, encuestas de campo y la revisión de especímenes de museos, e incluye información sobre la distribución, hábitat y ecología de 47 especies. Se supone que al menos nueve de estas especies son nativas o endémicas de las islas y al menos 32 especies se han introducido a través de vías asociadas a los humanos. Se desconoce el estado de las seis especies restantes. La información compilada en este estudio nos permitirá comprender mejor el papel ecológico de las especies indígenas y su potencial como indicadores de la salud del ecosistema. La información de referencia sobre la distribución de hormigas introducidas será indispensable para evaluar el impacto y la propagación dentro del archipiélago.We present the first comprehensive review of the ant taxa of the Galápagos archipelago. The review is based on data gathered during literature searches, field surveys, and the revision ofmuseum specimens, and includes information on the distribution, habitat, and ecology of 47 species. At least nine of these species are assumed to be native or endemic to the islands and at least 32 species have been introduced through human-associated pathways. The status of the remaining six species is unknown. The information compiled in this study will allow us to better understand the ecological role of indigenous species and their potential as indicators of ecosystem health. Baseline information on the distribution of introduced ants will be indispensable for assessing impact and spread within the archipelago

    Patternize: An R Package For Quantifying Color Pattern Variation

    The use of image data to quantify, study and compare variation in the colors and patterns of organisms requires the alignment of images to establish homology, followed by color-based segmentation of images. Here we describe an R package for image alignment and segmentation that has applications to quantify color patterns in a wide range of organisms. patternize is an R package that quantifies variation in color patterns obtained from image data. patternize first defines homology between pattern positions across specimens either through manually placed homologous landmarks or automated image registration. Pattern identification is performed by categorizing the distribution of colors using an RGB threshold, k-means clustering or watershed transformation. We demonstrate that patternize can be used for quantification of the color patterns in a variety of organisms by analyzing image data for butterflies, guppies, spiders and salamanders. Image data can be compared between sets of specimens, visualized as heatmaps and analyzed using principal component analysis (PCA). patternize has potential applications for fine scale quantification of color pattern phenotypes in population comparisons, genetic association studies and investigating the basis of color pattern variation across a wide range of organisms.NSF grant DEB-1257839 NIH grant 5P20GM103475-1
