Production and characterization of β-glucosidase from Gongronella butleri by solid-state fermentation
Among the enzymes of the cellulolytic complex, β-glucosidases are noteworthy due to the possibility of their application in different industrial processes, such as production of biofuels, winemaking, and development of functional foods. This study aimed to evaluate the production and characterization of β-glucosidase from the filamentous fungus Gongronella butleri, recently isolated from Cerrado soil and cultivated in agro-industrial residue substrates. The highest production of β-glucosidase, about 215.4 U/g of dry substrate (or 21.5 U/mL), was obtained by cultivation of the microorganism on wheat bran with 55% of the initial moisture, for 96 h at 30°C. This β-glucosidase showed higher catalytic activity at pH 4.5, and a temperature of 65°C. The original enzymatic activity was recovered in a pH range of 3.0-7.5 after 24 h of incubation. The enzyme retained 80% of its catalytic activity when incubated for 1 h at 50°C. The enzyme was strongly inhibited by glucose, an effect that was completely reversed by increasing substrate concentration in the reaction mixture, which is typical for competitive inhibition. High catalytic activity was observed in solutions containing up to 20% ethanol, allowing the application of this enzyme in processes with high alcohol concentrations (for example beverages and biofuels). The significant production of β-glucosidase by the selected strain, along with these enzyme characteristics, highlights the biotechnological potential of the fungus G. butleri.Key words: Microbial enzyme, biofuels, agro-industrial residues, cellulases, hemicellulases
Memories of Mathematics from students of Youngster and Adult Programs
Little reflection has been devoted to the incorporation of previous school experiences of adult students that reenter basic education. This article focus on the communication of reminiscences of Mathematics made by students of Youngster and Adult Education, trying to reveal them as a fundamental component in the constitution of adult students as subjects not only of their learning of Mathematics, but of the schooling process itself. More than as mere references to concepts or procedures of Mathematics learned in other opportunities, reminiscences are viewed here as memory effects that permeate the production of meaning. The text focus on their communication in the context of discursive practices that shape (and take shape in) the teaching-learning processes of maths in Youngster and Adult Education, taking it as an organized social action that institutes the shared memories of school mathematics as arbiter of collective legitimacy. In analyzing excerpts of interactions between the researcher and students of Youngster and Adult Education from an class equivalent to the 5th grade of basic education, the semantic-pragmatic dimensions of the remembrance manifested in this context are highlighted, a remembrance that is considered conscious of itself, metacognitive, and structured in and by the discourse. By its sociocultural nature, this remembrance, turned collective, answers to a call of the present, rebuilding the meaning of the past.Pouco se tem refletido sobre a incorporação da experiência escolar anterior de alunos adultos que se re-inserem na Educação Båsica. Este artigo focaliza a enunciação de reminiscências da matemåtica escolar protagonizada por alunos da Educação de Jovens e Adultos (EJA), procurando revelå-las como um componente fundamental na constituição do aluno adulto como sujeito não só da aprendizagem da matemåtica, mas do próprio processo de escolarização. Mais do que meras referências a conceitos ou procedimentos de matemåtica aprendidos em outras oportunidades, compreendem-se, aqui, as reminiscências como efeitos da memória que permeiam a produção de sentido. O texto focaliza sua enunciação no âmbito das pråticas discursivas que conformam os (e se conformam nos) processos de ensino-aprendizagem da matemåtica na EJA, tomando-a como ação social organizada, que institui a lembrança compartilhada da matemåtica escolar como årbitro da legitimidade coletiva. Ao se analisarem trechos de interaçþes entre a pesquisadora e alunos que cursam o equivalente à 5ª sÊrie do ensino fundamental num projeto de EJA, destacam-se as dimensþes semântico-pragmåticas da rememoração que nesse contexto se manifesta e que se considera autoconsciente, metacognitiva, e estruturada no discurso e pelo discurso. Novel and conserved microRNAs in soybean floral whorls
AbstractMicroRNAs (miRNAs) correspond to a class of endogenous small non-coding RNAs (19â24nt) that regulates the gene expression, through mRNA target cleavage or translation inhibition. In plants, miRNAs have been shown to play pivotal roles in a wide variety of metabolic and biological processes like plant growth, development, and response to biotic and abiotic stress. Soybean is one of the most important crops worldwide, due to the production of oil and its high protein content. The reproductive phase is considered the most important for soybean yield, which is mainly intended to produce the grains. The identification of miRNAs is not yet saturated in soybean, and there are no studies linking them to the different floral organs. In this study, three different mature soybean floral whorls were used in the construction of sRNA libraries. The sequencing of petal, carpel and stamen libraries generated a total of 10,165,661 sequences. Subsequent analyses identified 200 miRNAs sequences, among which, 41 were novel miRNAs, 80 were conserved soybean miRNAs, 31 were new antisense conserved soybean miRNAs and 46 were soybean miRNAs isoforms. We also found a new miRNA conserved in other plant species, and finally one miRNA-sibling of a soybean conserved miRNA. Conserved and novel miRNAs were evaluated by RT-qPCR. We observed a differential expression across the three whorls for six miRNAs. Computational predicted targets for miRNAs analyzed by RT-qPCR were identified and present functions related to reproductive process in plants. In summary, the increased accumulation of specific and novel miRNAs in different whorls indicates that miRNAs are an important part of the regulatory network in soybean flower
Stationary solutions for the parity-even sector of the CPT-even and Lorentz-covariance-violating term of the standard model extension
In this work, we focus on some properties of the parity-even sector of the
CPT-even electrodynamics of the standard model extension. We analyze how the
six non-birefringent terms belonging to this sector modify the static and
stationary classical solutions of the usual Maxwell theory. We observe that the
parity-even terms do not couple the electric and magnetic sectors (at least in
the stationary regime). The Green's method is used to obtain solutions for the
field strengths E and B at first order in the Lorentz- covariance-violating
parameters. Explicit solutions are attained for point-like and spatially
extended sources, for which a dipolar expansion is achieved. Finally, it is
presented an Earth-based experiment that can lead (in principle) to an upper
bound on the anisotropic coefficients as stringent as

Issues on Generating Primordial Anisotropies at the End of Inflation
We revisit the idea of generating primordial anisotropies at the end of
inflation in models of inflation with gauge fields. To be specific we consider
the charged hybrid inflation model where the waterfall field is charged under a
U(1) gauge field so the surface of end of inflation is controlled both by
inflaton and the gauge fields. Using delta N formalism properly we find that
the anisotropies generated at the end of inflation from the gauge field
fluctuations are exponentially suppressed on cosmological scales. This is
because the gauge field evolves exponentially during inflation while in order
to generate appreciable anisotropies at the end of inflation the spectator
gauge field has to be frozen and scale invariant. We argue that this is a
generic feature, that is, one can not generate observable anisotropies at the

Potentiation of 5-fluorouracil encapsulated in zeolites as drug delivery systems for in vitro models of colorectal carcinoma
The studies of potentiation of 5-fluorouracil (5-FU), a traditional drug used in the treatment of several cancers, including colorectal (CRC), were carried out with zeolites Faujasite in the sodium form, with different particle sizes (NaY, 700nm and nanoNaY, 150nm) and Linde type L in the potassium form (LTL) with a particle size of 80nm. 5-FU was loaded into zeolites by liquid-phase adsorption. Characterization by spectroscopic techniques (FTIR, 1H NMR and 13C and 27Al solid-state MAS NMR), chemical analysis, thermal analysis (TGA), nitrogen adsorption isotherms and scanning electron microscopy (SEM), demonstrated the successful loading of 5-FU into the zeolite hosts. In vitro drug release studies (PBS buffer pH 7.4, 37°C) revealed the release of 80-90% of 5-FU in the first 10min. To ascertain the drug release kinetics, the release profiles were fitted to zero-order, first-order, Higuchi, Hixson-Crowell, Korsmeyer-Peppas and Weibull kinetic models. The in vitro dissolution from the drug delivery systems (DDS) was explained by the Weibull model. The DDS efficacy was evaluated using two human colorectal carcinoma cell lines, HCT-15 and RKO. Unloaded zeolites presented no toxicity to both cancer cells, while all DDS allowed an important potentiation of the 5-FU effect on the cell viability. Complexes of Iron(II) with silylated pentalene ligands; building blocks for homo- and heterobimetallics
A range of iron(II) complexes incorporating the silylated pentalene ligands (Pnâ H = 1,4-{SiiPr3}2C8H5 and Pnâ = 1,4-{SiiPr3}2C8H4) have been investigated as model molecules/building blocks for metallocene-based polymers. Six complexes have been synthesised and extensively characterised by a range of techniques, including by cyclic voltammetry and X-ray diffraction studies. Amongst these compounds are the homobimetallic [CpâFe]2(Îź-Pnâ ) which is a fused analogue of biferrocene, and the 3d/4s heterobimetallic [CpâFe(Ρ5-Pnâ )][K] which forms an organometallic polymer in the solid state. DFT calculations on model mono-Fe(Ρ5-Pn) compounds reveal the charge densities on the uncoordinated carbon atoms of the pentalene ligand, and hence the potential for incorporating these units into heteronuclear bimetallic complexes is assessed
Learning difficulties : a portuguese perspective of a universal issue
In this article we present findings of a study that was conducted with the purpose of deepening the knowledge about the field of learning difficulties in Portugal. Therefore, within these findings we will discuss across several cultural boundaries, themes related with the existence of learning difficulties as a construct, the terminology, the political, social and scientific influences on the field, and the models of identification and of ongoing school support for students. While addressing the above-mentioned themes we will draw attention to the different, yet converging, international understandings of learning difficulties
Search for a W' boson decaying to a bottom quark and a top quark in pp collisions at sqrt(s) = 7 TeV
Results are presented from a search for a W' boson using a dataset
corresponding to 5.0 inverse femtobarns of integrated luminosity collected
during 2011 by the CMS experiment at the LHC in pp collisions at sqrt(s)=7 TeV.
The W' boson is modeled as a heavy W boson, but different scenarios for the
couplings to fermions are considered, involving both left-handed and
right-handed chiral projections of the fermions, as well as an arbitrary
mixture of the two. The search is performed in the decay channel W' to t b,
leading to a final state signature with a single lepton (e, mu), missing
transverse energy, and jets, at least one of which is tagged as a b-jet. A W'
boson that couples to fermions with the same coupling constant as the W, but to
the right-handed rather than left-handed chiral projections, is excluded for
masses below 1.85 TeV at the 95% confidence level. For the first time using LHC
data, constraints on the W' gauge coupling for a set of left- and right-handed
coupling combinations have been placed. These results represent a significant
