Measurement of the Crab nebula polarization at 90 GHz as a calibrator for CMB experiments
CMB experiments aiming at a precise measurement of the CMB polarization, such
as the Planck satellite, need a strong polarized absolute calibrator on the sky
to accurately set the detectors polarization angle and the cross-polarization
leakage. As the most intense polarized source in the microwave sky at angular
scales of few arcminutes, the Crab nebula will be used for this purpose. Our
goal was to measure the Crab nebula polarization characteristics at 90 GHz with
unprecedented precision. The observations were carried out with the IRAM 30m
telescope employing the correlation polarimeter XPOL and using two orthogonally
polarized receivers. We processed the Stokes I, Q, and U maps from our
observations in order to compute the polarization angle and linear polarization
fraction. The first is almost constant in the region of maximum emission in
polarization with a mean value of alpha_Sky=152.1+/-0.3 deg in equatorial
coordinates, and the second is found to reach a maximum of Pi=30% for the most
polarized pixels. We find that a CMB experiment having a 5 arcmin circular beam
will see a mean polarization angle of alpha_Sky=149.9+/-0.2 deg and a mean
polarization fraction of Pi=8.8+/-0.2%.Comment: Accepted for publication in A&A, 9 pages, 4 figure
How should discrepancy be assessed in perfectionism research? A psychometric analysis and proposed refinement of the Almost Perfect Scale–Revised
Research on perfectionism with the Almost Perfect Scale-Revised (APS-R) distinguishes
adaptive perfectionists versus maladaptive perfectionists based primarily on their responses to
the 12-item unidimensional APS-R discrepancy subscale, which assesses the sense of falling
short of standards. People described as adaptive perfectionists have high standards but low levels
of discrepancy (i.e., relatively close to attaining these standards). Maladaptive perfectionists have
perfectionistic high standards and high levels of discrepancy. In the current work, we re-examine
the psychometric properties of the APS-R discrepancy subscale and illustrate that this
supposedly unidimensional discrepancy measure may actually consists of more than one factor.
Psychometric analyses of data from student and community samples distinguished a pure fiveitem
discrepancy factor and a second four-item factor measuring dissatisfaction. The five-item
factor is recommended as a brief measure of discrepancy from perfection and the four-item
factor is recommended as a measure of dissatisfaction with being imperfect. Overall, our results
confirm past suggestions that most people with maladaptive perfectionism are characterized
jointly by chronic dissatisfaction as well as a sense of being discrepant due to having fallen short
of expectations. These findings are discussed in terms of their implications for the assessment of
perfectionism, as well as the implications for research and practice
A person-oriented approach to multidimensional perfectionism: perfectionism profiles in health and well-being
Advances in understanding of the perfectionism construct have been limited by an almost exclusive reliance on a variable-centered approach. This study utilized a person-oriented approach to examine Hewitt and Flett’s conceptualization of multidimensional perfectionism in relation to health and well-being. Levels of conscientiousness, extraversion, and neuroticism were also assessed. Cluster analyses were employed to examine within-person configurations of self-oriented, other-oriented, and socially prescribed perfectionism (SPP) in university students (n = 538) and adults with chronic illness (n = 773). Five unique configurations were found in both samples and three clusters replicated across samples. “Extreme perfectionists” with high scores across all perfectionism dimensions reported relatively poor physical health, psychological health, psychosocial resources, and well-being along with elevated neuroticism and conscientiousness. A group distinguished by elevated SPP also reported relatively poorer outcomes along with elevated neuroticism and lower conscientiousness. In contrast, “nonperfectionists” reported relatively elevated levels of health and well-being. These profiles differed in their links with health and well-being even after taking into account key differences in conscientiousness and neuroticism. Our results illustrate the importance of employing a person-oriented approach to the study of multidimensional perfectionism, especially as it relates to physical health, mental health, and subjective well-being
New pleiotropic effects of eliminating a rare tRNA from Streptomyces coelicolor, revealed by combined proteomic and transcriptomic analysis of liquid cultures
Background: In Streptomyces coelicolor, bldA encodes the only tRNA for a rare leucine codon, UUA. This tRNA is unnecessary for growth, but is required for some aspects of secondary metabolism and morphological development. We describe a transcriptomic and proteomic analysis of the effects of deleting bldA on cellular processes during submerged culture: conditions relevant to the industrial production of antibiotics.
Results: At the end of rapid growth, a co-ordinated transient up-regulation of about 100 genes, including many for ribosomal proteins, was seen in the parent strain but not the ΔbldA mutant. Increased basal levels of the signal molecule ppGpp in the mutant strain may be responsible for this difference. Transcripts or proteins from a further 147 genes classified as bldA-influenced were mostly expressed late in culture in the wild-type, though others were significantly transcribed
during exponential growth. Some were involved in the biosynthesis of seven secondary metabolites; and some have probable roles in reorganising metabolism after rapid growth. Many of the 147 genes were "function unknown", and may represent unknown aspects of Streptomyces biology. Only two of the 147 genes contain a TTA codon, but some effects of bldA could be traced to TTA codons in regulatory genes or polycistronic operons. Several proteins were affected posttranslationally by the bldA deletion. There was a statistically significant but weak positive global
correlation between transcript and corresponding protein levels. Different technical limitations of the two approaches were a major cause of discrepancies in the results obtained with them.
Conclusion: Although deletion of bldA has very conspicuous effects on the gross phenotype, the
bldA molecular phenotype revealed by the "dualomic" approach has shown that only about 2% of the genome is affected; but this includes many previously unknown effects at a variety of different levels, including post translational changes in proteins and global cellular physiology
Global spectral energy distribution of the Crab Nebula in the prospect of the Planck satellite polarisation calibration
Whithin the framework of the Planck satellite polarisation calibration, we
present a study of the Crab Nebula spectral energy distribution (SED) over more
than 6 decades in frequency ranging from 1 to . The Planck
satellite mission observes the sky from 30 to 857 GHz and therefore we focus on
the millimetre region. We use radio and submillimetre data from the WMAP
satellite between 23 and 94 GHz (from 13 to 3.18 mm) and from the Archeops
balloon experiment between 143 (2.1 mm) and 545 GHz (0.55 mm), and a compendium
of other Crab Nebula observations. The Crab SED is compared to models including
three main components : synchrotron which is responsible for the emission at
low and at high frequencies, dust which explains the excess of flux observed by
the IRAS satellite and an extra component on the millimetre regime. From this
analysis we conclude that the unpolarised emission of the Crab Nebula at
microwave and millimetre wavelengths is the same synchrotron emission that the
one observed in the radio domain. Therefore, we expect the millimetre emission
of the Crab nebula to be polarised with the same degree of polarisation and
orientation than the radio emission. We set upper limits on the possible errors
induced by any millimetre extra component on the reconstruction of the degree
and angle of polarisation at the percent level as a maximum. This result
strongly supports the choice by the Planck collaboration of the Crab nebula
emission for performing polarisation cross-checks in the range 30 (299 mm) to
353 GHz (0.849 mm)
Sociobiological Control of Plasmid copy number
Background:
All known mechanisms and genes responsible for the regulation of plasmid replication lie with the plasmid rather than the chromosome. It is possible therefore that there can be copy-up mutants. Copy-up mutants will have within host selective advantage. This would eventually result into instability of bacteria-plasmid association. In spite of this possibility low copy number plasmids appear to exist stably in host populations. We examined this paradox using a computer simulation model.

Model:
Our multilevel selection model assumes a wild type with tightly regulated replication to ensure low copy number. A mutant with slightly relaxed replication regulation can act as a “cheater” or “selfish” plasmid and can enjoy a greater within-host-fitness. However the host of a cheater plasmid has to pay a greater cost. As a result, in host level competition, host cell with low copy number plasmid has a greater fitness. Furthermore, another mutant that has lost the genes required for conjugation was introduced in the model. The non-conjugal mutant was assumed to undergo conjugal transfer in the presence of another conjugal plasmid in the host cell.

Results:
The simulatons showed that if the cost of carrying a plasmid was low, the copy-up mutant could drive the wild type to extinction or very low frequencies. Consequently, another mutant with a higher copy number could invade the first invader. This process could result into an increasing copy number. However above a certain copy number within-host selection was overcompensated by host level selection leading to a rock-paper-scissor (RPS) like situation. The RPS situation allowed the coexistence of high and low copy number plasmids. The non-conjugal “hypercheaters” could further arrest the copy numbers to a substantially lower level.

Conclusions:
These sociobiological interactions might explain the stability of copy numbers better than molecular mechanisms of replication regulation alone
The Big Three Perfectionism Scale: A new measure of perfectionism
This article introduces a new measure of dispositional perfectionism: the Big Three Perfectionism Scale (BTPS). The BTPS assesses three higher-order global factors (rigid perfectionism, self-critical perfectionism, narcissistic perfectionism) via 10 lower-order perfectionism facets (self-oriented perfectionism, self-worth contingencies, concern over mistakes, doubts about actions, self-criticism, socially prescribed perfectionism, other-oriented perfectionism, hypercriticism, grandiosity, entitlement). The present investigation examined the structure of the BTPS using exploratory factor analysis in Study 1 (288 undergraduates), and confirmatory factor analyses in Study 2 (352 community adults) and Study 3 (290 undergraduates). Additionally, in Study 3 the relationships among the BTPS, other measures of perfectionism, and the five-factor model of personality were investigated. Overall, findings provide first evidence for the reliability and validity of the BTPS as a multidimensional measure of perfectionism
Synthetic RNA Silencing of Actinorhodin Biosynthesis in Streptomyces coelicolor A3(2)
We demonstrate the first application of synthetic RNA gene silencers in Streptomyces coelicolor A3(2). Peptide nucleic acid and expressed antisense RNA silencers successfully inhibited actinorhodin production. Synthetic RNA silencing was target-specific and is a new tool for gene regulation and metabolic engineering studies in Streptomyces.Peer reviewe
