    Nel panorama nazionale si discute poco degli autori dei reati sessuali, infatti l’attenzione dell’opinione pubblica e dei diversi attori sociali è più rivolta alle vittime dei reati sessuali. Ancora poco si affronta il problema legato al trattamento dei pedofili e dei violentatori di donne, nonostante l’argomento meriti approfondimenti anche in ambito penitenziario. Appare necessario ed opportuno individuare, nell’ambito del lavoro trattamentale, un modello operativo finalizzato alla ricerca di strategie che orientino il condannato per reati sessuali verso un percorso di analisi degli agiti. Ciò assume una rilevanza particolare nel trattamento dei detenuti sex offenders, pur riconoscendo la valenza del percorso trattamentale rivolto a qualsiasi tipologia di detenuti. L’esperienza realizzata presso la Casa Circondariale di Castelvetrano, descritta nel presente articolo, ha consentito, attraverso l’uso di una metodologia di lavoro di gruppo, di rilevare a carico dei soggetti partecipanti un quadro di consistente difficoltà nei percorsi di approfondimento e di analisi personale. In Italy, there has been little discussion about sex offenders. In fact, public and social actors’ attention has focused more on the victims of sex crimes rather than on criminals, so little importance is given to resocialization programs for paedophiles and rapists, even if this topic is worth exploring particularly in prisons. In the treatment phase, it is necessary and considered appropriate to identify an operational model aimed to orientate the sex offenders towards a process of analysis of their deviant behaviours. Through the methodology of the experiential group, the project carried out at Castelvetrano prison has highlighted the severe difficulties in personal analysis encountered by the sex offenders involved

    822 Local radiotherapy synergizes with tumor-specific TCR redirected T cells in the rejection of prostate cancer

    Background Adoptive T cell therapy (ACT) has become a promising option for cancer patients. While tumor-infiltrating lymphocytes were initially exploited as a source of tumor reactive lymphocytes, T cells genetically redirected to the tumor by TCR/CAR gene transfer are now in clinical validation. In the case of solid tumors, unfavorable immunosuppressive microenvironments remain recognized barriers to therapeutic efficacy. We have recently reported that the therapeutic activity of ACT against poorly immunogenic and indolent prostate cancer is improved by the concurrent targeting of the tumor stroma by mean of T cells redirected to an ubiquitously expressed minor histocompatibility antigen or a tumor vessel targeted TNF derivative. We have now taken the concept further and hypothesized that local radiotherapy (RT), might also synergize with ACT by promoting lymphocyte endothelial transmigration and tumor recognition, and ultimately favor abscopal effects. Methods We investigated the combination of local RT and ACT in TRAMP (Transgenic Adenocarcinoma of the Mouse Prostate) mice and in mice bearing subcutaneous B16/B16-OVA (MO4) or TRAMP-C2/TRAMP-C2-OVA tumors. Local RT was delivered by X-RAD SmART (the Small Animal Radiation Therapy) microirradiator in single dose or hypo-fractioned regimens. ACT consisted of T cells engineered with tumor-specific TCRs. Immunogenic consequences were analyzed by Real-Time PCR, and flow cytometry (FACS) analyses. Prostate tumor debulking was evaluated by histological analyses. Results We found that local hypofractionated RT and ACT, while individually inefficacious in controlling tumor growth, concurred to the debulking of advanced prostate adenocarcinoma when used in combination in treating TRAMP mice. Mechanistically, exposing isolated tumor cells, or the TRAMP mouse prostate to hypo-fractionated RT regimens induced stronger type-I interferon (IFN-I) responses, when compared to single high dose. Acutely, hypofractionated RT promoted better immune tumor infiltration, among which TCR redirected effector cells. Conclusions Data support feasibility and efficacy of combining hypo-fractionated local RT with ACT in the form of TCR engineered T cells to promote prostate cancer recognition and eradication. Tumor debulking was observed in the absence of treatment-related toxicity. Systemic recirculation of TCR redirected T cells was observed. We are now investigating therapeutic effects at distal (metastatic) sites.     El libro recoge una colección de artículos producto de investigaciones adelantadas por docentes del Departamento de Humanidades y cuyo centro temático es la relación entre las personas y la felicidad. Cada capítulo brinda respuestas desde un ámbito específico disciplinar, a través de una metodología cualitativa, al problema antropológico y ético de la consecución de la felicidad o la realización humana personal. Desde la educación y la ética se plantea la transición de unas humanidades informativas a otras performativas, que integren formación moral y valores sobrenaturales, que aboguen por la empatía y la solidaridad como camino humano de felicidad. A partir de las claves antropológicas de Leonardo Polo se presenta a la persona como ser capaz de dotar de sentido su modo de estar en el mundo, más allá de la propia satisfacción felicitaria, pues ella misma es un sentido personal capaz de manifestarse en la tarea esperanzada. Así mismo, desde la antropología personalista se estudia la felicidad como proyecto de vida, pasando de la conflictividad hacia la espiritualidad y proponiendo decantadas transformaciones político-educativas. En el ámbito de las ciencias históricas se analiza el uso de los conceptos de persona y felicidad en el Magisterio de Juan xxiii subrayando las perspectivas sugeridas por el Papa y recogidas por los pontífices sucesivos.     Pathogenetics of alveolar capillary dysplasia with misalignment of pulmonary veins.

    Alveolar capillary dysplasia with misalignment of pulmonary veins (ACDMPV) is a lethal lung developmental disorder caused by heterozygous point mutations or genomic deletion copy-number variants (CNVs) of FOXF1 or its upstream enhancer involving fetal lung-expressed long noncoding RNA genes LINC01081 and LINC01082. Using custom-designed array comparative genomic hybridization, Sanger sequencing, whole exome sequencing (WES), and bioinformatic analyses, we studied 22 new unrelated families (20 postnatal and two prenatal) with clinically diagnosed ACDMPV. We describe novel deletion CNVs at the FOXF1 locus in 13 unrelated ACDMPV patients. Together with the previously reported cases, all 31 genomic deletions in 16q24.1, pathogenic for ACDMPV, for which parental origin was determined, arose de novo with 30 of them occurring on the maternally inherited chromosome 16, strongly implicating genomic imprinting of the FOXF1 locus in human lungs. Surprisingly, we have also identified four ACDMPV families with the pathogenic variants in the FOXF1 locus that arose on paternal chromosome 16. Interestingly, a combination of the severe cardiac defects, including hypoplastic left heart, and single umbilical artery were observed only in children with deletion CNVs involving FOXF1 and its upstream enhancer. Our data demonstrate that genomic imprinting at 16q24.1 plays an important role in variable ACDMPV manifestation likely through long-range regulation of FOXF1 expression, and may be also responsible for key phenotypic features of maternal uniparental disomy 16. Moreover, in one family, WES revealed a de novo missense variant in ESRP1, potentially implicating FGF signaling in the etiology of ACDMPV

    Este libro recoge una serie de artículos de investigación escritos por docentes y estudiantes que participan en los semilleros de investigación del Departamento de Humanidades de la Universidad Católica de Colombia. El centro temático de la obra son las reflexiones en relación con la persona, la intimidad y la sociedad. Cada capítulo brinda respuestas, desde un específico ámbito disciplinar, a través de una metodología cualitativa, al problema antropológico y ético fundamental del cultivo de la intimidad o interioridad personal y sus repercusiones en la vida social. En concreto, los textos estudian la familia como ámbito de intimidad con inspiración en la antropología trascendental de Leonardo Polo; enfocan su atención en el problema de reducir a la persona a productor y analizan la intimidad como apertura a lo espiritual, camino para recuperar una vida solidaria y fraterna; describen el papel de la intimidad entendida como gestora de grandes acontecimientos en el desarrollo histórico de Occidente, a partir del pensamiento de Agustín de Hipona; analizan el tema de la intimidad en el Magisterio de Pablo VI; abordan el concepto de intimidad familiar como lugar donde se recibe formación en valores humanos y religiosos; y presentan la relación que existe entre intimidad y desplazamiento. Con estas investigaciones se espera contribuir a la reflexión humanística alrededor del tema de la intimidad y estimular nuevas pautas de investigación que permitan entender mejor el difícil mundo en que vivimos.
