    A semi-schematic model for the center of mass dynamics in supercooled molecular liquids

    We introduce a semi-schematic mode-coupling model to describe the slow dynamics in molecular liquids, retaining explicitly only the description of the center of mass degrees of freedom. Angular degrees of freedom are condensed in a q-vector independent coupling parameter. We compare the time and q-dependence of the density fluctuation correlators with numerical data from a 250 ns long molecular dynamics simulation. Notwithstanding the choice of a network-forming liquid as a model for comparing theory and simulation, the model describes the main static and dynamic features of the relaxation in a broad q-vector range.Comment: Revtex, 2 figure

    Bypassing cellular EGF receptor dependence through epithelial-to-mesenchymal-like transitions

    Over 90% of all cancers are carcinomas, malignancies derived from cells of epithelial origin. As carcinomas progress, these tumors may lose epithelial morphology and acquire mesenchymal characteristics which contribute to metastatic potential. An epithelial-to-mesenchymal transition (EMT) similar to the process critical for embryonic development is thought to be an important mechanism for promoting cancer invasion and metastasis. Epithelial-to-mesenchymal transitions have been induced in vitro by transient or unregulated activation of receptor tyrosine kinase signaling pathways, oncogene signaling and disruption of homotypic cell adhesion. These cellular models attempt to mimic the complexity of human carcinomas which respond to autocrine and paracrine signals from both the tumor and its microenvironment. Activation of the epidermal growth factor receptor (EGFR) has been implicated in the neoplastic transformation of solid tumors and overexpression of EGFR has been shown to correlate with poor survival. Notably, epithelial tumor cells have been shown to be significantly more sensitive to EGFR inhibitors than tumor cells which have undergone an EMT-like transition and acquired mesenchymal characteristics, including non-small cell lung (NSCLC), head and neck (HN), bladder, colorectal, pancreas and breast carcinomas. EGFR blockade has also been shown to inhibit cellular migration, suggesting a role for EGFR inhibitors in the control of metastasis. The interaction between EGFR and the multiple signaling nodes which regulate EMT suggest that the combination of an EGFR inhibitor and other molecular targeted agents may offer a novel approach to controlling metastasis

    Hypoxia increases mutational load of breast cancer cells through frameshift mutations

    Tumor hypoxia-induced downregulation of DNA repair pathways and enhanced replication stress are potential sources of genomic instability. A plethora of genetic changes such as point mutations, large deletions and duplications, microsatellite and chromosomal instability have been discovered in cells under hypoxic stress. However, the influence of hypoxia on the mutational burden of the genome is not fully understood. Here, we attempted to elucidate the DNA damage response and repair patterns under different types of hypoxic stress. In addition, we examined the pattern of mutations exclusively induced under chronic and intermittent hypoxic conditions in two breast cancer cell lines using exome sequencing. Our data indicated that hypoxic stress resulted in transcriptional downregulation of DNA repair genes which can impact the DNA repair induced during anoxic as well as reoxygenated conditions. In addition, our findings demonstrate that hypoxic conditions increased the mutational burden, characterized by an increase in frameshift insertions and deletions. The somatic mutations were random and non-recurring, as huge variations within the technical duplicates were recognized. Hypoxia also resulted in an increase in the formation of potential neoantigens in both cell lines. More importantly, these data indicate that hypoxic stress mitigates DNA damage repair pathways and causes an increase in the mutational burden of tumor cells, thereby interfering with hypoxic cancer cell immunogenicity

    The short-sequence designs of isochores from the human genome

    The human genome, a typical mammalian genome, is made up of long (≈1-Mb, on average) regions, the isochores, that are fairly homogeneous in base composition and belong in five families characterized by different GC levels. An analysis of di- and tri-nucleotide densities in the isochores from the five families has shown large differences. These different “short-sequence designs:” (i) account for the fractionation of human DNA (and vertebrate DNA in general) when using sequence-specific ligands in density gradients, (ii) are very similar in whole isochores and in the corresponding intergenic sequences and introns, (iii) are reflected in different codon usages, (iv) lead to amino acid differences that increase the thermal stability of the proteins encoded by genes located in increasingly GC-rich isochore families, and (v) correspond to different chromatin structures

    L'histoire des justices militaires a jusqu'il y a peu été largement négligée. Institution hybride, elle a longtemps fait l'objet de jugements à l'emporte-pièce, tant chez les militaires que chez les juristes. On a ainsi vu se multiplier critiques acerbes et plaidoyers pro domo autour de cette institution si particulière. Une série de séminaires menés dans le cadre du projet de la Maison des sciences de l'Homme, de 2004 à 2008, avait pour objectif de parcourir, en perspective comparée, les évolutions de la justice militaire depuis le XVIe siècle. Le présent volume reprend reprend une vingtaine de ces contributions, orientées sur le premier XXe siècle, pour comprendre les tensions, les pratiques et les limites de la justice militaire pendant et autour des deux guerres mondiales. Parcourant l'Europe occidentale, il se veut méthodologique et initiateur, éclairant une réalité transnationale à l'aide d'études de cas, inscrites dans le temps et l'espace.Until recently, the history of military justice has been largely neglected. As a hybrid concept, it has for many years been the subject of hasty judgments from both military institutions and lawyers. Members of the military institutions kept repeating harsh criticisms while the lawyers kept justifying themselves. This volume brings together 20 contributions that focus on the early 20th century in Western Europe with the aim of examining the tensions, practices and limitations of military justice during the two World Wars. In doing so, it uses a methodological and original approach, from the perspective of transnational reality and uses case studies from a range of countries at different times. The contributions were all presented during multiple seminars held at the Maison des sciences de l'Homme between 2004 and 2008. Using a comparative perspective, these seminars explored the evolution of the military justice system since the 16th century
