Composição química e atividade biológica de espécies de nectandra e stevia rebaudiana
The growing of the interest in natural products, the study about native medicinal plants has been relevant as the phytochemical constituents and biological activities, in contrast to Europe plants, which have been extensively studied. This study was divided into three articles, the theme of the first two articles was the biochemical characterization of native species of Nectandra genre. For a year have been collected, in the middle of each season, leaves and bark of the species Nectandra lanceolata, Nectandra grandiflora and Nectandra megapotâmica in the Edgardia Farm and Lageado Farm belonging to the Campus Botucatu - UNESP, located in Botucatu, Sao Paulo. The first article aimed to evaluate the effect of seasonality in phenolic content and antioxidant activity in leaves and barck of Nectandras. The results showed that the sampling time influenced the presence of phenolic compounds in the species in different parts of the plant, giving greater prominence to the species N. megapotamica. As for the antioxidant activity whose species was evidence N. lanceolata. The second article assessed the influence of seasonality in content, chemical and antimicrobial activity of strains of Escherichia coli sp. and Staphylococcus aureus sp. The results of this study showed that species N. grandiflora and N. lanceolata showed a lot of similarity in their volatile compounds during the year, differently of N. megapotamica. The species N. megapotamica and N. lanceolata proved promising on the antibacterial activity. The third article, accomplished during Sanduiche Doctoral stage in Padova, aimed to evaluate the effect of different doses of macronutrients (NPK) on the yield and quality of Stevia rebaudiana Berth. The experiment was conducted during the summer season in Italy in greenhouse, with a factorial split-splot 3 x 3 x3.
Seasonality effects on chemical composition, antibacterial activity and essential oil yield of three species of Nectandra
In this study, we aimed to determine whether seasonality affects the content, chemical composition, and antimicrobial activity of essential oils (EOs) from the leaves of three species of Nectandra (Nectandra megapotamica, Nectandra grandiflora, and Nectandra lanceolata) native to the Atlantic rainforest, Sao Paulo state, Brazil. In addition, we identified the compounds potentially related to the antimicrobial activity. Leaves were randomly collected in the middle of winter (August), spring (November), summer (February), and autumn (May). The influence of seasonality on the content and chemical composition of EOs from the Nectandra species was evident in this study. The EOs from N. lanceolata and N. grandiflora were characterized by similarities in the chemical composition and had a higher relative proportion of oxygenated sesquiterpenes. N. megapotamica presented a different chemical profile, with plenty of monoterpenic and sesquiterpenic hydrocarbons. Changes in the EO chemical profile because of seasonality were shown by the similarities between the EOs obtained in spring and autumn and the differences between the EOs obtained in summer and winter. The EO from the leaves of N. megapotamica harvested in winter and spring showed the highest control of the growth of Escherichia coli, and this antimicrobial action can be related to the monoterpenes \u3b1-pinene and \u3b2-pinene as well as myrcene and limonene. The minimum inhibitory concentration (MIC) of the EO from the leaves of N. lanceolata harvested in summer and autumn was lower against the gram-positive bacterium Staphylococcus aureus and can be related to the sesquiterpene hydrocarbons isobicyclogermacrenal, epi-zizanone, and germacrene B
Storage and stability of IgG and IgM monoclonal antibodies dried on filter paper and utility in <it>Neisseria meningitidis </it>serotyping by Dot-blot ELISA
Abstract Background A simple filter paper method was developed for, the transport and storage of monoclonal antibodies (Mabs) at room temperature or -20°C after spotting on filter paper, for subsequent serotyping of outer membrane antigens of N.meningitidis by dot-blot ELISA. Methods Monoclonal antibodies (Mabs) were spotted within a 0.5–1 cm diameter area of Whatman grade 903 paper, which were stored individually at room temperature or at -20°C. These MAbs were stored and analyzed after periods of one week, 4 weeks, 12 months, or 13 years in the case of frozen Mab aliquots, or after 4 weeks at -20°C or at room temperature (RT) in the case of Mabs dried on filter paper strips. Assays were performed in parallel using dot-blot ELISA. In addition to the MAbs specific for serotyping class 1, 2 or 3, we used a larger number of Mabs for polysaccharides, lipooligosaccharides (LOS), class 5 and cross-reactive antigens for native outer membrane of N.meningitidis. The Mabs dried on filter paper were eluted with phosphate-buffered saline (PBS) containing 0.2% gelatin. Results Mabs of the isotypes IgG and IgM dried on filter papers were not affected by duration of storage. The detection by serotyping Mabs was generally consistent for dried filter paper MAb samples stored frozen for over 1 year at -20°C, and although decreased reactive antibody titers were found after storage, this did not interfere with the specificity of the Mabs used after 13 years as dry spots on filter paper. Conclusion The use of filter paper is an inexpensive and convenient method for collecting, storing, and transporting Mab samples for serotyping studies. In addition, the samples occupy little space and can be readily transported without freezing. The efficiency of using immunoglobulin G (IgG) or M (IgM) eluted was found to be consistent with measurement of IgG or IgM titers in most corresponding, ascites Mabs stored frozen for over 1 year. The application of meningococcal typing methods and designations depend on the question being asked.</p
