Effects of Climate Change on Peatland Reservoirs: A DOC Perspective
Peatland reservoirs are global hotspots for drinking water provision and are likely to become more important as demand per capita rises and the climate changes. Dissolved organic carbon (DOC) is associated with harmful disinfection byproducts and reduced aesthetic quality, and its removal is the major treatment cost. Littoral zones are known to be disproportionately important for DOC production through macrophyte inputs, and such communities are predicted to expand with warming in northern regions. However, little is known about autochthonous DOC contributions and their response to climatic change. Here we exposed mesocosms to elevated CO2 (eCO2), warming and a combined treatment across a trophic gradient. Regression analysis indicated that while sediments, macrophytes, and phytoplankton are important DOC sources (P < 0.05), benthic algal biomass showed the strongest relationship with DOC (P < 0.05), suggesting it is an underestimated source. DOC removal indicators, namely phenol oxidase (depolymerization) and respiration (mineralization) were inversely related to DOC concentration in oligohumic (P < 0.05) and oligotrophic (P < 0.1) systems, suggesting heterotrophic processes are important in DOC removal. DOC concentrations increased across all systems (P < 0.05), irrespective of trophic status, due to increased photoautotrophic inputs (macrophyte, pelagic, and benthic algae) under eCO2, warming, and combined scenarios, with inhibited depolymerization and mineralization under eCO2, even when combined with warming (P < 0.05 and P < 0.05 excepting the oligo-mesotrophic reservoir P < 0.1 respectively). Increased DOC loads of all fractions, regardless of provenance, are predicted in a future climate and, thus, investment in techniques to remove a greater range of DOC fractions is proposed to help “future proof” drinking water supplies
Min-sum 2-paths problems
An orientation of an undirected graph G is a directed graph obtained by replacing each edge {u,v} of G by exactly one of the arcs (u,v) or (v,u). In the min-sum k -paths orientation problem, the input is an undirected graph G and ordered pairs (s i ,t i ), where i∈{1,2,…,k}. The goal is to find an orientation of G that minimizes the sum over all i∈{1,2,…,k} of the distance from s i to t i . In the min-sum k edge-disjoint paths problem, the input is the same, however the goal is to find for every i∈{1,2,…,k} a path between s i and t i so that these paths are edge-disjoint and the sum of their lengths is minimum. Note that, for every fixed k≥2, the question of N P-hardness for the min-sum k-paths orientation problem and for the min-sum k edge-disjoint paths problem has been open for more than two decades. We study the complexity of these problems when k=2. We exhibit a PTAS for the min-sum 2-paths orientation problem. A by-product of this PTAS is a reduction from the min-sum 2-paths orientation problem to the min-sum 2 edge-disjoint paths problem. The implications of this reduction are: (i) an NP-hardness proof for the min-sum 2-paths orientation problem yields an NP-hardness proof for the min-sum 2 edge-disjoint paths problem, and (ii) any approximation algorithm for the min-sum 2 edge-disjoint paths problem can be used to construct an approximation algorithm for the min-sum 2-paths orientation problem with the same approximation guarantee and only an additive polynomial increase in the running time
Small changes in water levels and groundwater nutrients alter nitrogen and carbon processing in dune slack soils
Dune slacks are biodiverse seasonal wetlands which experience considerable fluctuation in water table depth. They are under threat from lowered water tables due to climate change and water abstraction and from eutrophication. The biological effects caused by the interactions of these pressures are poorly understood, particularly on soil processes. We used a mesocosm experiment and laboratory assays to study the impact of lowered water tables, groundwater nitrogen contamination, and their synergistic effects on soil microbial processes and greenhouse gas emissions. This study showed that just a 10 cm decrease in water table depth led to a reduction in denitrification and to a corresponding increase in soil nitrogen content. Meanwhile N2O emissions occurred for longer durations within dune slack soils subject to higher concentrations of groundwater nitrogen contamination. The results from extracellular enzyme assays suggest that decomposition rates increase within drier soils shown by the increase in β-glucosidase activity, with further sensitivity to groundwater nitrogen contamination shown by the increase in phenol oxidase activity. Dune slack soils with a 10 cm lower water table had significantly lower methane emissions, nearly 5 times lower in the drier soils. Our findings demonstrate that dune slacks are sensitive to both small changes in groundwater levels and to groundwater nitrogen contamination. The biological impacts from lowered water tables are likely to be intensified where there is also groundwater nitrogen contamination
Impact of distinct poxvirus infections on the specificities and functionalities of CD4+ T cell responses.
UNLABELLED: The factors that determine CD4+ T cell (TCD4+) specificities, functional capacity, and memory persistence in response to complex pathogens remain unclear. We explored these parameters in the C57BL/6 mouse through comparison of two highly related (\u3e92% homology) poxviruses: ectromelia virus (ECTV), a natural mouse pathogen, and vaccinia virus (VACV), a heterologous virus that nevertheless elicits potent immune responses. In addition to elucidating several previously unidentified major histocompatibility complex class II (MHC-II)-restricted epitopes, we observed many qualitative and quantitative differences between the TCD4+ repertoires, including responses not elicited by VACV despite complete sequence conservation. In addition, we observed functional heterogeneity between ECTV- and VACV-specific TCD4+ at both a global and individual epitope level, particularly greater expression of the cytolytic marker CD107a from TCD4+ following ECTV infection. Most striking were differences during the late memory phase where, in contrast to ECTV, VACV infection failed to elicit measurable epitope-specific TCD4+ as determined by intracellular cytokine staining. These findings illustrate the strong influence of epitope-extrinsic factors on TCD4+ responses and memory.
IMPORTANCE: Much of our understanding concerning host-pathogen relationships in the context of poxvirus infections stems from studies of VACV in mice. However, VACV is not a natural mouse pathogen, and therefore, the relevance of results obtained using this model may be limited. Here, we explored the MHC class II-restricted TCD4+ repertoire induced by mousepox (ECTV) infection and the functional profile of the responding epitope-specific TCD4+, comparing these results to those induced by VACV infection under matched conditions. Despite a high degree of homology between the two viruses, we observed distinct specificity and functional profiles of TCD4+ responses at both acute and memory time points, with VACV-specific TCD4+ memory being notably compromised. These data offer insight into the impact of epitope-extrinsic factors on the resulting TCD4+ responses
Light nuclei in galactic globular clusters : constraints on the self-enrichment scenario from nucleosynthesis
Hydrogen-burning is the root cause of the star-to-star abundance variations
of light nuclei in Galactic globular clusters (GC). In the present work we
constrain the physical conditions that gave rise to the observed abundance
patterns of Li, C, N, O, Na, Mg, Al, as well as Mg isotopes in the typical case
of NGC6752. We perform nucleosynthesis calculations at constant temperature,
adopting realistic initial abundances for the proto-cluster gas. We use a
detailed nuclear reaction network and state-of-the-art nuclear reaction rates.
Although simplistic, our analysis provides original results and new constraints
on the self-enrichment scenario for GCs. Our parametrized calculations allow us
to determine a narrow range of temperature where the observed extreme
abundances of all light elements and isotopes in NGC6752 are nicely reproduced
simultaneously. This agreement is obtained after mixing of the H-processed
material with 30 % of unprocessed gas. The observed C-N, O-Na, Mg-Al, Li-Na and
F-Na anticorrelations, as well as the behaviour of the Mg isotopes can be
recovered by assuming mixing with even larger dilution factors. Li production
by the stars that build up the other abundance anomalies is not mandatory in
the case of NGC 6752. Observations of O, Na, Mg and Al constrain the
temperature range for H-burning; such temperatures are encountered in the two
main candidate ``polluters'' proposed for GCs, namely massive AGBs and the most

Grain Boundary Induced Magneto-Far Infrared Resonances in Superconducting YBaCuO Thin Films
Spectral features induced by 45 in-plane misoriented grains have
been observed in the far infrared magneto-transmission of YBaCuO thin films. Two strong dispersive features are found at 80 and
160 and a weaker one at 116 . The data can be well
represented by Lorentzian oscillator contributions to the conductivity. Several
possible interpretations are discussed. We conclude that the resonances are due

Epithelial Immunization Induces Polyfunctional CD8+ T Cells and Optimal Mousepox Protection.
We assessed several routes of immunization with vaccinia virus (VACV) in protecting mice against ectromelia virus (ECTV). By a wide margin, skin scarification provided the greatest protection. Humoral immunity and resident-memory T cells notwithstanding, several approaches revealed that circulating, memory CD8(+) T cells primed via scarification were functionally superior and conferred enhanced virus control. Immunization via the epithelial route warrants further investigation, as it may also provide enhanced defense against other infectious agents
An Introduction to Gas Accretion onto Galaxies
Evidence for gas accretion onto galaxies can be found throughout the
universe. In this chapter, I summarize the direct and indirect signatures of
this process and discuss the primary sources. The evidence for gas accretion
includes the star formation rates and metallicities of galaxies, the evolution
of the cold gas content of the universe with time, numerous indirect indicators
for individual galaxies, and a few direct detections of inflow. The primary
sources of gas accretion are the intergalactic medium, satellite gas and
feedback material. There is support for each of these sources from observations
and simulations, but the methods with which the fuel ultimately settles in to

Shedding light on the elusive role of endothelial cells in cytomegalovirus dissemination.
Cytomegalovirus (CMV) is frequently transmitted by solid organ transplantation and is associated with graft failure. By forming the boundary between circulation and organ parenchyma, endothelial cells (EC) are suited for bidirectional virus spread from and to the transplant. We applied Cre/loxP-mediated green-fluorescence-tagging of EC-derived murine CMV (MCMV) to quantify the role of infected EC in transplantation-associated CMV dissemination in the mouse model. Both EC- and non-EC-derived virus originating from infected Tie2-cre(+) heart and kidney transplants were readily transmitted to MCMV-naïve recipients by primary viremia. In contrast, when a Tie2-cre(+) transplant was infected by primary viremia in an infected recipient, the recombined EC-derived virus poorly spread to recipient tissues. Similarly, in reverse direction, EC-derived virus from infected Tie2-cre(+) recipient tissues poorly spread to the transplant. These data contradict any privileged role of EC in CMV dissemination and challenge an indiscriminate applicability of the primary and secondary viremia concept of virus dissemination
