    Digestibilidade aparente de rações contendo complexo enzimático para tilápia-do-nilo

    Avaliou-se a digestibilidade aparente de cinco rações isoproteicas e isoenergéticas contendo 0; 0,1; 0,2; 0,3 e 0,4g do complexo enzimático Bioenzimaplus (lipase, protease e carboidrase)/kg de ração na alimentação da tilápia-do-nilo. Foram utilizados 100 peixes distribuídos em cinco aquários de alimentação e cinco aquários de coleta de fezes. Os cinco tratamentos foram arranjados em delineamento inteiramente ao acaso com cinco repetições. Houve efeito linear de tratamento sobre a digestibilidade aparente (CDa) para proteína bruta e extrato etéreo. O CDa da proteína bruta e do extrato etéreo variaram de 81,60% a 84,93% e de 74,19% a 82,69%, respectivamente. Para carboidrato e energia bruta, o maior nível de suplementação resultou em maior CDa, 57,85% e 63,78%, respectivamente( P<0,05). Não foi observada diferença entre tratamentos no CDa da matéria seca, apresentando valores de 49,47% a 58,55%. A inclusão de complexo enzimático - lipase, protease e carboidrase - em dietas para tilápia-do-nilo melhorou a digestibilidade da proteína, do extrato etéreo, do carboidrato e da energia das rações.Apparent digestibility (ADC) of five isoproteic and isoenergetic diets containing 0, 0.1, 0.2, 0.3, and 0.4g/kg of enzymatic complex Bioenzimaplus (lipase, protease, and carbohydrase) for Nile tilapia was evaluated. A hundred fish were randomly assigned in five feeding aquaria, and five collecting feces ones. Treatments were composed by five enzymatic complex levels arranged in a completely randomized design in five repetitions. Significant differences (P<0.05) were observed with the inclusion of enzymatic complex in the diets for ADC of crude protein and fat (linear effect). ADC of crude protein and fat varied from 81.60% to 84.94% and 74.19% to 85.69%, respectively. The highest enzyme supplementation level showed better digestibility values for carbohydrate and gross energy (57.85% and 63.78%, respectively). No difference was observed for ADC of dry matter showing values between 49.47% and 58.55%. The use of enzymes in Nile tilapia diets improved the digestibilities of protein, fat, carbohydrate, and gross energy digestibility

    Experimental study of a vertical column of grains submitted to a series of impulses

    We report physical phenomena occurring in a vertical Newton's cradle system. A dozen of metallic spheres are placed in a vertical tube. Therefore, the gravity induces a non-uniform pre-compression of the beads and a restoring force. An electromagnetic hammer hits the bottom bead at frequencies tuned between 1 and 14Hz. The motion of the beads are recorded using a high-speed camera. For low frequencies, the pulses travel through the pile and expel a few beads from the surface. Then, after a few bounces of these beads, the system relaxes to the chain of contacting grains. When the frequency is increased, the number of fluidized beads increases. In the fluidized part of the pile, adjacent beads are bouncing in opposition of phase. This phase locking of the top beads is observed even when the bottom beads experience chaotic motions. While the mechanical energy increases monotically with the bead vertical position, heterogeneous patterns in the kinetic energy distribution are found when the system becomes fluidized

    Requirement of the N-Terminal Extension for Vacuolar Trafficking and Transport Activity of Yeast Ycf1p, an ATP-binding Cassette Transporter

    Ycf1p is the prototypical member of the yeast multidrug resistance-associated protein (MRP) subfamily of ATP-binding cassette (ABC) transporters. Ycf1p resides in the vacuolar membrane and mediates glutathione-dependent transport processes that result in resistance to cadmium and other xenobiotics. A feature common to many MRP proteins that distinguishes them from other ABC transporters is the presence of a hydrophobic N-terminal extension (NTE), whose function is not clearly established. The NTE contains a membrane spanning domain (MSD0) with five transmembrane spans and a cytosolic linker region (L0). The goal of this study was to determine the functional significance of the NTE of Ycf1p by examining the localization and functional properties of Ycf1p partial molecules, expressed either singly or together. We show that MSD0 plays a critical role in the vacuolar membrane trafficking of Ycf1p, whereas L0 is dispensable for localization. On the other hand, L0 is required for transport function, as determined by monitoring cadmium resistance. We also examine an unusual aspect of Ycf1p biology, namely, the posttranslational proteolytic processing that occurs within a lumenal loop of Ycf1p. Processing is shown to be Pep4p dependent and thus serves as a convenient marker for proper vacuolar localization. The processed fragments associate with each other, suggesting that these natural cleavage products contribute together to Ycf1p function

    Resposta hemática de tilápias-do-nilo alimentadas com dietas suplementadas com colina e submetidas a estímulo por baixa temperatura Hematic response of Nile tilapia fed diets supplemented with choline and submitted to stimulus by low temperature

    Esta pesquisa foi realizada com o objetivo de avaliar a resposta hemática de tilápias-do-nilo (Oreochromis niloticus) arraçoadas com dietas suplementadas com colina e submetidas a estímulo por baixa temperatura. O período experimental foi realizado em duas etapas: a primeira, de 109 dias, e a segunda, de 7 dias. Durante a primeira etapa, foram utilizados 192 alevinos com peso médio inicial de 4 g, distribuídos em 32 tanques-rede de 200 L instalados em aquários de mil litros. As rações foram formuladas de modo a apresentar 28,0% de proteína digestível e 3.100,0 kcal ED/kg e mesma concentração de aminoácidos. O delineamento experimental foi inteiramente casualizado com oito tratamentos e quatro repetições. As rações foram suplementadas com colina (cloreto de colina 60,0%), de modo a apresentar 100,0; 200,0; 400,0; 600,0; 800,0; 1.000,0 e 1.200,0 mg/kg de ração, e avaliadas em comparação a uma ração sem suplementação. Após o período de 109 dias, foram efetuadas as análises hematológicas dos peixes. Após as análises, os peixes foram transferidos para a sala de desafio e distribuídos em 24 aquários, onde foram mantidos a 17ºC durante sete dias. Após esse período, foram feitas as mesmas análises do período anterior ao desafio. A suplementação de colina não influenciou a eritropoiese ao estímulo pelo frio. A suplementa��ão dietética de colina não interfere na síntese de eritrócitos e leucócitos e a temperatura de 17,0ºC determina linfopenia e neutrofilia.<br>The aim of this study was to evaluate the hematic response of Nile tilapia (Oreochromis niloticus) fed diets supplemented with choline and submitted to temperature stress. The experimental period was realized in two phases: the first, during 109 days, and the second, for seven days. During the first stage, 192 fingerlings with average initial weight of 4 g were distributed in 32 net cages (200 L) allocated in 1,000-L aquaria. The diets were formulated to present 28% of digestible protein and 3,100 kcal DE/kg and the same concentration of amino acids. It was used a complete random experimental design with eight treatments and four replicates. The diets were choline supplemented (60% choline chloride) in order to present 100.0; 200.0; 400.0; 600.0; 800.0; 1,000.0 and 1,200.0 mg/kg of diet and evaluated by comparing to a non-supplemented diet. After the 109-day period, the hematological analyses of the fish were performed. After these analyzes, fish were transferred to the challenge room, distributed in 24 aquaria, and kept at 17ºC during seven days. After this period, the same analyzes of the period previous to the challenge were done. Choline supplementation did not affected erythropoiesis to stimulus by the cold temperature. Dietary choline supplementation does not affect erythrocyte and leukocyte synthesis and the 17ºC temperature determines lymphopenia and neutrophilia
