    Blood flow cytometry in Mugil cephalus and Carassius auratus: a comparative study

    Blood and its parameters can give specific indications on the welfare of fishes.Many endogenous and exogenous factors exert influences on the characteristics of blood. The correct interpretation of fish hematology for a given species depends on the availability of reference values. The purpose of the present study was therefore to build a database with data on haematological profiles of Mugil cephalus (Linnaeus 1758) and goldfish Carassius auratus (Linnaeus 1758), in particular of some blood cells, namely red blood cells (RBC), white blood cells (WBC) and thrombocytes (TC). All the blood parameters studied showed significant differences in the two fish species considered, using flow cytometry and optical microscopy coupled with an automated system. In particular, RBC showed an increase in mullets in respect to goldfish, while WBC and TC decreased. The differences found may be due to the environmental conditions and the different eating habits of the two species. The results of this research will allow to better understand how the different dietary habits and environmental conditions can influence the haematological parameters of fishes. Flow cytometry represents a modern diagnostic technique in human. Moreover, the technique used by combining flow cytometry with automated haematological counting, has proved very effective in the early evaluation of haematological parameters of various fish species

    Our review delves into the realm of peroral endoscopic myotomies (POEMs) in the upper gastrointestinal tract (UGT). In recent years, POEMs have brought about a revolution in the treatment of UGT motility disorders. Esophageal POEM, the first to be introduced, has now been validated as the primary treatment for achalasia. Subsequently developed, G-POEM displays promising results in addressing refractory gastroparesis. Over time, multiple endoscopic myotomy techniques have emerged for the treatment of Zenker’s diverticulum, including Z-POEM, POES, and hybrid approaches. Despite the well-established efficacy outcomes, new challenges arise in the realm of POEMs in the UGT. For esophageal POEM, the future scenario lies in customizing the myotomy extent to the minimum necessary, while for G-POEM, it involves identifying patients who can optimally benefit from the treatment. For ZD, it is crucial to validate an algorithm that considers various myotomy options according to the diverticulum’s size and in relation to individual patients. These challenges align with the concept of precision endoscopy, personalizing the technique for each subject. Within our text, we comprehensively examine each myotomy technique, analyzing indications, outcomes, and adverse events. Additionally, we explore the emerging challenges posed by myotomies within the context of the evolving field of precision endoscopy

    The work aimed to study the induction of morphological alterations in M. galloprovincialis in the field and its suitability to be integrated into a sensitive, simple, and cost-effective cell-based multimarker approach for the detection of the stress status induced by pollution in coastal marine environments in view of ecotoxicological biomonitoring and assessment application. Cellular morphometric alterations was paralleled by the analysis of standardized biomarkers such as lysosomal membrane destabilization, and genotoxocity biomarkers such as micronuclei and binuclated cells frequencies were investigated. The study was carried out by means of a transplanting experiment in the field, using caged organisms from an initial population exposed in the field in two multi-impacted coastal sites of the central Mediterranean area, Bagnoli in the eastern Tyrrhenian Sea and Augusta-Melilli-Priolo in the western Ionian Sea. Capo Miseno (NA) for the Tyrrhenian area and Brucoli (ME) for the Ionian area were chosen as control sites. Hemocyte enlargement and filopodial elongation increased frequencies were observed in organisms exposed to the impacted sites. These morphometric alterations showed strong agreement with the lysosomal membrane destabilization and biomarkers of genotoxicity, suggesting their usefulness in detecting the pollutant-induced stress syndrome related to genotoxic damage
