    Metagenetic and Volatilomic Approaches to Elucidate the Effect of Lactiplantibacillus plantarum Starter Cultures on Sicilian Table Olives

    The present study aimed to evaluate the effect of selected Lactiplantibacillus plantarum strains on both microbiota composition and volatile organic compound profile of Sicilian table olives. Two mixed cultures, named O1 and O2, were set up for pilot-plan scale fermentations at 5% of NaCl. Uninoculated table olives at 5 and 8% (C5 and C8) of salt were used as control. The fermentation process was monitored until 80 days through a dual approach, which included both classical microbiological and 16S amplicon-based sequencing and volatilomics analyses. Compared with control samples (C5 and C8), experimental samples, inoculated with starter cultures (O1 and O2), exhibited a faster acidification with a more pronounced drop in pH. Metagenetics data revealed significant differences of microbiota composition among samples, highlighting the dominance of lactobacilli in both experimental samples; a high occurrence of Enterobacter genus only in control samples with 5% of NaCl; and the presence of Bacteroides, Faecalibacterium, Klebsiella, and Raoultella genera only in control samples with 8% of NaCl. Furthermore, microbiota composition dynamics, through the fermentation process, significantly affected the volatile organic compounds of the final products, whereas no compounds involved in off-odors metabolites were detected in all samples investigated. In conclusion, the addition of the proposed starter cultures and the use of low concentrations of sodium chloride positively affected the microbiota and volatile organic compounds, ensuring the microbiological safety and the pleasant flavors of the final product

    Evidenza di un caso di epatite infettiva del cane in Sicilia

    L’Adenovirus Canino di tipo 1 (CAV-1), appartenente al genere Mastadenovirus, famiglia Adenoviridae, è l’agente causale dell’epatite infettiva del cane (ICH). Il virus replica negli endoteli vascolari e negli epatociti, causando epatite necrotico emorragica acuta. I sintomi neurologici sono rari e sono causati da una vasculite del SNC. Negli ultimi anni, la diffusa vaccinazione ha ridotto la circolazione di CAV-1 ed i casi clinici segnalati sono diventati rari ed isolati. In questo lavoro, gli Autori descrivono un caso clinico in un cucciolo di 2 mesi, non vaccinato, con febbre, prostrazione, convulsioni, vomito, diarrea ed esito infausto in una settimana. A seguito dell’esame autoptico, sono stati prelevati campioni di vari organi (cuore, milza, polmone, rene, encefalo, fegato, intestino). Questi sono stati analizzati mediante PCR ed isolamento su colture cellulari sensibili, per la ricerca dei principali agenti virali causa di malattia nel cane: parvovirus, cimurro, CAV-1 e CAV-2, coronavirus, rotavirus. Tutti gli organi esaminati sono risultati positivi sia in PCR che all’isolamento su MDCK solo per CAV-1. Il genoma estratto è stato amplificato secondo la metodica descritta da Hu et al. (2001), in grado di discriminare CAV-1 da CAV-2. L’isolamento su MDCK ha prodotto effetto citopatico al secondo passaggio ed il virus isolato è stato identificato mediante immunofluorescenza diretta e PCR. Il presente lavoro costituisce un contributo alla esigua disponibilità bibliografica sulle infezioni da CAV-1. La vaccinazione sistematica condotta nell’ultimo decennio ha sensibilmente ridotto i casi di malattia. Infatti, gli ultimi casi risalgono al 2001 in Basilicata e Puglia dove sono stati descritti rispettivamente un caso di CAV-1 con sintomatologia classica ed un altro caratterizzato anche da sintomi neurologici. La presente indagine dimostra l’attuale circolazione di CAV-1 e la sporadica comparsa di casi clinici. La vaccinazione sistematica con CAV-2 rimane il mezzo più efficace di protezione della popolazione canina e costituisce l’unico metodo di controllo della malattia

    Archaeometric Study of Two Tanagra Type Statuettes of Unknown Provenance to Support Forensic Study

    This paper is concerned with a morphological-stylistic and archaeometric study of two small pottery statues, confiscated by the Cosenza Carabinieri Unit for the Protection of Cultural Heritage and Anti-Counterfeiting (Calabria, Italy). The research aimed to establish the authenticity of the artworks and to verify a possible origin from the same workshop manufacturing, by providing indications about the textural features and raw materials used for their production. For these purposes, the analytical approach involved the use of minero-petrographic and physical analysis, as follows: petrographic analysis (OM), X-ray diffraction (XRD) and thermoluminescence tests (TL). The preliminary observation, which highlights differences in the stylistic features of the two statuettes as well as in the color, morphology and distribution of the white-greyish patina, is further confirmed by the TL investigations. The TL test revealed an ancient production only for one of the analyzed finds and the investigations on the raw materials allowed to relate this to a possible local historical-artistic context. The second statuette, on the other hand, is attributable to a modern production as confirmed by TL measurement

    This paper addresses an archaeometric study of a marble bas-relief seized by the Cosenza Carabinieri Unit for the Protection of Cultural Heritage and Anti-Counterfeiting (Calabria, Italy). The research aimed to collect data on the authenticity of the artwork, providing indications about the compositional features of the sampled materials. An analytical approach based on the use of SEM-EDX and FT-IR techniques along with a stylistic evaluation of the artwork, allowed us to answer the requests posed by the institutions and to set the work in a different historical context from the Roman production

    Effects of different stress parameters on growth and on oleuropein-degrading abilities of lactiplantibacillus plantarum strains selected as tailored starter cultures for naturally table olives

    The use of β-glucosidase positive strains, as tailored-starter cultures for table olives fermentation, is a useful biotechnological tool applied to accelerate the debittering process. Nowadays, strains belonging to Lactiplantibacillus plantarum species are selected for their high versatility and tolerance to stress conditions. The present study investigated the effect of different stress factors (pH, temperature and NaCl) on growth and on oleuropein-degrading abilities of selected L. plantarum strains. In addition, the presence of the beta-glucosidase gene was investigated by applying a PCR based approach. Results revealed that, overall, the performances of the tested strains appeared to be robust toward the different stressors. However, the temperature of 16 °C significantly affected the growth performance of the strains both singularly and in combination with other stressing factors since it prolongs the latency phase and reduces the maximum growth rate of strains. Similarly, the oleuropein degradation was mainly affected by the low temperature, especially in presence of low salt content. Despite all strains displayed the ability to reduce the oleuropein content, the beta-glucosidase gene was detected in five out of the nine selected strains, demonstrating that the ability to hydrolyze the oleuropein is not closely related to the presence of beta-glucosidase. Data of the present study suggest that is extremely important to test the technological performances of strains at process conditions in order to achieve a good selection of tailored starter cultures for table olives

    Respiratory Syncytial Virus: New Challenges for Molecular Epidemiology Surveillance and Vaccination Strategy in Patients with ILI/SARI.

    Abstract: Several respiratory pathogens are responsible for influenza-like illness (ILI) and severe respiratory infections (SARI), among which human respiratory syncytial virus (hRSV) represents one of the most common aetiologies. We analysed the hRSV prevalence among subjects with ILI or SARI during the five influenza seasons before the emergence of SARS-CoV-2 epidemic in Sicily (Italy). Respiratory specimens from ILI outpatients and SARI inpatients were collected in the framework of the Italian Network for the Influenza Surveillance and molecularly tested for hRSV-A and hRSV-B. Overall, 8.1% of patients resulted positive for hRSV. Prevalence peaked in the age-groups <5 years old (range: 17.6–19.1%) and ≥50 years old (range: 4.8–5.1%). While the two subgroups co-circulated throughout the study period, hRSV-B was slightly predominant over hRSV-A, except for the season 2019–2020 when hRSV-A strongly prevailed (82.9%). In the community setting, the distribution of hRSV subgroups was balanced (47.8% vs. 49.7% for hRSV-A and hRSV-B, respectively), while most infections identified in the hospital setting were caused by hRSV-B (69.5%); also, this latter one was more represented among hRSV cases with underlying diseases, as well as among those who developed a respiratory complication. The molecular surveillance of hRSV infections may provide a valuable insight into the epidemiological features of ILI/SARI. Our findings add new evidence to the existing knowledge on viral aetiology of ILI and SARI in support of public health strategies and may help to define high-risk categories that could benefit from currently available and future vaccines

    Performance evaluation of a commercial protective coating through field-exposure tests on three stone substrates

    In the last decades, there have been several studies on Cultural Heritage regarding the performance of protective and consolidating coatings for the prevention of decay. A coating must have several characteristics such as efficiency, breathability, and must be durable and reversible. In this research work, the performance of a commercial protective product (Fosbuild FBLE 200) was evaluated. This coating is composed of a TiO2 nanopowder dispersed in an aqueous solution of an acrylic polymer. The product, which exhibits depolluting, antimicrobial, water-repellent and self-cleaning properties, has been applied on three different lithotypes: Carrara marble, Noto stone, and Comiso stone. Field-exposure tests were carried out in two different outdoor environments (Catania and Palermo) in order to assess its suitability. Promising results were obtained for the Carrara marble after one year of exposure; however, a decrease in effectiveness was observed at the end of the second year

    A Multi-Analytical Diagnostic on an Outdoor Wall Painting: The Study on the Déesis of St. Maria Annunziata’s Church, Motta San Giovanni (Reggio Calabria, Italy)

    This article concerns the diagnostic campaign aimed at analyzing the mural painting representing the iconographic theme of the Deesis of the Church of St. Maria Annunziata, Motta San Giovanni, in the province of Reggio Calabria. In 1951, a flood caused the collapse of the building and the consequent breaking of the apse into two parts. The present study focused on the left side of the apse, hosting the figures of Christ and Mary, in order to plan the best conservation intervention strategy. For this purpose, non-invasive investigations and laboratory analytical methods were conducted in order to characterize the constituent materials and to identify the forms of alteration and degradation present on the surface of the painting. In particular, Raman spectroscopy, optical microscopy, scanning electron microscopy coupled to the chemical analysis by an EDS probe, Fourier-transform infrared spectroscopy and ion chromatography were employed. The results highlighted the presence of a single layer of plaster made with a lime-based binder. The chromatic palette of the painting is characterized by ochres and carbon black mixed with lime to obtain the different shades. Finally, the definition of the nature of the deposits and of the overlaid materials was fundamental in order to identify the best products and methods to restore the readability of the work
