Prognostic and therapeutic considerations of antibodies against c-ter apolipoprotein A-1 in the general population.
Autoantibodies against apolipoprotein A1 (anti-apoA1 IgGs) and its C-terminal region (cter apoA1) have emerged as an independent biomarker for cardiovascular disease. Cter apoA1 mimetic peptides were shown to reverse the deleterious anti-apoA1 IgG effects in vitro. We evaluated the association of anti-cter apoA1 IgGs with overall mortality in the general population and tested the ability of a cter apoA1 mimetic peptide to reverse the anti-apoA1 IgG-induced inflammatory response and mortality in vitro and in vivo, respectively.
Anti-cter apoA1 IgGs were measured in serum samples of 6386 participants of the CoLaus study of which 5220 were followed for a median duration of 5.6 years. The primary outcome was overall mortality. The peptide inhibitory concentration 50% (IC <sub>50</sub> ) was determined in vitro on HEK-Blue-4 and RAW cells. ApoE <sup>-/-</sup> mice were exposed to 16 weeks of anti-apoA1IgG passive immunisation with and without peptide co-incubation.
Anti-cter apoA1 IgGs were associated with higher interleukin 6 levels and independently predicted overall mortality; an increase of one standard deviation of anti-cter apoA1 IgG level was associated with an 18% increase in mortality risk (hazard ratio: 1.18, 95% confidence interval: 1.04-1.33; P = 0.009). The cterApoA1 analogue reversed the antibody-mediated inflammatory response with an IC <sub>50</sub> of 1 µm in vitro but did not rescue the significant anti-apoA1 IgG-induced mortality rate in vivo (69% vs. 23%, LogRank P = 0.02).
Anti-cter apoA1 IgG independently predicts overall mortality in the general population. Despite being effective in vitro, our cter apoA1 analogue did not reverse the anti-apoA1 IgG-induced mortality in mice. Our data suggest that these autoantibodies are not readily treatable through cognate peptide immunomodulation

Size Effects of Brittle Particles in Aerosol Deposition—Molecular Dynamics Simulation
Copyright © The Author(s) 2021. Up to now, the role of particle sizes on the impact behavior of ceramic particles in aerosol deposition not yet fully understood. Hence, with the aim to supply a more general understanding, modeling series of low strain rate compression and high-speed impact were performed by molecular dynamics on single-crystalline particles in sizes of 10-300 nm that are tuned to match mechanical properties of TiO2-anatase. The modeling results reveal that particles with original diameter of 25-75 nm exhibit three different impact behaviors that could be distinguished as (i) rebounding, (ii) bonding and (iii) fragmentation, depending on their initial impact velocity. In contrast, particles larger than 75 nm do not exhibit the bonding behavior. Detailed stress and strain field distributions reveal that combination of “localized inelastic deformation” along the slip systems and “shear localization” cause bonding of the small and large particles to the substrate. The analyses of associated temperature rise by the inelastic deformation revealed that heat diffusion at these small scales depend on size. Whereas small particles could reach a rather homogeneous temperature distribution, the evolved heat in the larger ones keeps rather localized to areas of highest deformation and may support deformation and the formation of dense layers in aerosol deposition.young investigator research group FOCUS H2 (BMBF EnMat-514-211)
Leavers and Remainers After Brexit: More United Than Divided After All?
Since the British “Brexit referendum” in 2016, tensions between ‘leave’ and ‘remain’ voters have been growing. Using a novel analytical approach based on the full distribution of responses rather than their arithmetic means, Study 1 (N=1,506) showed on average 90% of overlap among Leavers and Remainers across a range of important variables. Even on the variables that are commonly used to illustrate how Leavers and Remainers differ (e.g., prejudice against immigrants), the amount of overlap was larger than 50%. In Study 2 (N=206), we demonstrate the importance of focusing on similarities between groups: Presenting the full distribution of responses from Leavers and Remainers rather than their mean differences caused more accurate perceptions of the actual differences and similarities between both groups and higher perceived intergroup harmony. We conclude that similarities between Leavers and Remainers are substantial and that our proposed approach may help to de-escalate tensions between these and other groups
