Rapporti tra Corte costituzionale italiana e giudice ordinario nella dinamica del rinvio pergiudiziale alla Corte di giustizia
Il lavoro esamina la questione relativa al se la Corte costituzionale italiana, nei casi in cui sia investita di questione incidentale di legittimit\ue0 costituzionale, possa qualificarsi come giurisdizione nazionale abilitata a proporre rinvio pregiudiziale alla Corte di giustizia dell'Unione europea. Preso atto dell'orientamento attuale della Corte costituzionale italiana, volto a negare la propria posizione come giurisdizione abilitata al rinvio pregiudiziale, il lavoro, attraverso un esame della giurisprudenza della Corte di giustizia e degli obblighi nascenti dai Trattati, si propone di dimostrare come risulti conforme al diritto dell'Unione europea ritenere che la Corte costituzionale rientri nella nozione di giurisdizione nazionale abilitata al rinvio pregiudiziale
Action of Ganoderma lucidum mycelial growth filtrates on Erysiphe dffusa and embryotoxicity assessment in a chicken embryo model
This work aimed to evaluate the antimicrobial effect of Ganoderma lucidum mycelial growth filtrates (MGF) on the phytopathogen Erysiphe diffusa and their potential effects on the embryonic development of Gallus gallus. The antimicrobial activity was evaluated on E. diffusa spores by the microdilution broth method. To evaluate embryotoxic and teratogenic effects, fertile eggs of G. gallus received injections of solutions containing the filtrates of G. lucidum through the air chamber. After three days of incubation, we opened the eggs and evaluated egg viability, embryo survival, malformation occurrence, embryonic staging and heart rate. Live embryos were prepared using whole mount technique and the morphological analysis was performed. We used the generalized linear model to fit embryotoxicity and teratogenicity data. We verified that G. lucidum MGF showed inhibitory activity in vitro against E. diffusa and the minimum inhibitory concentrations ranged from 5 to 10 mg/mL. We could also observe that the filtrates did not present embryotoxic or teratogenic effects on the early embryonic development of G. gallus, but induced significant differences in the embryonic mean heart rate and on the stage of embryonic development
Atrial natriuretic peptide effects on intracellular pH changes and ROS production in HEPG2 cells: Role of p38 MAPK and phospholipase D
Aims: The present study was performed to evaluate Atrial Natriuretic Peptide ( ANP) effects on intracellular pH, phospholipase D and ROS production and the possible relationship among them in HepG2 cells. Cancer extracellular microenvironment is more acidic than normal tissues and the activation of NHE- 1, the only system able to regulate pHi homeostasis in this condition, can represent an important event in cell proliferation and malignant transformation. Methods: The ANP effects on pHi were evaluated by fluorescence spectrometry. The effects on p38 MAPK and ROS production were evaluated by immunoblots and analysis of DCF- DA fluorescence, respectively. RT- PCR analysis and Western blotting were used to determine the ANP effect on mRNA NHE- 1 expression and protein levels. PLD- catalyzed conversion of phosphatidylcholine to phosphatydilethanol ( PetOH), in the presence of ethanol, was monitored by thin layer chromatography. Results: A significant pHi decrease was observed in ANP- treated HepG2 cells and this effect was paralleled by the enhancement of PLD activity and ROS production. The ANP effect on pHi was coupled to an increased p38 MAPK phosphorylation and a down- regulation of mRNA NHE- 1 expression and protein levels. Moreover, the relationship between PLD and ROS production was demonstrated by calphostin- c, a potent inhibitor of PLD. At the same time, all assessed ANP- effects were mediated by NPR- C receptors. Conclusion: Our results indicate that ANP recruits a signal pathway associated with p38 MAPK, NHE- 1 and PLD responsible for ROS production, suggesting a possible role for ANP as novel modulator of ROS generation in HepG2 cells. Conjugated polymers mediate intracellular Ca2+ signals in circulating endothelial colony forming cells through the reactive oxygen species-dependent activation of Transient Receptor Potential Vanilloid 1 (TRPV1)
Endothelial colony forming cells (ECFCs) represent the most suitable cellular substrate to induce revascularization of ischemic tissues. Recently, optical excitation of the light-sensitive conjugated polymer, regioregular Poly (3-hexyl-thiophene), rr-P3HT, was found to stimulate ECFC proliferation and tube formation by activating the non-selective cation channel, Transient Receptor Potential Vanilloid 1 (TRPV1). Herein, we adopted a multidisciplinary approach, ranging from intracellular Ca2+ imaging to pharmacological manipulation and genetic suppression of TRPV1 expression, to investigate the effects of photoexcitation on intracellular Ca2+ concentration ([Ca2+](i)) in circulating ECFCs plated on rr-P3HT thin films. Polymer-mediated optical excitation induced a long-lasting increase in [Ca2+](i) that could display an oscillatory pattern at shorter light stimuli. Pharmacological and genetic manipulation revealed that the Ca2+ response to light was triggered by extracellular Ca2+ entry through TRPV1, whose activation required the production of reactive oxygen species at the interface between rr-P3HT and the cell membrane. Light-induced TRPV1-mediated Ca2+ entry was able to evoke intracellular Ca2+ release from the endoplasmic reticulum through inositol-1,4,5-trisphosphate receptors, followed by store-operated Ca2+ entry on the plasma membrane. These data show that TRPV1 may serve as a decoder at the interface between rr-P3HT thin films and ECFCs to translate optical excitation in pro-angiogenic Ca2+ signals
The role of adipose derived stem cells in the treatment of rotator cuff tears: From basic science to clinical application
Over the last decade, regenerative medicine has become increasingly popular throughout the scientific community. The poor healing capacity at the tendon-bone interface makes the rotator cuff an appealing target for biologic agents. Adipose derived stem cells are mesenchymalcells with the capacity for self-renewal and mul-tipotential differentiation. They have been recently proposed, both in isolation and as adjuvants to existing surgical therapies, for the treatment of rotator cuff tears. Several studies have been carried out in this research field, starting from the biological characteristics of adipose derived stem cells, their preparation and culture, up to the application in the experimental field on animal models and on humans. The purpose of this study was to provide a state of the art about the current basic science and clinical literature for the effectiveness of adipose derived stem cells in the treatment of rotator cuff tears
Arthroscopic treatment of an unusual distal clavicle ostheochondroma causing rotator cuff impingement: Case report and literature review
Chronic shoulder impingement is one of the most common causes of shoulder pain. Intrinsic, extrinsic and secondary factors play a role in this syndrome; however the etiology of the pathology is still under debate. In rare cases, it can be caused by tumors, such as an osteochondroma. In the present study, a 49-year-old patient presented with shoulder pain for 6 months. Initially he underwent conservative treatment, without relief of symptoms. X-rays and MRI were then performed and showed the presence of an exostotic formation on the undersurface of the lateral third of the clavicle. The formation was arthroscopically removed. Histologic examination confirmed the diagnosis of osteochondroma. After surgery, the patient resumed fully activities with no symptoms within 3 months. At 1 year follow up, there are still no clinical or radiological signs of recurrence. This is, to our knowledge, the first case where an arthroscopic approach was used to remove an ostochondroma of the distal third of the clavicle
A familial form of epidermolysis bullosa simplex associated with a pathogenic variant in krt5
Epidermolysis bullosa simplex is a disease that belongs to a group of genodermatoses characterised by the formation of superficial bullous lesions caused by minor mechanical trauma to the skin. The skin fragility observed in the EBS is mainly caused by pathogenic variants in the KRT5 and KRT14 genes that compromise the mechanical stability of epithelial cells. By performing DNA sequencing in a female patient with EBS, we found the pathogenic variant c.967G>A (p.Val323Met) in the KRT5 gene. This variant co-segregated with EBS in the family pedigree and was transmitted in an autosomal dominant inheritance manner. This is the first report showing a familial form of EBS due to this pathogenic variant
