Dietary Leucine - An Environmental Modifier of Insulin Resistance Acting on Multiple Levels of Metabolism
Environmental factors, such as the macronutrient composition of the diet, can have a profound impact on risk of diabetes and metabolic syndrome. In the present study we demonstrate how a single, simple dietary factorâleucineâcan modify insulin resistance by acting on multiple tissues and at multiple levels of metabolism. Mice were placed on a normal or high fat diet (HFD). Dietary leucine was doubled by addition to the drinking water. mRNA, protein and complete metabolomic profiles were assessed in the major insulin sensitive tissues and serum, and correlated with changes in glucose homeostasis and insulin signaling. After 8 weeks on HFD, mice developed obesity, fatty liver, inflammatory changes in adipose tissue and insulin resistance at the level of IRS-1 phosphorylation, as well as alterations in metabolomic profile of amino acid metabolites, TCA cycle intermediates, glucose and cholesterol metabolites, and fatty acids in liver, muscle, fat and serum. Doubling dietary leucine reversed many of the metabolite abnormalities and caused a marked improvement in glucose tolerance and insulin signaling without altering food intake or weight gain. Increased dietary leucine was also associated with a decrease in hepatic steatosis and a decrease in inflammation in adipose tissue. These changes occurred despite an increase in insulin-stimulated phosphorylation of p70S6 kinase indicating enhanced activation of mTOR, a phenomenon normally associated with insulin resistance. These data indicate that modest changes in a single environmental/nutrient factor can modify multiple metabolic and signaling pathways and modify HFD induced metabolic syndrome by acting at a systemic level on multiple tissues. These data also suggest that increasing dietary leucine may provide an adjunct in the management of obesity-related insulin resistance
Epilepsia en niños atendidos en el Hospital Nacional Cayetano Heredia de Lima, PerĂș, 2010- 2016.
La epilepsia es la principal causa de consulta neurolĂłgica en niños peruanos. Sin embargo, hay escasa informaciĂłn sobre sus caracterĂsticas clĂnicas y epidemiolĂłgicas. Objetivos: Presentar las caracterĂsticas clĂnicas de los niños epilĂ©pticos atendidos en el Hospital Cayetano Heredia, entre los años 2010 y 2016. Material y mĂ©todos: Se realizĂł un estudio observacional descriptivo, retrospectivo, tipo serie de casos, basado en las historias clĂŹnicas de en niños menores de 14 años con diagnĂłstico de epilepsia, seguimiento clĂnico, registro electroencefalogrĂ fico y neuroimagenes. Resultados: Se incluyeron 193 pacientes. El 47,2% (91/193) presentaron su primera crisis epilĂ©ptica antes del año de edad, cuya etiologĂa fue secundaria en el 59,3% (54/91), destacando los factores perinatales y las malformaciones cerebrales. La etiologĂa primaria fue mĂĄs frecuente en los niños que iniciaron las crisis despuĂ©s del año de edad. Las crisis generalizadas se presentaron en 64,2% (124/193) sin diferencias por grupos etarios. El 69,9% (135/193) fueron controlados con un solo fĂĄrmaco, en tanto que el 15% (29/193) fueron refractarios al tratamiento. La comorbilidad ocurriĂł en el 68,4% (132/193), siendo frecuentes el retraso del desarrollo psicomotor, el retardo mental y la parĂĄlisis cerebral. Conclusiones: La mayorĂa de los niños presentaron su primera crisis antes del año de edad, generalmente de causa secundaria. En los niños mayores fue prevalente la etiologĂa primaria. La mayorĂa fueron controlados con monoterapia, pero se registrĂł un porcentaje de refractariedad al tratamiento. La comorbilidad fue muy frecuente
Antibody Immunity to Zika Virus among Young Children in a Flavivirus-Endemic Area in Nicaragua
Objective: To understand the dynamics of Zika virus (ZIKV)-specific antibody immunity in children born to mothers in a flavivirus-endemic region during and after the emergence of ZIKV in the Americas. Methods: We performed serologic testing for ZIKV cross-reactive and type-specific IgG in two longitudinal cohorts, which enrolled pregnant women and their children (PW1 and PW2) after the beginning of the ZIKV epidemic in Nicaragua. Quarterly samples from children over their first two years of life and maternal blood samples at birth and at the end of the two-year follow-up period were studied. Results: Most mothers in this dengue-endemic area were flavivirus-immune at enrollment. ZIKV-specific IgG (anti-ZIKV EDIII IgG) was detected in 82 of 102 (80.4%) mothers in cohort PW1 and 89 of 134 (66.4%) mothers in cohort PW2, consistent with extensive transmission observed in Nicaragua during 2016. ZIKV-reactive IgG decayed to undetectable levels by 6â9 months in infants, whereas these antibodies were maintained in mothers at the year two time point. Interestingly, a greater contribution to ZIKV immunity by IgG3 was observed in babies born soon after ZIKV transmission. Finally, 43 of 343 (13%) children exhibited persistent or increasing ZIKV-reactive IgG at â„9 months, with 10 of 30 (33%) tested demonstrating serologic evidence of incident dengue infection. Conclusions: These data inform our understanding of protective and pathogenic immunity to potential flavivirus infections in early life in areas where multiple flaviviruses co-circulate, particularly considering the immune interactions between ZIKV and dengue and the future possibility of ZIKV vaccination in women of childbearing potential. This study also shows the benefits of cord blood sampling for serologic surveillance of infectious diseases in resource-limited settings
Arboviruses as an unappreciated cause of non-malarial acute febrile illness in the Dschang Health District of western Cameroon
Acute febrile illness is a common problem managed by clinicians and health systems globally, particularly in the Tropics. In many regions, malaria is a leading and potentially deadly cause of fever; however, myriad alternative etiologies exist. Identifying the cause of fever allows optimal management, but this depends on many factors including thorough knowledge of circulating infections. Arboviruses such as dengue (DENV) cause fever and may be underdiagnosed in sub-Saharan Africa where malaria is a major focus. We examined cases of fever in western Cameroon that tested negative for malaria and found 13.5% (13/96) were due to DENV, with 75% (9/12) of these being DENV serotype 2 infections. Two complete DENV2 genomes were obtained and clustered closely to recent isolates from Senegal and Burkina Faso. The seroprevalence of DENV in this region was 24.8% (96/387). Neutralizing antibodies to DENV2 were detected in all (15/15) seropositive samples tested. Chikungunya (CHIKV) is an arthritogenic alphavirus that is transmitted by Aedes mosquitoes, the same principal vector as DENV. The seroprevalence for CHIKV was 15.7% (67/427); however, CHIKV did not cause a single case of fever in the 96 subjects tested. Of note, being seropositive for one arbovirus was associated with being seropositive for the other (Χ2 = 16.8, p<0.001). Taken together, these data indicate that Aedes-transmitted arboviruses are endemic in western Cameroon and are likely a common but underappreciated cause of febrile illness. This work supports the need for additional study of arboviruses in sub-Saharan Africa and efforts to improve diagnostic capacity, surveillance systems, and arbovirus prevention strategies

60°N was +2.0°C, relative to the 1981â2010 average value (Fig. 5.1). This marks a new high for the record. The average annual surface air temperature (SAT) anomaly for 2016 for land stations north of starting in 1900, and is a significant increase over the previous highest value of +1.2°C, which was observed in 2007, 2011, and 2015. Average global annual temperatures also showed record values in 2015 and 2016. Currently, the Arctic is warming at more than twice the rate of lower latitudes
Antimicrobial resistance among migrants in Europe: a systematic review and meta-analysis
BACKGROUND: Rates of antimicrobial resistance (AMR) are rising globally and there is concern that increased migration is contributing to the burden of antibiotic resistance in Europe. However, the effect of migration on the burden of AMR in Europe has not yet been comprehensively examined. Therefore, we did a systematic review and meta-analysis to identify and synthesise data for AMR carriage or infection in migrants to Europe to examine differences in patterns of AMR across migrant groups and in different settings. METHODS: For this systematic review and meta-analysis, we searched MEDLINE, Embase, PubMed, and Scopus with no language restrictions from Jan 1, 2000, to Jan 18, 2017, for primary data from observational studies reporting antibacterial resistance in common bacterial pathogens among migrants to 21 European Union-15 and European Economic Area countries. To be eligible for inclusion, studies had to report data on carriage or infection with laboratory-confirmed antibiotic-resistant organisms in migrant populations. We extracted data from eligible studies and assessed quality using piloted, standardised forms. We did not examine drug resistance in tuberculosis and excluded articles solely reporting on this parameter. We also excluded articles in which migrant status was determined by ethnicity, country of birth of participants' parents, or was not defined, and articles in which data were not disaggregated by migrant status. Outcomes were carriage of or infection with antibiotic-resistant organisms. We used random-effects models to calculate the pooled prevalence of each outcome. The study protocol is registered with PROSPERO, number CRD42016043681. FINDINGS: We identified 2274 articles, of which 23 observational studies reporting on antibiotic resistance in 2319 migrants were included. The pooled prevalence of any AMR carriage or AMR infection in migrants was 25·4% (95% CI 19·1-31·8; I2 =98%), including meticillin-resistant Staphylococcus aureus (7·8%, 4·8-10·7; I2 =92%) and antibiotic-resistant Gram-negative bacteria (27·2%, 17·6-36·8; I2 =94%). The pooled prevalence of any AMR carriage or infection was higher in refugees and asylum seekers (33·0%, 18·3-47·6; I2 =98%) than in other migrant groups (6·6%, 1·8-11·3; I2 =92%). The pooled prevalence of antibiotic-resistant organisms was slightly higher in high-migrant community settings (33·1%, 11·1-55·1; I2 =96%) than in migrants in hospitals (24·3%, 16·1-32·6; I2 =98%). We did not find evidence of high rates of transmission of AMR from migrant to host populations. INTERPRETATION: Migrants are exposed to conditions favouring the emergence of drug resistance during transit and in host countries in Europe. Increased antibiotic resistance among refugees and asylum seekers and in high-migrant community settings (such as refugee camps and detention facilities) highlights the need for improved living conditions, access to health care, and initiatives to facilitate detection of and appropriate high-quality treatment for antibiotic-resistant infections during transit and in host countries. Protocols for the prevention and control of infection and for antibiotic surveillance need to be integrated in all aspects of health care, which should be accessible for all migrant groups, and should target determinants of AMR before, during, and after migration. JWST-TST DREAMS: Quartz Clouds in the Atmosphere of WASP-17b
Clouds are prevalent in many of the exoplanet atmospheres that have been
observed to date. For transiting exoplanets, we know if clouds are present
because they mute spectral features and cause wavelength-dependent scattering.
While the exact composition of these clouds is largely unknown, this
information is vital to understanding the chemistry and energy budget of
planetary atmospheres. In this work, we observe one transit of the hot Jupiter
WASP-17b with JWST's MIRI LRS and generate a transmission spectrum from 5-12
m. These wavelengths allow us to probe absorption due to the
vibrational modes of various predicted cloud species. Our transmission spectrum
shows additional opacity centered at 8.6 m, and detailed atmospheric
modeling and retrievals identify this feature as SiO(s) (quartz) clouds.
The SiO(s) clouds model is preferred at 3.5-4.2 versus a cloud-free
model and at 2.6 versus a generic aerosol prescription. We find the
SiO(s) clouds are comprised of small m particles,
which extend to high altitudes in the atmosphere. The atmosphere also shows a
depletion of HO, a finding consistent with the formation of
high-temperature aerosols from oxygen-rich species. This work is part of a

will use Guaranteed Time Observations to perform Deep Reconnaissance of

A Critical Tryptophan and Ca2+ in Activation and Catalysis of TPPI, the Enzyme Deficient in Classic Late-Infantile Neuronal Ceroid Lipofuscinosis
Tripeptidyl aminopeptidase I (TPPI) is a crucial lysosomal enzyme that is deficient in the fatal neurodegenerative disorder called classic late-infantile neuronal ceroid lipofuscinosis (LINCL). It is involved in the catabolism of proteins in the lysosomes. Recent X-ray crystallographic studies have provided insights into the structural/functional aspects of TPPI catalysis, and indicated presence of an octahedrally coordinated Ca(2+).Purified precursor and mature TPPI were used to study inhibition by NBS and EDTA using biochemical and immunological approaches. Site-directed mutagenesis with confocal imaging technique identified a critical W residue in TPPI activity, and the processing of precursor into mature enzyme.NBS is a potent inhibitor of the purified TPPI. In mammalian TPPI, W542 is critical for tripeptidyl peptidase activity as well as autocatalysis. Transfection studies have indicated that mutants of the TPPI that harbor residues other than W at position 542 have delayed processing, and are retained in the ER rather than transported to lysosomes. EDTA inhibits the autocatalytic processing of the precursor TPPI.We propose that W542 and Ca(2+) are critical for maintaining the proper tertiary structure of the precursor proprotein as well as the mature TPPI. Additionally, Ca(2+) is necessary for the autocatalytic processing of the precursor protein into the mature TPPI. We have identified NBS as a potent TPPI inhibitor, which led in delineating a critical role for W542 residue. Studies with such compounds will prove valuable in identifying the critical residues in the TPPI catalysis and its structure-function analysis
Adapting Agriculture to Climate Change: A Synopsis of Coordinated National Crop Wild Relative Seed Collecting Programs across Five Continents
The Adapting Agriculture to Climate Change Project set out to improve the diversity,
quantity, and accessibility of germplasm collections of crop wild relatives (CWR). Between 2013 and
2018, partners in 25 countries, heirs to the globetrotting legacy of Nikolai Vavilov, undertook seed
collecting expeditions targeting CWR of 28 crops of global significance for agriculture. Here, we
describe the implementation of the 25 national collecting programs and present the key results. A total
of 4587 unique seed samples from at least 355 CWR taxa were collected, conserved ex situ, safety
duplicated in national and international genebanks, and made available through the Multilateral
System (MLS) of the International Treaty on Plant Genetic Resources for Food and Agriculture (Plant
Treaty). Collections of CWR were made for all 28 targeted crops. Potato and eggplant were the most
collected genepools, although the greatest number of primary genepool collections were made for
rice. Overall, alfalfa, Bambara groundnut, grass pea and wheat were the genepools for which targets
were best achieved. Several of the newly collected samples have already been used in pre-breeding

A communal catalogue reveals Earthâs multiscale microbial diversity
Our growing awareness of the microbial worldâs importance and diversity contrasts starkly with our limited understanding of its fundamental structure. Despite recent advances in DNA sequencing, a lack of standardized protocols and common analytical frameworks impedes comparisons among studies, hindering the development of global inferences about microbial life on Earth. Here we present a meta-analysis of microbial community samples collected by hundreds of researchers for the Earth Microbiome Project. Coordinated protocols and new analytical methods, particularly the use of exact sequences instead of clustered operational taxonomic units, enable bacterial and archaeal ribosomal RNA gene sequences to be followed across multiple studies and allow us to explore patterns of diversity at an unprecedented scale. The result is both a reference database giving global context to DNA sequence data and a framework for incorporating data from future studies, fostering increasingly complete characterization of Earthâs microbial diversity
