CMB-S4 Science Book, First Edition
This book lays out the scientific goals to be addressed by the
next-generation ground-based cosmic microwave background experiment, CMB-S4,
envisioned to consist of dedicated telescopes at the South Pole, the high
Chilean Atacama plateau and possibly a northern hemisphere site, all equipped
with new superconducting cameras. CMB-S4 will dramatically advance cosmological
studies by crossing critical thresholds in the search for the B-mode
polarization signature of primordial gravitational waves, in the determination
of the number and masses of the neutrinos, in the search for evidence of new
light relics, in constraining the nature of dark energy, and in testing general
relativity on large scales
CMB-S4: Forecasting Constraints on Primordial Gravitational Waves
CMB-S4---the next-generation ground-based cosmic microwave background (CMB)
experiment---is set to significantly advance the sensitivity of CMB
measurements and enhance our understanding of the origin and evolution of the
Universe, from the highest energies at the dawn of time through the growth of
structure to the present day. Among the science cases pursued with CMB-S4, the
quest for detecting primordial gravitational waves is a central driver of the
experimental design. This work details the development of a forecasting
framework that includes a power-spectrum-based semi-analytic projection tool,
targeted explicitly towards optimizing constraints on the tensor-to-scalar
ratio, , in the presence of Galactic foregrounds and gravitational lensing
of the CMB. This framework is unique in its direct use of information from the
achieved performance of current Stage 2--3 CMB experiments to robustly forecast
the science reach of upcoming CMB-polarization endeavors. The methodology
allows for rapid iteration over experimental configurations and offers a
flexible way to optimize the design of future experiments given a desired
scientific goal. To form a closed-loop process, we couple this semi-analytic
tool with map-based validation studies, which allow for the injection of
additional complexity and verification of our forecasts with several
independent analysis methods. We document multiple rounds of forecasts for
CMB-S4 using this process and the resulting establishment of the current
reference design of the primordial gravitational-wave component of the Stage-4
experiment, optimized to achieve our science goals of detecting primordial
gravitational waves for at greater than , or, in the

Design and baseline characteristics of the finerenone in reducing cardiovascular mortality and morbidity in diabetic kidney disease trial
Background: Among people with diabetes, those with kidney disease have exceptionally high rates of cardiovascular (CV) morbidity and mortality and progression of their underlying kidney disease. Finerenone is a novel, nonsteroidal, selective mineralocorticoid receptor antagonist that has shown to reduce albuminuria in type 2 diabetes (T2D) patients with chronic kidney disease (CKD) while revealing only a low risk of hyperkalemia. However, the effect of finerenone on CV and renal outcomes has not yet been investigated in long-term trials.
Patients and Methods: The Finerenone in Reducing CV Mortality and Morbidity in Diabetic Kidney Disease (FIGARO-DKD) trial aims to assess the efficacy and safety of finerenone compared to placebo at reducing clinically important CV and renal outcomes in T2D patients with CKD. FIGARO-DKD is a randomized, double-blind, placebo-controlled, parallel-group, event-driven trial running in 47 countries with an expected duration of approximately 6 years. FIGARO-DKD randomized 7,437 patients with an estimated glomerular filtration rate >= 25 mL/min/1.73 m(2) and albuminuria (urinary albumin-to-creatinine ratio >= 30 to <= 5,000 mg/g). The study has at least 90% power to detect a 20% reduction in the risk of the primary outcome (overall two-sided significance level alpha = 0.05), the composite of time to first occurrence of CV death, nonfatal myocardial infarction, nonfatal stroke, or hospitalization for heart failure.
Conclusions: FIGARO-DKD will determine whether an optimally treated cohort of T2D patients with CKD at high risk of CV and renal events will experience cardiorenal benefits with the addition of finerenone to their treatment regimen.

Humins in the environment: early stage insights on ecotoxicological aspects
International audienceWith the growing interest of a circular economy, the use of lignocellulosic residues such as lignins and humins as potential renewable feedstock for biorefining processes looks more and more promising. With humins, many challenges are still remaining for their sustainable use, starting from the need of providing reference data reflecting actual usable feedstocks of such materials. With this perspective, this paper offers a first outlook on the potential environmental fate of those materials and components, all related to furanics, a family of compounds for which toxicity is still a matter of debate. During the assessment, the conventional OECD ecotoxicity and biodegradability tests demanded by the European REACH regulation for a primary evaluation of environmental hazards were performed in combination with fish immunomarker tests to study the possible long-term effects on aquatic ecosystems. These first results are promising as humins did not exhibit any immediate ecotoxicological concerns and hence would allow considering their use in environmental-friendly applications
Insights on Thermal and Fire Hazards of Humins in Support of Their Sustainable Use in Advanced Biorefineries
International audienceThe concept of circular economy is aiming at promoting innovative use of wastes, residues, or byproducts into marketable chemicals and intermediates, while enhancing process economics and environmental sustainability. Within biorefineries, the focus has now shifted toward âhuminsâ, polymeric condensation byproducts far less investigated than lignins. Humins are produced in many biobased value chains that start from polysaccharides pretreatment, targeting access to C6 and C5 sugars and further conversion to furanics- or levulinics-based platform chemicals. They are complex and versatile substrates, and combustible because of their organic nature. A first insight on safety issues of humins is therefore proposed in this paper, considering the growing interest in humins as new biobased feedstocks for various applications. The current study provides characterization details necessary to establish safety information on a global context. This first attempt has brought in preliminary safety data mainly focusing on thermal and fire hazards of humins, to ensure safe storage, handling, transportation, and processing. The obtained results show that overall fire risk of humins so far is similar to that of the conventional cellulosic fuels such as woody materials. Further consolidation of safety information shall be done with a scenario-based risk assessment approach for conditioning safe use of humins in further innovative applications
