The major transcriptional regulatory protein of herpes simplex virus type 1 includes a protease resistant DNA binding domain
Herpes simplex virus type 1 expresses five immediateearly (IE) polypeptides. In the absence of functional Vmw175 (the product of IE gene 3) activation of transcription of later classes of viral genes and repression of IE gene expression does not occur. The recognition of specific DNA sequences by Vmw175 requires, as determined by sensitivity to mutation, a part of the protein highly conserved in the corresponding proteins of related herpes viruses. However, mutations in other parts of the protein can also disrupt specific DNA binding. This paper shows that the DNA binding domain of Vmw175 can be liberated as a functional unit by digestion with proteinase K. Analysis of mutant Vmw175 proteinsshowed that the proteinase K resistant domain has an amino terminus between amino acid residues 229 and 292, while its carboxy terminus is between residues 495 and 518. Mutations outside this region which affect DNA binding by the intact protein do not eliminate binding of the proteinase K resistant domain. This implies that direct DNA binding by Vmw175 involves a linear subsection of the polypeptide, and that mutations in other parts of the polypeptide which affect DNA binding of the whole protein do so by indirect means
Core test reactor shield cooling system analysis
System requirements for cooling the shield within the vacuum vessel for the core test reactor are analyzed. The total heat to be removed by the coolant system is < 22,700 Btu/hr, with an additional 4600 Btu/hr to be removed by the 2-inch thick steel plate below the shield. This latter heat is radiated to the walls of the vacuum chamber where the heat is removed by the cooling coils wrapped around the outside of the chamber. The maximum temperature of the concrete in the shield can be kept below 200°F if the shield plug walls are kept below 160°F. The walls of the two "donut" shaped shield segments, which are cooled by the water from the shield and vessel cooling system, should operate below 95°F. The walls of the center plug, which are cooled with nitrogen, should operate below 100°F
High level expression and purification of herpes simplex virus type 1 immediate early polypeptide Vmw110
Herpes simplex virus type 1 (HSV-1) encodes five immediate early (IE) polypeptides. This paper reports the construction of a baculovirus vector which expresses large amounts of Vmw110, the product of IE gene 1. The expressed protein has been purified to near homogeneity and has a mobility on SDS polyacrylamide gels identical to that of Vmw110 produced during HSV-1 infection. Characterisation of its properties indicated that it forms dimers and perhaps higher order oligomers in solution and that the purified protein binds to both single stranded and double stranded calf thymus DNA cellulose columns. However, filter binding experiments were unable to detect any stable association of Vmw110 with DNA in solution

If the polaron coupling constant is large enough, bipolarons or
multi-polarons will form. When passing through the critical from

Clouds, shadows, or twilight? Mayfly nymphs recognise the difference
1. We examined the relative changes in light intensity that initiate night-time locomotor activity changes in nymphs of the mayfly, Stenonema modestum (Heptageniidae). Tests were carried out in a laboratory stream to examine the hypothesis that nymphs increase their locomotion in response to the large and sustained reductions in relative light intensity that take place during twilight but not to short-term daytime light fluctuations or a minimum light intensity threshold. Ambient light intensity was reduced over a range of values representative of evening twilight. Light was reduced over the same range of intensities either continuously or in discrete intervals while at the same time nymph activity on unglazed tile substrata was video recorded.
2. Nymphs increased their locomotor activity during darkness in response to large, sustained relative light decreases, but not in response to short-term, interrupted periods of light decrease. Nymphs did not recognise darkness unless an adequate light stimulus, such as large and sustained relative decrease in light intensity, had taken place.
3. We show that nymphs perceive light change over time and respond only after a lengthy period of accumulation of light stimulus. The response is much lengthier than reported for other aquatic organisms and is highly adaptive to heterogeneous stream environments
Determining the Drift Time of Charge Carriers in P-Type Point-Contact HPGe Detectors
An algorithm to measure the drift time of charge carriers in p-type point
contact (PPC) high-purity germanium (HPGe) detectors from the signals processed
with a charge-sensitive preamplifier is introduced. It is demonstrated that the
drift times can be used to estimate the distance of charge depositions from the
point contact and to characterize losses due to charge trapping. A correction
for charge trapping effects over a wide range of energies is implemented using
the measured drift times and is shown to improve the energy resolution by up to

Possibility of spontaneous CP violation in the nonminimal supersymmetric standard model with two neutral Higgs singlets
A supersymmetric standard model with two Higgs doublets and two Higgs
singlets is investigated if it can accommodate the possibility of spontaneous
CP violation. Assuming the degeneracy of the scalar quark masses of the third
generation, we find that spontaneous CP violation in the Higgs sector is viable
in our model. In the case of spontaneous CP violation, the masses of the
lightest two neutral Higgs bosons are estimated to be 80 and 125 GeV for some
parameter values in our model, which, are consistent with LEP2 data.Comment: 18 pages, 3figure
