    Transciptome Analysis Illuminates the Nature of the Intracellular Interaction in a Vertebrate-Algal Symbiosis

    During embryonic development, cells of the green alga Oophila amblystomatis enter cells of the salamander Ambystoma maculatum forming an endosymbiosis. Here, using de novo dual-RNA seq, we compared the host salamander cells that harbored intracellular algae to those without algae and the algae inside the animal cells to those in the egg capsule. This two-by-two-way analysis revealed that intracellular algae exhibit hallmarks of cellular stress and undergo a striking metabolic shift from oxidative metabolism to fermentation. Culturing experiments with the alga showed that host glutamine may be utilized by the algal endosymbiont as a primary nitrogen source. Transcriptional changes in salamander cells suggest an innate immune response to the alga, with potential attenuation of NF-κB, and metabolic alterations indicative of modulation of insulin sensitivity. In stark contrast to its algal endosymbiont, the salamander cells did not exhibit major stress responses, suggesting that the host cell experience is neutral or beneficial

    Zeeman Slowers for Strontium based on Permanent Magnets

    We present the design, construction, and characterisation of longitudinal- and transverse-field Zeeman slowers, based on arrays of permanent magnets, for slowing thermal beams of atomic Sr. The slowers are optimised for operation with deceleration related to the local laser intensity (by the parameter ϵ\epsilon), which uses more effectively the available laser power, in contrast to the usual constant deceleration mode. Slowing efficiencies of up to \approx 1818 % are realised and compared to those predicted by modelling. We highlight the transverse-field slower, which is compact, highly tunable, light-weight, and requires no electrical power, as a simple solution to slowing Sr, well-suited to spaceborne application. For 88^{88}Sr we achieve a slow-atom flux of around 6×1096\times 10^9 atoms\,s1^{-1} at 3030 ms1^{-1}, loading approximately 5×1085\times 10^8 atoms in to a magneto-optical-trap (MOT), and capture all isotopes in approximate relative natural abundances

    Co-Cultures of Oophila Amblystomatis Between Ambystoma Maculatum and Ambystoma Gracile Hosts Show Host-Symbiont Fidelity

    A unique symbiosis occurs between embryos of the spotted salamander (Ambystoma maculatum) and a green alga (Oophila amblystomatis). Unlike most vertebrate host-symbiont relationships, which are ectosymbiotic, A. maculatum exhibits both an ecto- and an endo-symbiosis, where some of the green algal cells living inside egg capsules enter embryonic tissues as well as individual salamander cells. Past research has consistently categorized this symbiosis as a mutualism, making this the first example of a “beneficial” microbe entering vertebrate cells. Another closely related species of salamander, Ambystoma gracile, also harbors beneficial Oophila algae in its egg capsules. However, our sampling within the A. gracile range consistently shows this to be a strict ectosymbiotic interaction—with no sign of tissue or presumably cellular entry. In this study we swapped cultured algae derived from intracapsular fluid of different salamander hosts to test the fidelity of tissue entry in these symbioses. Both A. maculatum and A. gracile embryos were raised in cultures with their own algae or algae cultured from the other host. Under these in vitro culture conditions A. maculatum algae will enter embryonic A. maculatum tissues. Additionally, although at a much lower frequency, A. gracile derived algae will also enter A. maculatum host tissues. However, neither Oophila strain enters A. gracile hosts in these co-culture conditions. These data reveal a potential host-symbiont fidelity that allows the unique endosymbiosis to occur in A. maculatum, but not in A. gracile. However, preliminary trials in our study found that persistent endogenous A. maculatum algae, as opposed to the cultured algae used in subsequent trials, enters host tissues at a higher frequency. An analysis of previously published Oophila transcriptomes revealed dramatic differences in gene expression between cultured and intracapsular Oophila. These include a suite of genes in protein and cell wall synthesis, photosynthesis, central carbon metabolism suggesting the intracapsular algae are assimilating ammonia for nitrogen metabolism and may be undergoing a life-cycle transition. Further refinements of these co-culture conditions could help determine physiological differences between cultured and endogenous algae, as well as rate-limiting cues provided for the alga by the salamander

    Child Abuse & Neglect: The inclusion of disability as a condition for termination of parental rights

    Objectives: All 50 states and the District of Columbia have statutes outlining the grounds for terminating parental rights (TPR) in relation to child abuse and neglect. Although recent research has found that parents with disabilities are not more likely to maltreat their children than parents without disabilities (Glaun & Brown, 1999; Oyserman, Mowbray, Meares, & Firminger, 2000), studies have found very high rates of TPR of parents with disabilities (Accardo & Whitman, 1989). The objective of this study is to examine how states are including disability in their TPR statutes. Methods: This study used legal document analysis, consisting of a comprehensive Boolean search of the state codes of the 50 states and District of Columbia (DC) relating to TPR, using the most recent state code available on Lexis-Nexis in August 2005. TPR and related statutes were searched for contemporary and historical disability related terms and their common cognates, such as: “mental,” “disability,” “handicap,” and “incapacity.” Two researchers independently conducted the searches, and the searches were reconciled. A code list was then developed to measure for inclusion of disability, preciseness, scope, use of language, and references to accessibility or fairness. Statutes were then reanalyzed, and groupings developed. Results: Thirty-seven states included disability-related grounds for termination of parental rights, while 14 states did not include disability language as grounds for termination. Many of these state codes used outdated terminology, imprecise definitions, and emphasized disability status rather than behavior. All of the 14 states that do not include disability in TPR grounds allowed for termination based on neglectful parental behavior that may be influenced by a disability. Conclusions: The use of disability language in TPR statutes can put an undue focus on the condition of having a disability, rather than parenting behavior. Implications: This paper recommends that states consider removing disability language from their statutes, as such language risks taking the emphasis away from the assessment based on parenting behavior

    Prevalence of children with disabilities in the child welfare system and out of home placement: An examination of administrative records

    This article explores the prevalence and characteristics of children with disabilities within the child welfare system using administrative data from the State of Minnesota. This study finds that more than a fifth (22%) of children with substantiated maltreatment are labeled in administrative records as having a disability, and more than one quarter of children (27.9%) over age five. The most common type of disability among children with substantiated maltreatment was emotional disturbance, while other common disabilities included intellectual and developmental disabilities and learning disabilities. Using logistic regression, this study finds that children with substantiated maltreatment with disabilities were about two times more likely to be in out of home placement than children with substantiated maltreatment without disabilities

    Gender differences in child care and work: An interdisciplinary perspective

    During the past several decades, women have increased their labor-force participation while remaining primarily responsible for the care of children. Men have not correspondingly increased their home activity, and women's earnings have not reached parity with men's. A primary reason that women have failed to gain equality in the labor market may be the remaining inequalities in provision of child rearing. We consider the constraints on child rearing faced by men and review literature on animal behavior to elucidate the conditions that facilitate parental investment by males. Some factors appear to have congruent effects, in general, on paternal behavior in many species: the benefit to young of male help, the male capacity for providing help, and paternity certainty.     Gender, Sexual Orientation, and Truth-of-Consensus in Studies of Physical Attractiveness

    Truth-of-consensus methodology presently holds that sex differences in perceptions of physical attractiveness are negligible and may be routinely ignored during prescaling. No determination has been made in the literature of the effects of sexual orientation on this perceptual process. The data presented herein suggest that while sex and sexual orientation of judge are largely irrelevant to prescaling of female stimuli, these variables are important when judging male stimuli. In particular, male homosexuals and male heterosexuals differ significantly in ranking male facial photographs. Thus, experimenters wishing to treat attractiveness levels as known quantities should control for this difference, especially when using a small number of judges for prescaling

    The menstrual cycle and components of human female sexual behaviour

    In non-human primates, gonadal hormones alter attractivity and proceptivity to produce maximal sexual interaction around ovulation. A review of the relevant literature for humans revealed little comparable data. Hormonal correlations with human sexual behaviour have been difficult to demonstrate, and the most prominent peak of conscious sexual desire appears to occur in the mid-follicular rather than the ovulatory phase. There is evidence, however, for a mid-cycle peak in female sexual initiation.     Young-adult children of alcoholic parents: protective effects of positive family functioning

    The occurrence of alcoholism is clustered within families, but the detrimental effect of a positive family history may vary with the degree of family impairment involved. In this study we assessed the effects of family history and family environment on alcohol misuse. From ongoing studies we recruited parents who had a child aged 18–30, 20 with a DSM-III-R alcohol dependence diagnosis, 20 without. The child then completed a multidimensional assessment. The young-adult participants included 20 men and 20 women (mean age=24.8). Differences by family history were restricted to substance abuse behaviors. While a high level of alcohol problems occurred in both groups, those with an alcohol-dependent parent were more likely to be heavy drinkers and showed more symptoms of alcohol dependence. Overall psychological adjustment did not differ between the groups, however. Alcohol misuse measures did correlate moderately with symptoms of poor emotional health. The most important correlates of alcohol misuse measures in this study were exposure to parental alcoholism, abusive punishment, and psychological symptoms, with some separation of effects in the two subgroups. Psychological symptoms had a stronger relationship with misuse in subjects with social-drinking parents, while abuse was more associated in the group with an alcohol-dependent parent. These results confirm the importance of environmental interactions with familial risk. A biological vulnerability from an alcohol-dependent parent was not sufficient or necessary for the participants in this study to develop alcohol dependence as a young adult, although there was an increased risk.
