    Effect of luminescence transport through adipose tissue on measurement of tissue temperature by using ZnCdS nanothermometers

    The spectra of luminescence of ZnCdS nanoparticles (ZnCdS NPs) were measured and analyzed in a wide temperature range: from room to human body and further to a hyperthermic temperature resulting in tissue morphology change. The results show that the signal of luminescence of ZnCdS NPs placed within the tissue is reasonably good sensitive to temperature change and accompanied by phase transitions of lipid structures of adipose tissue. It is shown that the presence of a phase transition in adipose tissue upon its heating (polymorphic transformations of lipids) leads to a nonmonotonic temperature dependence of the intensity of luminescence for the nanoparticles introduced into adipose tissue. This is due to a change in the light scattering by the tissue. The light scattering of adipose tissue greatly distorts the results of temperature measurements. The application of these nanoparticles is possible for temperature measurements in very thin or weakly scattering samples

    The relevance of the stated problem of the crisis of 6-7-year old’s is dictated by the recently growing tendency to bring back children to preschool institutions after a short period at school by parents who notice the child’s painful condition associated with school maladaptation. In this regard, many researchers and practicing educators recommend reviewing the start dates for children at school so that the child starts school at the age of seven. Other teachers notice a significant increase in the work load on primary schoolchildren connected to the introduction of the Federal State Educational Standard for Primary General Education and the corresponding forms of control and assessment activities. Thus, there was a need to develop new technologies to preserve the health of younger students and allow them to overcome the crisis of the onset of education. The purpose of this article is to describe the developed technology aimed at helping 6-7-year old to overcome the age crisis and prevent school maladaptation with the help of early formation of stress tolerance. The main method in the study is the design of a technology for early formation of stress tolerance and prediction of the results of its implementation in the experimental groups of 6-7-year-old children. The technology is based on the principles of continuity of the educational process between preschool educational organizations and schools. With the implementation of the developed technology the following results were achieved: increased motivation for success and higher somatic health level manifested in sport and creative activities. The above-mentioned results in their turn contributed to crisis prevention, school deprivation of first graders and comfortable overcoming of the crisis of 6-7 year olds. The materials of the article can be useful to organizers of sport and creative activities in preschool educational institutions, educators of preparatory groups, teachers, methodologists in preschool and primary education, as well as physical education teachers and primary school teachers. The materials of the article can be used in the system of advanced training of teachers of physical education and elementary school, as well as in the preparation for professional activities in preschool educational institutions and elementary schools of students of teacher training colleges and universities.La relevancia del problema declarado de la crisis de los niños de 6 a 7 años está dictada por la tendencia cada vez mayor de traer a los niños a las instituciones preescolares después de un corto período en la escuela por parte de los padres que notan la condición dolorosa del niño asociada con la mala adaptación escolar. En este sentido, muchos investigadores y educadores en ejercicio recomiendan revisar las fechas de inicio de los niños en la escuela para que el niño comience la escuela a la edad de siete años. Otros maestros notan un aumento significativo en la carga de trabajo de los escolares de primaria relacionados con la introducción del Estándar Educativo del Estado Federal para la Educación General Primaria y las formas correspondientes de actividades de control y evaluación. Por lo tanto, era necesario desarrollar nuevas tecnologías para preservar la salud de los estudiantes más jóvenes y permitirles superar la crisis del inicio de la educación. El propósito del artículo es describir la tecnología desarrollada destinada a ayudar a niños de 6 a 7 años a superar la crisis de la edad y prevenir la mala adaptación escolar con la ayuda de la formación temprana de tolerancia al estrés. El método principal en el estudio es el diseño de una tecnología para la formación temprana de tolerancia al estrés y la predicción de los resultados de su implementación en los grupos experimentales de niños de 6-7 años. La tecnología se basa en los principios de continuidad del proceso educativo entre las organizaciones educativas preescolares y las escuelas. Con la implementación de la tecnología desarrollada se lograron los siguientes resultados: mayor motivación para el éxito y un mayor nivel de salud somática manifestado en actividades deportivas y creativas. Los resultados mencionados a su vez contribuyeron a la prevención de crisis, la privación escolar de los alumnos de primer grado y la superación cómoda de la crisis de los niños de 6 a 7 años. Los materiales del artículo pueden ser útiles para los organizadores de actividades deportivas y creativas en instituciones educativas preescolares, educadores de grupos preparatorios, maestros, metodólogos en educación preescolar y primaria, así como maestros de educación física y maestros de primaria.     Modeling Energy-Efficient Consumption at Industrial Enterprises

    A possibility to reduce costs incurred in both product manufacturing and energy consumption constitutes energy-efficient consumption at industrial enterprises. Manufacturers operating in various economic sectors adopt similar conceptual models of energy consumption, which allows developing a universal solution for modeling such processes. The central problem of modeling lies in finding an adequate objective function based on a sufficient set of parameters and characteristics of the efficiency of an enterprise's power circuit. The article justifies the performance indicators of industrial enterprises' power supply systems. The literature review proves that economic feasibility of energy efficiency is especially obvious if a massive modernization program is implemented. The estimates produced indicate that the largest portion of the potential energy savings is attributed to implementation of energy-saving projects in industrial buildings and other structures. We present a mathematical model designed to perform a comparative analysis of technical and economic features of two modernization scenarios of the heat and power supply system of an industrial enterprise circuit. Performing a simulation calculation based on performance aspects of Russian companies, the paper outlines the advantages of integrated modernization and analyzes the influence of various factors. The results reveal that there is an inverse correlation between specific heat consumption of buildings under thermal modernization and the length of the thermal network. As specific energy consumption in the thermal modernization scenario increases, the payback period for capital investments decreases. The effect of this dynamics is virtually proportional. Compared with total costs for various heat sources, the share of capital costs differs significantly; however, the share of running costs is dominant for all types of units. At the same time, the length of an enterprise's heat circuit weakly affects the payback period of capital costs.     Temperature sensing of adipose tissue heating with the luminescent upconversion nanoparticles as nanothermometer: In vitro study

    The luminescence spectra of upconversion nanoparticles (UCNPs) imbedded in fat tissue were measured in a wide temperature range, from room to human body and further to hyperthermic temperatures. The two types of synthesized UCNP [NaYF4:Yb3+, Er3+] specimens, namely, powdered as-is and embedded into polymer film, were used. The results show that the luminescence of UCNPs placed under the adipose tissue layer is reasonably good sensitive to temperature change and reflects phase transitions of lipids in tissue cells. The most likely, multiple phase transitions are associated with the different components of fat cells such as phospholipids of cell membrane and lipids of fat droplets. In the course of fat cell heating, lipids of fat droplet first transit from a crystalline form to a liquid crystal form and then to a liquid form, which is characterized by much less scattering. The phase transitions of lipids were observed as the changes of the slope of the temperature dependence of UCNP luminescence intensity. The obtained results confirm a high sensitivity of the luminescent UCNPs to the temperature variations within tissues and show a strong potential for providing a controllable tissue thermolysis

    Overexpression of the SuUR gene induces reversible modifications at pericentric, telomeric and intercalary heterochromatin of Drosophila melanogaster polytene chromosomes

    The SuUR (suppressor of underreplication) gene controls late replication and underreplication of DNA in Drosophila melanogaster polytene chromosomes: its mutation suppresses DNA underreplication whereas additional doses of the normal allele strongly enhances underreplication. The SuUR protein is localized in late replicating and underreplicating regions. The N-terminal part of the SuUR protein shares modest similarity with the ATPase/helicase domain of SWI2/SNF2 chromatin remodeling factors, suggesting a role in modification of chromatin structure. Here we describe novel structural modifications of polytene chromosomes (swellings) and show that SuUR controls chromatin organization in polytene chromosomes. The swellings develop as the result of SuUR ectopic expression in the transgene system Sgs3-GAL4; UAS-SuUR(+). They are reminiscent of chromosome puffs and appear in similar to190 regions of intercalary, pericentric and telomeric heterochromatin; some of them attain tremendous size. The swellings are temperature sensitive: they are maximal at 29C and are barely visible at 18degreesC. Shifting from 29degreesC to 18degreesC results in the complete recovery of the normal structure of chromosomes. The swellings are transcriptionally inactive, since they do not incorporate [H-3]uridine. The SuUR protein is not visualized in regions of maximally developed swellings. Regular ecdysone-inducible puffs are not induced in cells where these swellings are apparent

    Delivery and reveal of localization of upconversion luminescent microparticles and quantum dots in the skin in vivo by fractional laser microablation, multimodal imaging, and optical clearing

    Delivery and spatial localization of upconversion luminescent microparticles [Y 2 O 3 ;Yb, Er] (mean size ~1.6 μm) and quantum dots (QDs) (CuInS 2 ZnS nanoparticles coated with polyethylene glycol-based amphiphilic polymer, mean size ~20 nm) inside rat skin was studied in vivo using a multimodal optical imaging approach. The particles were embedded into the skin dermis to the depth from 300 to 500 μm through microchannels performed by fractional laser microablation. Low-frequency ultrasound was applied to enhance penetration of the particles into the skin. Visualization of the particles was revealed using a combination of luminescent spectroscopy, optical coherence tomography, confocal microscopy, and histochemical analysis. Optical clearing was used to enhance the image contrast of the luminescent signal from the particles. It was demonstrated that the penetration depth of particles depends on their size, resulting in a different detection time interval (days) of the luminescent signal from microparticles and QDs inside the rat skin in vivo. We show that luminescent signal from the upconversion microparticles and QDs was detected after the particle delivery into the rat skin in vivo during eighth and fourth days, respectively. We hypothesize that the upconversion microparticles have created a long-time depot localized in the laser-created channels, as the QDs spread over the surrounding tissues

    Seasonal hydrological and hydrochemical surveys in the Voevoda Bay (Amur Bay, Japan Sea)

    Hydrological and hydrochemical surveys were conducted in the Voevoda Bay in May, August, and October, 2011 and February, 2012, in total 140 stations. Free water exchange of the bay with the Amur Bay is observed, with exception of its inner bights Kruglaya and Melkovodnaya. The water exchange is maintained by anticyclonic circulation with the inflow along the southern coast and outflow along the northern coast of the Voyevoda Bay. However, the opposite cyclonic circulation is observed in the Melkovodanaya Bight because of its coastal line patterns and fresh water discharge by the river. Dissolved oxygen content and partial pressure of CO2 in the bay waters are determined mostly by intensive processes of production and destruction of organic matter. There are three main groups of primary producers there, as diatom algae, sea grass Zostera marina , and periphyton. Specific chemical regime is formed in the Melkovodnaya Bight, in particular in winter when primary production depends on the ice cover and is driven by variations of photosynthetically active radiation passed through the ice. Seasonal variability of production-destruction processes intensity is discussed on the data of chemical parameters changes

    Whole genome screening procures a holistic hold of the Russian chicken gene pool heritage and demographic history

    Simple Summary: A collection of native farm animal breeds can be considered as a gene pool and a national heritage. Long-term artificial selection in domesticated animals has certain effects on their genomes, which can be investigated using genome-wide screens for DNA sequence variation, that is, so-called single nucleotide polymorphism (SNP) screens. Here, we looked at the genomes of 19 Russian chicken gene pool breeds, both native and imported, evaluating the contrasting egg, meat and dual-purpose types. Based on genetic diversity statistics, we identified differences between the breeds using many DNA markers (SNPs) that may represent genomic regions that are being selected for, either within a specific breed or shared between breeds. Our research will be helpful for further understanding the genomic diversity and demographic history of Russian domestic chickens. This would be essential for their successful breeding. Abstract: A study for genomic variation that may reflect putative selective signaling and be associated with economically important traits is instrumental for obtaining information about demographic and selection history in domestic animal species and populations. A rich variety of the Russian chicken gene pool breeds warrants a further detailed study. Specifically, their genomic features can derive implications from their genome architecture and selective footprints for their subsequent breeding and practical efficient exploitation. In the present work, whole genome genotyping of 19 chicken breeds (20 populations with up to 71 samples each) was performed using the Chicken 50 K BeadChip DNA chip. The studied breed sample included six native Russian breeds of chickens developed in the 17th–19th centuries, as well as eight Russian chicken breeds, including the Russian White (RW), created in the 20th century on the basis of improving local chickens using breeds of foreign selection. Five specialized foreign breeds of chickens, including the White Leghorn (WL), were used along with other breeds representing the Russian gene pool. The characteristics of the genetic diversity and phylogenetic relationships of the native breeds of chickens were represented in comparison with foreign breeds. It was established that the studied native breeds demonstrate their own genetic structure that distinguishes them from foreign breeds, and from each other. For example, we previously made an assumption on what could cause the differences between two RW populations, RW1 and RW2. From the data obtained here, it was verified that WL was additionally crossed to RW2, unlike RW1. Thus, inherently, RW1 is a purer population of this improved Russian breed. A significant contribution of the gene pool of native breeds to the global genetic diversity of chickens was shown. In general, based on the results of a multilateral survey of this sample of breeds, it can be concluded that phylogenetic relationships based on their genetic structure and variability robustly reflect the known, previously postulated and newly discovered patterns of evolution of native chickens. The results herein presented will aid selection and breeding work using this gene pool

Domestic animals are subject to long-term artificial selection that leaves certain footprints in their genomes one can explore using genome-wide SNP screen. We examined here genomes of two contrasting chicken breeds, the native egg-type Russian White and meat-type White Cornish. Using three statistics, we identified genomic regions under putative selection, both breed-specific and shared between two breeds, that harbor key candidate genes for economically important traits. Our findings will be useful in further understanding selection history and genomic diversity in domestic chickens that would be pivotal in their productive breeding. Abstract: Comparison of genomic footprints in chicken breeds with different selection history is a powerful tool in elucidating genomic regions that have been targeted by recent and more ancient selection. In the present work, we aimed at examining and comparing the trajectories of artificial selection in the genomes of the native egg-type Russian White (RW) and meat-type White Cornish (WC) breeds. Combining three different statistics (top 0.1% SNP by FST value at pairwise breed comparison, hapFLK analysis, and identification of ROH island shared by more than 50% of individuals), we detected 45 genomic regions under putative selection including 11 selective sweep regions, which were detected by at least two different methods. Four of such regions were breed-specific for each of RW breed (on GGA1, GGA5, GGA8, and GGA9) and WC breed (on GGA1, GGA5, GGA8, and GGA28), while three remaining regions on GGA2 (two sweeps) and GGA3 were common for both breeds. Most of identified genomic regions overlapped with known QTLs and/or candidate genes including those for body temperatures, egg productivity, and feed intake in RW chickens and those for growth, meat and carcass traits, and feed efficiency in WC chickens. These findings were concordant with the breed origin and history of their artificial selection. We determined a set of 188 prioritized candidate genes retrieved from the 11 overlapped regions of putative selection and reviewed their functions relative to phenotypic traits of interest in the two breeds. One of the RW-specific sweep regions harbored the known domestication gene, TSHR. Gene ontology and functional annotation analysis provided additional insight into a functional coherence of genes in the sweep regions. We also showed a greater candidate gene richness on microchromosomes relative to macrochromosomes in these genomic areas. Our results on the selection history of RW and WC chickens and their key candidate genes under selection serve as a profound information for further conservation of their genomic diversity and efficient breeding

    Selective footprints and genes relevant to cold adaptation and other phenotypic traits are unscrambled in the genomes of divergently selected chicken breeds

    Background: The genomes of worldwide poultry breeds divergently selected for performance and other phenotypic traits may also be affected by, and formed due to, past and current admixture events. Adaptation to diverse environments, including acclimation to harsh climatic conditions, has also left selection footprints in breed genomes. Results: Using the Chicken 50K_CobbCons SNP chip, we genotyped four divergently selected breeds: two aboriginal, cold tolerant Ushanka and Orloff Mille Fleur, one egg-type Russian White subjected to artificial selection for cold tolerance, and one meat-type White Cornish. Signals of selective sweeps were determined in the studied breeds using three methods: (1) assessment of runs of homozygosity islands, (2) FST based population differential analysis, and (3) haplotype differentiation analysis. Genomic regions of true selection signatures were identified by two or more methods or in two or more breeds. In these regions, we detected 540 prioritized candidate genes supplemented them with those that occurred in one breed using one statistic and were suggested in other studies. Amongst them, SOX5, ME3, ZNF536, WWP1, RIPK2, OSGIN2, DECR1, TPO, PPARGC1A, BDNF, MSTN, and beta-keratin genes can be especially mentioned as candidates for cold adaptation. Epigenetic factors may be involved in regulating some of these important genes (e.g., TPO and BDNF). Conclusion: Based on a genome-wide scan, our findings can help dissect the genetic architecture underlying various phenotypic traits in chicken breeds. These include genes representing the sine qua non for adaptation to harsh environments. Cold tolerance in acclimated chicken breeds may be developed following one of few specific gene expression mechanisms or more than one overlapping response known in cold-exposed individuals, and this warrants further investigation
