Chromosome 9p deletion in clear cell renal cell carcinoma predicts recurrence and survival following surgery
BACKGROUND: Wider clinical applications of 9p status in clear cell renal cell carcinoma (ccRCC) are limited owing to the lack of validation and consensus for interphase fluorescent in situ hybridisation (I-FISH) scoring technique. The aim of this study was to analytically validate the applicability of I-FISH in assessing 9p deletion in ccRCC and to clinically assess its long-term prognostic impact following surgical excision of ccRCC. METHODS: Tissue microarrays were constructed from 108 renal cell carcinoma (RCC) tumour paraffin blocks. Interphase fluorescent in situ hybridisation analysis was undertaken based on preset criteria by two independent observers to assess interobserver variability. 9p status in ccRCC tumours was determined and correlated to clinicopathological variables, recurrence-free survival and disease-specific survival. RESULTS: There were 80 ccRCCs with valid 9p scoring and a median follow-up of 95 months. Kappa statistic for interobserver variability was 0.71 (good agreement). 9p deletion was detected in 44% of ccRCCs. 9p loss was associated with higher stage, larger tumours, necrosis, microvascular and renal vein invasion, and higher SSIGN (stage, size, grade and necrosis) score. Patients with 9p-deleted ccRCC were at a higher risk of recurrence (P=0.008) and RCC-specific mortality (P=0.001). On multivariate analysis, 9p deletion was an independent predictor of recurrence (hazard ratio 4.323; P=0.021) and RCC-specific mortality (hazard ratio 4.603; P=0.007). The predictive accuracy of SSIGN score improved from 87.7% to 93.1% by integrating 9p status to the model (P=0.001). CONCLUSIONS: Loss of 9p is associated with aggressive ccRCC and worse prognosis in patients following surgery. Our findings independently confirm the findings of previous reports relying on I-FISH to detect 9p (CDKN2A) deletion
Microsatellite alteration and immunohistochemical expression profile of chromosome 9p21 in patients with sporadic renal cell carcinoma following surgical resection.
BACKGROUND: Long-term prognostic significance of loss of heterozygosity on chromosome 9p21 for localized renal cell carcinoma following surgery remains unreported. The study assessed the frequency of deletions of different loci of chromosome 9p along with immunohistochemical profile of proteins in surgically resected renal cancer tissue and correlated this with long-term outcomes. METHODS: DNA was extracted from renal tumours and corresponding normal kidney tissues in prospectively collected samples of 108 patients who underwent surgical resection for clinically localized disease between January 2001 and December 2005, providing a minimum of 9 years follow-up for each participant. After checking quality of DNA, amplified by PCR, loss of heterozygosity (LOH) on chromosome 9p was assessed using 6 microsatellite markers in 77 clear cell carcinoma. Only 5 of the markers showed LOH (D9S1814, D9S916, D9S974, D9S942, and D9S171). Protein expression of p15(INK4b), p16(INK4a), p14(ARF), CAIX, and adipose related protein (ADFP) were demonstrated by immunostaining in normal and cancer tissues. Loss of heterozygosity for microsatellite analysis was correlated with tumour characteristics, recurrence free, cancer specific, and overall survival, including significance of immunohistochemical profile of protein expressions. RESULTS: The main deletion was found at loci telomeric to CDKN2A region at D9S916. There was a significant correlation between frequency of LOH stage (p = 0.005) and metastases (p = 0.006) suggesting a higher LOH for advanced and aggressive renal cell carcinoma. Most commonly observed LOH in the 3 markers: D9S916, D9S974, and D9S942 were associated with poor survival, and were statistically significant on multivariate analysis. Immunohistochemical expression of p14, p15, and p16 proteins were either low or absent in cancer tissue compared to normal. CONCLUSIONS: Loss of heterozygosity of p921 chromosome is associated with aggressive tumours, and predicts cancer specific or recurrence free survival on long-term follow-up. Egyptian truffles as a source of antimicrobial and antioxidant agents
Edible desert truffles are obligate symbiotic macro hypogenous ascomycetes fruit bodies. These are growing in depth between 5 and 10 cm. These consider a miracle of nature and an unexploited source of therapeutic compounds with antimicrobial, antioxidant, and wonderful food, especially for Bedouins. The current study investigated the activity of premature and mature ethyl acetate truffle extracts (white and Red) collected from the Western Egyptian Desert as antimicrobial and antioxidant. This study evaluated in vitro the efficacy of antimicrobial activity of organic truffles extract, and their effect on various pathogens (Gram-negative, Gram-positive bacteria, filamentous fungi, and yeast) by using agar well diffusion. TEM micrographs had been done for the most effective crude extracts. Furthermore, the activity of DPPH scavenging was studied for both mature truffles. Both truffles extracts had antibacterial activity more than antifungal activity. The selected extracts exhibited an inhibitory effect on the cell wall and protoplasm of pathogens. Terfezia sp. had antioxidant activity more than Tirmania sp. This investigation concluded that the truffle extracts could be considered a promising antibiotic and antioxidant drug in near future
Design and performance evaluation of vehicular visible light communication system under different weather conditions and system parameters
Vehicular visible light communication is an emerging technology that allows wireless communication between vehicles or between vehicles and infrastructure. In this paper, a vehicular visible light communication system is designed using a non-return to zero on-off keying modulation scheme under the effect of different weather conditions such as clear, haze, and fog. The first model is a light emitting diode-based system and the second is a laser diode-based system. For both models, the influence of system parameters such as beam divergence, transceiver aperture diameters, and receiver responsivity is studied. The impact of the use of the trans-impedance amplifier is also investigated for both models. It was concluded that in the presence of the amplifier, output power of the light emitting diode and laser diode model are increased by 98.46 μW and 0.4719 W, respectively. The performance of the two proposed models is evaluated through bit error rate, quality factor, eye diagram, and output power to have some insightful results about the quality of service for the two proposed models. Under a specific weather condition, the performance of the system would be critical and other techniques should be applied. The maximum achievable link distance for the laser-based and light-emitting diode-based systems is 190 m at a data rate of 25 Gbps and 80 m at a data rate of 60 kbps, respectively, under the same system parameters and weather conditions. The obtained results provide a full idea about the availability of constructing our proposed model in a practical environment, showing a higher performance of the laser diode-based model than that of the light emitting diode-based model
