    ANTIMICROBIAL RESISTANCE PATTERNS OF PROTEUS ISOLATES FROM CLINICAL SPECIMENS

    Total of 6840 clinical samples were collected from King Fahd Hospital at Medina, Kingdom of Saudi Arabia (KSA). Clinical samples were screened for Proteus spp. It is found that Proteus spp isolates representing about 3% of all positive samples isolated from different clinical sources. Males were found to be more vulnerable than females in acquiring Proteus infections. Results showed that the greatest number of Proteus spp. isolates from clinical specimens were isolated from wound and sputum swabs specimens representing about 88% of all clinical specimens.Thirteen different antibiotics representing different families of antibiotics were tested on Proteus spp. specimens for studying its antimicrobial sensitivity pattern. Results of antimicrobial sensitivity tests revealed that imipenem (IMP) antibiotic was the most effective antibiotic against Proteus spp. with 91% of antimicrobial sensitivity. Imipenem (Imp) was followed by amikacin (AK) with 61% of antimicrobial sensitivity.Results are recommending prescribing of imipenem (IMP) antibiotic in the treatment of Proteus spp infections. It is recommended also to prescribe amikacin (AK) in the case of sensitivity to imipenem (IMP). In acute cases, Proteus infections could require use of combined antimicrobial therapy (imipenem- amikacin). Results also demonstrated that percentage of Proteus spp. infections was highest during summer season representing about 40% of infections all year along. Summer season was followed by winter season with about 32% of infections

    Ultrasonographic Appearance and Echo-pattern Characterization of Donkeys’ Internal Reproductive Organs

    The current study was achieved to designate the accessory sex glands, ampullae and urethra of 25 healthy male donkeys using the ultrasonography. Animals were classified according to their age into young peri-mature (i.e. ≀2 years; n=7) and old mature (>3 years; n=18). The examination was done per rectum by using Magic 2200 scanner with a 6 MHz linear transrectal transducer. The position of each gland was allocated, and length and width were measured. The dimensions of prostate gland lobes and ampulla ductus deferens significantly differed between the examined groups. The echo-pattern analysis of vesicular gland and ampulla ductus deferens, in terms of mean pixel value (MPV), pixel intensity (PI) and pixel number (PN), significantly differed between premature and mature donkeys. Age was significantly correlated with all sex glands measurements. There was a substantial confident relationship between MPV and PI and the dimensions of vesicular and ampulla ductus deferens. In the meantime, PN was clearly negatively correlated with vesicular gland length and width. In conclusions, ultrasound is a feasible non-invasive diagnostic tool that could be used to characterize the accessory glands along the course of the pelvic urethra of male donkeys reared in Egypt. The addressed measures as well as acoustic physiognomies of the secondary sex organs are considered a promising guide in fertility potential prediction and/or the discrimination of the pathological conditions of male reproductive organs

    Evaluation of Hospital Wastewater Treatment Using Sewage Treatment Plant for Heavy Metals, Radionuclides, and Some Pharmaceuticals: A Case Study

    This is the first study in Oman to evaluate the efficiency of a sewage treatment plant (STP) for hospital wastewater (HWW) treatment for heavy metals, radionuclides, and some selected pharmaceuticals. A sewage treatment plant (STP) at Sultan Qaboos University (SQU) receives HWW, from Sultan Qaboos University Hospital (SQUH), and municipal wastewater from non-medical facilities at SQU. Representative samples of HWW (before mixing with municipal wastewater at STP), STP-treated wastewater (TWW), and STP mixing sludge, were collected and analyzed. A method for analyzing pharmaceuticals including metformin, atenolol, chlorpheniramine, triprolidine, diphenhydramine, and citalopram was developed and validated using LC-MS-MS. HWW and TWW show low concentrations of heavy metals. Radionuclides found in HWW include Cs137, K40, Ra226, Th234, I131, Tl208, Zn65 Ac228, Sb125, Bi124 and Be7. Diphenhydramine (2.24 mg/L), chlorpheniramine (0.293 mg/L) and atenolol (0.0260 mg/L) were found in HWW. Heavy metals, radionuclides, and pharmaceuticals were found less in TWW than in HWW. STP sewage sludge showed higher levels of these pollutants than HWW or TWW. Concentrations of diphenhydramine, chlorpheniramine, and citalopram were 137, 0.950, and 169 mg/kg, respectively in dried sewage sludge. The study confirms the ineffectiveness of STP treatment to completely remediate HWW. HWW should be considered hazardous and requires physico-chemical treatment before mixing with municipal wastewater. Keywords: Hospital, pharmaceuticals, radionuclides, heavy metals, wastewater

    The impact of multi-enzyme fortification on growth performance, intestinal morphology, nutrient digestibility, and meat quality of broiler chickens fed a standard or low-density diet

    This research aimed to study the impact of supplementation of three multi-enzyme levels (0, 0.1, and 0.2% of feed) and two levels of dietary treatments [standard diet (SD) and low-density diet (LDD)] on growth performance, carcass traits, digestibility, and meat quality of broilers from 1 to 38 days of age. A total of 216 1-day-old Arbor Acres broiler chicks were randomly assigned to a factorial experiment (2 × 3) comprising six dietary treatments, each with six replicates and each replicate with six chickens. The results showed that the LDD significantly reduced body weight gain by 5.0%, compared with the SD. Multi-enzymes significantly improved body weight gain and the production index (PI) relative to the SD. The feed conversion ratio was significantly enhanced with increased multi-enzymes from 1 to 21 days. A significant relation between the multi-enzyme concentration and type of dietary treatment was observed in body weight gain and feed conversion ratio from 1 to 21 days of age. Nitrogen-free extract digestibility was significantly increased by using the SD diet compared with using the LDD. Multi-enzyme supplementation improved the digestibility of dry matter, crude protein, crude fiber, and nitrogen-free extract in the LDD. A significant relationship was found between the multi-enzyme concentration and type of dietary treatment on the pancreas, liver, and intestinal length percentages. The meat dry matter concentration was significantly higher in the LDD group than in the SD group. The low-density diet significantly reduced the total revenue compared with the SD, whereas broilers fed the SD recorded significantly higher total revenue and economic efficiency than those fed the LDD. The low-density diet significantly increased economic efficiency compared with the SD. Multi-enzymes significantly increased the total revenue, net revenue, and economic efficiency than the standard set. In conclusion, using multi-enzymes in broiler diets improved body weight gain. The LDD with multi-enzymes showed enhanced body weight gain compared with the SD without multi-enzymes

    Development of new thiazolidine-2,4-dione hybrids as aldose reductase inhibitors endowed with antihyperglycaemic activity: design, synthesis, biological investigations, and in silico insights

    This research study describes the development of new small molecules based on 2,4-thiazolidinedione (2,4-TZD) and their aldose reductase (AR) inhibitory activities. The synthesis of 17 new derivatives of 2,4-TZDs hybrids was feasible by incorporating two known bioactive scaffolds, benzothiazole heterocycle, and nitro phenacyl moiety. The most active hybrid (8b) was found to inhibit AR in a non-competitive manner (0.16 ”M), as confirmed by kinetic studies and molecular docking simulations. Furthermore, the in vivo experiments demonstrated that compound 8b had a significant hypoglycaemic effect in mice with hyperglycaemia induced by streptozotocin. Fifty milligrams per kilogram dose of 8b produced a marked decrease in blood glucose concentration, and a lower dose of 5 mg/kg demonstrated a noticeable antihyperglycaemic effect. These outcomes suggested that compound 8b may be used as a promising therapeutic agent for the treatment of diabetic complications

    A descriptive analysis of relations between parents' self-reported smoking behavior and infants' daily exposure to environmental tobacco smoke

    <p>Abstract</p> <p>Background</p> <p>The aims of the present study were to examine relations between parents' self-reported smoking behavior and infants' daily exposure to environmental tobacco smoke, as assessed by urinary cotinine-to-creatinine ratio (CCR), and to describe the CCR over seven days among infants at home.</p> <p>Methods</p> <p>A convenience sample of 27 households was drawn. Each household had to have at least one daily tobacco smoker and one child up to three years of age. Over a seven-day period, urine samples were obtained from the child daily. To examine relations between parents' self-reported smoking and infants' daily CCR, generalized estimating equation (GEE) analysis was used.</p> <p>Results</p> <p>The data revealed that infants from households with indoor smoking had higher CCRs than infants in households with outdoor smoking. CCRs were higher in girls than in boys. Older infants had lower CCRs than younger infants. Smoking outside the home versus inside the home, infant's gender, and infants' age accounted for 68% of the variance in CCR in a GEE data analysis model. No increase or decrease of CCR over time was found.</p> <p>Conclusion</p> <p>The findings suggest that parents' self-reported smoking indoors at home versus outdoors is predictive of CCR among infants three and younger. Higher CCR concentrations in girls' urine need further examination. Furthermore, significant fluctuations in daily CCR were not apparent in infants over a seven-day time period.</p

    Self-oligomerization regulates stability of survival motor neuron protein isoforms by sequestering an SCF<sup>Slmb</sup> degron

    Spinal muscular atrophy (SMA) is caused by homozygous mutations in human SMN1. Expression of a duplicate gene (SMN2) primarily results in skipping of exon 7 and production of an unstable protein isoform, SMNΔ7. Although SMN2 exon skipping is the principal contributor to SMA severity, mechanisms governing stability of survival motor neuron (SMN) isoforms are poorly understood. We used a Drosophila model system and label-free proteomics to identify the SCFSlmb ubiquitin E3 ligase complex as a novel SMN binding partner. SCFSlmb interacts with a phosphor degron embedded within the human and fruitfly SMN YG-box oligomerization domains. Substitution of a conserved serine (S270A) interferes with SCFSlmb binding and stabilizes SMNΔ7. SMA-causing missense mutations that block multimerization of full-length SMN are also stabilized in the degron mutant background. Overexpression of SMNΔ7S270A, but not wild-type (WT) SMNΔ7, provides a protective effect in SMA model mice and human motor neuron cell culture systems. Our findings support a model wherein the degron is exposed when SMN is monomeric and sequestered when SMN forms higher-order multimers
