Modified approach to estimating daily methane emissions of dairy cows by measuring filtered eructations during milking
The aim of this study was to compare metrics for quantifying enteric methane (CH4) emissions from individual cows during milking using frequent spot measurements and peak analysis methods. An infrared gas analyser was used to measure the CH4 emitted by cows, and eructation peaks were identified using a Signal Processing Toolbox provided by Matlab. CH4 emissions were quantified by gas peak height, peak amplitude and average concentration, and were expressed in grams per day and CH4 yield (grams per kilogram of dry matter intake (DMI)). Peak analysis measurements of CH4 were obtained from 36 cows during 2,474 milkings, during which cows were fed a ration containing between 39 and 70 % forage. Spot measurements of CH4 were compared to a separate dataset of 196 chamber CH4 records from another group of 105 cows, which were fed a ration containing between 25 and 80 % forage. The results showed that the metrics of CH4 peak height and CH4 peak amplitude demonstrated similar positive relationships between daily CH4 emissions and DMI (both r = 0.37), and a negative relationship between CH4 yield and DMI (r = -0.43 and -0.38 respectively) as observed in the chamber measurements (r = 0.57 for daily emissions and r = -0.40 for CH4 yield). The CH4 metrics of peak height and peak amplitude were highly repeatable (ranging from 0.76 to 0.81), comparable to the high repeatability of production traits (ranging from 0.63 to 0.99) and were more repeatable than chamber CH4 measurements (0.31 for daily emissions and 0.03 for CH4 yield). With the aim to evaluate the effectiveness of techniques and measures for control of cereal bunts and smuts in organic farming systems, field trials were carried out in multiple locations from 2004 to 2006. The relevant fungal diseases included in the trials were common bunt (Tilletia tritici), dwarf bunt (T. controversa), loose smut (Usti-lago tritici, U. nuda), covered smut (U. hordei) and black loose smut (U. avenae). The following treatments were found to be effective. Common bunt: different physical methods, Tillecur, acetic acid. Loose smut of barley and wheat: warm and hot water treatment, seed treatment with ethanol (more trials needed). Covered smut of barley: warm water treatment, ethanol, Lebermooser. Loose smut of oats: hot water treat-ment, ethanol. Field resistant varieties are available (although in some cases only in limited number) for common bunt, loose smut of wheat and barley and covered smut. The status of varietal resistance against dwarf bunt is only insufficiently known. In case of black loose smut research on resistant varieties is currently being conducted
Column amounts of trace gases from ground based FTIR measurements in the late north polar winters 1990 and 1991
Two FTIR spectrometers were employed in the late winters 1990 and 1991 in Esrange, North Sweden, and in Ny Aalesund, Spitsbergen to detect zenith column amounts of several trace gases. Time series of column amounts of the trace gases O3, N2O, CH4, HNO3, NO2, CHl, and HF have been derived from the measured spectra. Additionally, some information on the vertical distribution of HCl could be obtained by analyzing the spectral line shapes. The results are interpreted in terms of dynamical and chemical processes
Comparative Responses in Lung Function Measurements with Tiotropium in Adolescents and Adults, and Across Asthma Severities:A Post Hoc Analysis
INTRODUCTION: Airway obstruction is usually assessed by measuring forced expiratory volume in 1 s (FEV1), forced vital capacity (FVC) and peak expiratory flow (PEF). This post hoc study investigated comparative responses of lung function measurements in adults and adolescents (full analysis set, N = 3873) following treatment with tiotropium Respimat®. METHODS: Lung function outcomes were analysed from five phase III trials in adults (≥ 18 years) with symptomatic severe, moderate and mild asthma (PrimoTinA-asthma®, MezzoTinA-asthma® and GraziaTinA-asthma®, respectively), and one phase III trial in adolescents (12-17 years) with symptomatic moderate asthma (RubaTinA-asthma®). Changes from baseline versus placebo in FEV1, FVC, PEF and FEV1/FVC ratio with tiotropium 5 µg or 2.5 µg added to at least stable inhaled corticosteroids at week 24 (week 12 in GraziaTinA-asthma) were analysed. RESULTS: All lung function measures improved in all studies with tiotropium 5 µg (mean change from baseline versus placebo), including peak FEV1 (110-185 mL), peak FVC (57-95 mL) and morning PEF (15.8-25.6 L/min). Changes in adolescents were smaller than those in adults, and were statistically significant primarily for FEV1 and PEF, but not for FVC. CONCLUSION: Consistent improvements were seen across all lung function measures with the addition of tiotropium to other asthma treatments in adults across all severities, whereas the improvements with tiotropium in adolescents primarily impacted measures of flow rather than lung volume. This may reflect less pronounced airway remodelling and air trapping in adolescents with asthma versus adults

Over the last century, eosinophils have been regarded ambiguously either as 'friends' or 'foes'. Recent developments have greatly enhanced our understanding of the role and function of eosinophils in health and disease. Pathogenic eosinophilic inflammation can lead to severe diseases in various organs, such as the gastrointestinal tract, airways, heart and skin. In a 2-day focus workshop of the German Society for Allergology and Clinical Immunology (DGAKI), the state of the art was discussed and practical recommendations for diagnosis and treatment of eosinophilic diseases, with a particular focus on new biologics, such as anti-interleukin 5 and anti-interleukin 5R, were derived
Contribution of macrophages to neural survival and intracochlear tissue remodeling responses following cochlear implantation
BACKGROUND: Cochlear implants (CIs) restore hearing to deafened patients. The foreign body response (FBR) following cochlear implantation (post-CI) comprises an infiltration of macrophages, other immune and non-immune cells, and fibrosis into the scala tympani, a space that is normally devoid of cells. This FBR is associated with negative effects on CI outcomes including increased electrode impedances and loss of residual acoustic hearing. This study investigates the extent to which macrophage depletion by an orally administered CSF-1R specific kinase (c-FMS) inhibitor, PLX-5622, modulates the tissue response to CI and neural health.
MAIN TEXT: 10- to 12-week-old CX3CR1 + /GFP Thy1 + /YFP mice on C57BL/6J/B6 background was fed chow containing 1200 mg/kg PLX5622 or control chow for the duration of the study. 7 days after starting the diet, 3-channel cochlear implants were implanted in the ear via the round window. Serial impedance and neural response telemetry (NRT) measurements were acquired throughout the study. Electric stimulation began 7 days post-CI until 28 days post-CI for 5 h/day, 5 days/week, with programming guided by NRT and behavioral responses. Cochleae harvested at 10, 28 or 56 days post-CI were cryosectioned and labeled with an antibody against α-smooth muscle actin (α-SMA) to identify myofibroblasts and quantify the fibrotic response. Using IMARIS image analysis software, the outlines of scala tympani, Rosenthal canal, modiolus, and lateral wall for each turn were traced manually to measure region volume. The density of nuclei, CX3CR1 + macrophages, Thy1 + spiral ganglion neuron (SGN) numbers, and the ratio of the α-SMA + volume/scala tympani volume were calculated. Cochlear implantation in control diet subjects caused infiltration of cells, including macrophages, into the cochlea. Fibrosis was evident in the scala tympani adjacent to the electrode array. Mice fed PLX5622 chow showed reduced macrophage infiltration throughout the implanted cochleae across all time points. However, scala tympani fibrosis was not reduced relative to control diet subjects. Further, mice treated with PLX5622 showed increased electrode impedances compared to controls. Finally, treatment with PLX5622 decreased SGN survival in implanted and contralateral cochleae.
CONCLUSION: The data suggest that macrophages play an important role in modulating the intracochlear tissue response following CI and neural survival
Grazing management for soil carbon in Australia: A review
The livestock industry accounts for a considerable proportion of agricultural greenhouse gas emissions, and in response, the Australian red meat industry has committed to an aspirational target of net-zero emissions by 2030. Increasing soil carbon storage in grazing lands has been identified as one method to help achieve this, while also potentially improving production and provision of other ecosystem services. This review examined the effects of grazing management on soil carbon and factors that drive soil carbon sequestration in Australia. A systematic literature search and meta-analysis was used to compare effects of stocking intensity (stocking rate or utilisation) and stocking method (i.e, continuous, rotational or seasonal grazing systems) on soil organic carbon, pasture herbage mass, plant growth and ground cover. Impacts on below ground biomass, soil nitrogen and soil structure are also discussed. Overall, no significant impact of stocking intensity or method on soil carbon sequestration in Australia was found, although lower stocking intensity and incorporating periods of rest into grazing systems (rotational grazing) had positive effects on herbage mass and ground cover compared with higher stocking intensity or continuous grazing. Minimal impact of grazing management on pasture growth rate and below-ground biomass has been reported in Australia. However, these factors improved with grazing intensity or rotational grazing in some circumstances. While there is a lack of evidence in Australia that grazing management directly increases soil carbon, this meta-analysis indicated that grazing management practices have potential to benefit the drivers of soil carbon sequestration by increasing above and below-ground plant production, maintaining a higher residual biomass, and promoting productive perennial pasture species. Probabilistic Model-Based Safety Analysis
Model-based safety analysis approaches aim at finding critical failure
combinations by analysis of models of the whole system (i.e. software,
hardware, failure modes and environment). The advantage of these methods
compared to traditional approaches is that the analysis of the whole system
gives more precise results. Only few model-based approaches have been applied
to answer quantitative questions in safety analysis, often limited to analysis
of specific failure propagation models, limited types of failure modes or
without system dynamics and behavior, as direct quantitative analysis is uses
large amounts of computing resources. New achievements in the domain of
(probabilistic) model-checking now allow for overcoming this problem.
This paper shows how functional models based on synchronous parallel
semantics, which can be used for system design, implementation and qualitative
safety analysis, can be directly re-used for (model-based) quantitative safety
analysis. Accurate modeling of different types of probabilistic failure
occurrence is shown as well as accurate interpretation of the results of the
analysis. This allows for reliable and expressive assessment of the safety of a
system in early design stages
