    TP53-induced glycolysis and apoptosis regulator promotes proliferation and invasiveness of nasopharyngeal carcinoma cells

    The TP53induced glycolysis and apoptosis regulator (TIGAR) is the protein product of the p53 target gene, C12orf5. TIGAR blocks glycolysis and promotes cellular metabolism via the pentose phosphate pathway; it promotes the production of cellular nicotinamide adenine dinucleotide phosphate (NADPH), which leads to enhanced scavenging of intracellular reactive oxygen species, and inhibition of oxidative stressinduced apoptosis in normal cells. Our previous study identified a novel nucleoside analog that inhibited cellular growth and induced apoptosis in nasopharyngeal carcinoma (NPC) cell lines via downregulation of TIGAR expression. Furthermore, the growth inhibitory effects of cMet tyrosine kinase inhibitors were ameliorated by the overexpression of TIGAR in the NPC cell lines. These results indicate a significant role for TIGAR expression in the survival of NPCs. The present study aimed to further define the function of TIGAR expression in NPC cells. In total, 36 formalinfixed, paraffinembedded NPC tissue samples were obtained for the immunohistochemical determination of TIGAR expression. The effects of TIGAR expression on cell proliferation, NADPH production and cellular invasiveness were also assessed in NPC cell lines. Overall, TIGAR was overexpressed in 27/36 (75%) of the NPC tissues compared with the adjacent noncancer epithelial cells. Similarly, TIGAR overexpression was also observed in a panel of six NPC cell lines compared with normal NP460 hTert and Het1A cell lines. TIGAR overexpression led to increased cellular growth, NADPH production and invasiveness of the NPC cell lines, whereas a knockdown of TIGAR expression resulted in significant inhibition of cellular growth and invasiveness. The expression of the two mesenchymal markers, fibronectin and vimentin, was increased by TIGAR overexpression, but reduced following TIGARknockdown. The present study revealed that TIGAR overexpression led to increased cellular growth, NADPH production and invasiveness, and the maintenance of a mesenchymal phenotype, in NPC tissues.published_or_final_versio

    Validation of the Chinese version of underwood's daily spiritual experience scale-transcending cultural boundaries?

    Background: Daily spiritual experience (DSE) refers to one's interaction with the transcendent in day-to-day life. Underwood's Daily Spiritual Experience Scale mic(DSES) was developed to measure this experiential component of religiousness and spirituality. Addressing ordinary daily experiences rather than particular beliefs, DSES has transcultural applicability potential. Purpose: The current study aimed to develop and evaluate the Chinese version of DSES (DSES-C). Method: The 16-item scale was translated faithfully through standard translation/back-translation procedures. The term "God" required an extended definition embracing both a humanized and a philosophical higher power in Chinese culture. The translated scale plus a battery of validation scales were administered to staff of a large rehabilitation service complex, resulting in 245 completed questionnaires. Results: Exploratory factor analysis revealed a similar factor structure as the original English version and similar problems with items 13 (compassion) and 14 (mercy). After carefully deliberating on the Chinese conceptualization of spirituality and balancing psychometric properties, the one-factor 16-item structure of the English version was supported. DSES-C showed high internal consistency (Cronbach's alpha=0.97). Construct validity was supported by correlations with validation scales in expected directions. Conclusion: The psychometric properties of DSES were similar to the English version in factor structure, internal consistency, and convergence/divergence construct validity. © 2009 International Society of Behavioral Medicine.link_to_subscribed_fulltex

    Practice of breastfeeding and factors that affect breastfeeding in Hong Kong

    Objectives. To describe the patterns of and factors affecting breastfeeding and to find out any significant relationship between breastfeeding and health of the child. Design. Cohort study. Setting. Postnatal ward of the Prince of Wales Hospital. Participants. A total of 243 infants born in 1998 to 2001 at the hospital. Each infant was followed up for 3 years. Home visits were carried out at 3, 15, 24, and 36 months of age by medical students from the Chinese University of Hong Kong. A questionnaire was completed at each visit. Independent sample t-tests and Pearson Chi squared tests were used. Results. Of the 243 subjects, 213 provided data on the method of infant feeding. There were 66.7% of mothers initiating breastfeeding, with a median duration of 1 month. Only 13.4% met the World Health Organization's recommendations on breastfeeding. Breastfeeding was found to have a statistically significant relationship with (i) the infant's birth order and (ii) the mother's and father's education level. During follow-up, 44.6% of the infants were hospitalised but there was no significant relationship between breastfeeding and number of hospitalisations. Conclusions. The current breastfeeding rate in Hong Kong falls below expectations when compared with other developed nations. To raise this rate, more support is needed for families with parents having a lower education level or more than two children, as they are the least likely to breastfeed. This might be achieved by encouraging antenatal class attendance, counselling of husbands, and more support for breastfeeding from doctors.link_to_subscribed_fulltex

    STAT3 activation contributes directly to Epstein-Barr virus-mediated invasiveness of nasopharyngeal cancer cells in vitro

    Nasopharyngeal cancer (NPC) is an Epstein-Barr virus (EBV)-associated head and neck cancer prevalent in Asia. Although with reasons not fully understood, the intrinsic invasiveness of NPC is believed to be EBV-linked. Recently, EBV was found to induce STAT3 activation. Constitutive STAT3 activation correlated with advanced clinical staging in NPC. We hypothesized that STAT3 activation by EBV directly contributes to the intrinsic invasiveness of NPC cells. Phospho-STAT3-Tyr705 was detected in high percentage of NPC tumors (7/10 cases). Using a paired NPC cell line model, HONE-1 and the EBV-infected counterpart, HONE-1-EBV, we found that HONE-1-EBV expressed a higher level of phospho-STAT3-Tyr705 and was ∼11-fold more invasive than HONE-1. In HONE-1-EBV, STAT3 siRNA targeting inhibited both spontaneous and serum-induced invasion, as well as cell growth. Conversely, activation of STAT3 (by expressing an activated STAT3 mutant, namely STAT3C) in the parental HONE-1, mimicking EBV-induced STAT3 activation, significantly enhanced its invasiveness and proliferation, which was accompanied by increased expression of markers of mesenchymal status, proliferation and anti-apoptosis. Our results demonstrated that EBV-induced STAT3 activation is responsible for NPC cell proliferation and invasion. This was further confirmed by a small molecule inhibitor of JAK/STAT3, JSI-124. JSI-124 inhibited STAT3 activation in HONE-1-EBV, with subsequent growth inhibition, induction of PARP cleavage, abrogation of anchorage-independent growth and invasion. We found that EBV-independent activation of STAT3 by a growth factor, EGF, also contributed to NPC invasion. In conclusion, EBV-induced STAT3 activation directly contributes to the intrinsic invasiveness of NPC cells and STAT3 targeting may be beneficial in treating aggressive NPC. © 2009 UICC.link_to_subscribed_fulltex

    A computer-aided design system for foot-feature-based shoe last customization

    There has been a growing trend among shoe manufacturers to introduce customized shoes to satisfy varying customer style, fit, and comfort needs, thus to increase the product's added value. This study presents a computer-aided design (CAD) system for designing a customized shoe last based on the chosen shoe style and customer's foot features. The CAD system first automatically extracts 18 important foot features from a laser-scanned customer's foot. Then, it applies a global grading with a local deformation approach that can deform the base shoe last with the customer's chosen style to the customized shoe last based on the extracted foot features while maintaining the customer's chosen style. Finally, the system evaluates the final foot shoe last fit and represents the fit in a contoured figure. The experimental results show that the proposed CAD system can be adopted by shoe manufacturers to make customized shoes with the customer's chosen style and foot size and shape.close121
