On massless electron limit for a multispecies kinetic system with external magnetic field
We consider a three-dimensional kinetic model for a two species plasma
consisting of electrons and ions confined by an external nonconstant magnetic
field. Then we derive a kinetic-fluid model when the mass ratio tends
to zero. Each species initially obeys a Vlasov-type equation and the
electrostatic coupling follows from a Poisson equation. In our modeling, ions
are assumed non-collisional while a Fokker-Planck collision operator is taken
into account in the electron equation. As the mass ratio tends to zero we show
convergence to a new system where the macroscopic electron density satisfies an
anisotropic drift-diffusion equation. To achieve this task, we overcome some
specific technical issues of our model such as the strong effect of the
magnetic field on electrons and the lack of regularity at the limit. With
methods usually adapted to diffusion limit of collisional kinetic equations and
including renormalized solutions, relative entropy dissipation and velocity
averages, we establish the rigorous derivation of the limit model

Detection of subtelomere imbalance using MLPA: validation, development of an analysis protocol, and application in a diagnostic centre
BACKGROUND: Commercial MLPA kits (MRC-Holland) are available for detecting imbalance at the subtelomere regions of chromosomes; each kit consists of one probe for each subtelomere. METHODS: For validation of the kits, 208 patients were tested, of which 128 were known to be abnormal, corresponding to 8528 genomic regions overall. Validation samples included those with trisomy 13, 18 and 21, microscopically visible terminal deletions and duplications, sex chromosome abnormalities and submicroscopic abnormalities identified by multiprobe FISH. A robust and sensitive analysis system was developed to allow accurate interpretation of single probe results, which is essential as breakpoints may occur between MLPA probes. RESULTS: The validation results showed that MLPA is a highly efficient technique for medium-throughput screening for subtelomere imbalance, with 95% confidence intervals for positive and negative predictive accuracies of 0.951-0.996 and 0.9996-1 respectively. A diagnostic testing strategy was established for subtelomere MLPA and any subsequent follow-up tests that may be required. The efficacy of this approach was demonstrated during 15 months of diagnostic testing when 455 patients were tested and 27 (5.9%) abnormal cases were detected. CONCLUSION: The development of a robust, medium-throughput analysis system for the interpretation of results from subtelomere assays will be of benefit to other Centres wishing to implement such an MLPA-based service
Dissymmetrical U-Shaped Ï-Stacked Supramolecular Assemblies by Using a Dinuclear CuI Clip with Organophosphorus Ligands and Monotopic Fully Ï-Conjugated Ligands
International audienceReactions between the U-shape binuclear CuI complex A bearing short metal-metal distances and the cyano-capped monotopic Ï-conjugated ligands 1-5 carrying gradually bulkier polyaromatic terminal fragments lead to the formation of Ï-stacked supramolecular assemblies 6-10 respectively in 50-80 % yields. These derivatives have been characterized by multinuclear NMR spectroscopy and X-ray diffraction studies. Their solid state structures show the selective formation of U-shaped supramolecular assemblies in which two monotopic Ï-conjugated systems present large (6,7,9) or medium (8,10) intramolecular Ï-overlap revealing Ï-Ï interactions. These assemblies self-organize into head-to-tail Ï-stacked dimers that in turn self-assemble affording infinite columnar Ï-stacks. The nature, the extentand the complexity of the intermolecular contacts within the head-to-tail Ï-stacked dimer depends on the nature of the terminal polyaromatic fragment carried by the cyano-capped monotopic ligand but it does not alter the result of the self-assembling process. These results demonstrate that the dinuclear molecular clip A bearing short metal-metal distance allows selective supramolecular assembling processes driven by the formation of intra- and intermolecular short Ï-Ï interactions in the resulting self-assembled structures demonstrating that their shape is not only dictated by the symmetry of the building blocks. This approach opens perspectives toward the formation of extented Ï-stacked columns based on dissymmetrical and functionnal Ï-conjugated systems
Serum levels of selenium and smoking habits at age 50 influence long term prostate cancer risk; a 34 year ULSAM follow-up
Background: Serum selenium level (s-Se) has been associated with prostate cancer (PrCa) risk. We investigated the relation between s-Se, smoking and non-screening detected PrCa and explored if polymorphisms in two DNA repair genes: OGG1 and MnSOD, influenced any effect of s-Se. Methods: ULSAM, a population based Swedish male cohort (n = 2322) investigated at age 50 for s-Se and s-Se influencing factors: serum cholesterol, erythrocyte sedimentation rate and smoking habits. At age 71 a subcohort, (n = 1005) was genotyped for OGG1 and MnSOD polymorphisms. Results: In a 34-year-follow-up, national registries identified 208 PrCa cases further confirmed in medical records. Participants with s-Se in the upper tertile had a non-significantly lower risk of PrCa. Smokers with s-Se in the two lower tertiles (<= 80 mu g/L) experienced a higher cumulative incidence of PrCa than smokers in the high selenium tertile (Hazard Ratio 2.39; 95% CI: 1.09-5.25). A high tertile selenium level in combination with non-wt rs125701 of the OGG1 gene in combination with smoking status or rs4880 related variation of MnSOD gene appeared to protect from PrCa. Conclusions: S-Se levels and smoking habits influence long-term risk of PrCa. Smoking as a risk factor for PrCa in men with low s-Se is relevant to explore further. Exploratory analyses of variations in OGG1 and MnSOD genes indicate that hypotheses about patterns of exposure to selenium and smoking combined with data on genetic variation in genes involved in DNA repair can be valuable to pursue

Do genetic factors protect for early onset lung cancer? A case control study before the age of 50 years
<p>Abstract</p> <p>Background</p> <p>Early onset lung cancer shows some familial aggregation, pointing to a genetic predisposition. This study was set up to investigate the role of candidate genes in the susceptibility to lung cancer patients younger than 51 years at diagnosis.</p> <p>Methods</p> <p>246 patients with a primary, histologically or cytologically confirmed neoplasm, recruited from 2000 to 2003 in major lung clinics across Germany, were matched to 223 unrelated healthy controls. 11 single nucleotide polymorphisms of genes with reported associations to lung cancer have been genotyped.</p> <p>Results</p> <p>Genetic associations or gene-smoking interactions was found for <it>GPX1(Pro200Leu) </it>and <it>EPHX1(His113Tyr)</it>. Carriers of the Leu-allele of <it>GPX1(Pro200Leu) </it>showed a significant risk reduction of OR = 0.6 (95% CI: 0.4â0.8, p = 0.002) in general and of OR = 0.3 (95% CI:0.1â0.8, p = 0.012) within heavy smokers. We could also find a risk decreasing genetic effect for His-carriers of <it>EPHX1(His113Tyr) </it>for moderate smokers (OR = 0.2, 95% CI:0.1â0.7, p = 0.012). Considered both variants together, a monotone decrease of the OR was found for smokers (OR of 0.20; 95% CI: 0.07â0.60) for each protective allele.</p> <p>Conclusion</p> <p>Smoking is the most important risk factor for young lung cancer patients. However, this study provides some support for the T-Allel of <it>GPX1(Pro200Leu) </it>and the C-Allele of <it>EPHX1(His113Tyr) </it>to play a protective role in early onset lung cancer susceptibility.</p
USP18 inhibits NF-ÎșB and NFAT activation during Th17 differentiation by deubiquitinating the TAK1âTAB1 complex
Reversible ubiquitin modification of cell signaling molecules has emerged as a critical mechanism by which cells respond to extracellular stimuli. Although ubiquitination of TGF-ÎČâactivated kinase 1 (TAK1) is critical for NF-ÎșB activation in T cells, the regulation of its deubiquitination is unclear. We show that USP18, which was previously reported to be important in regulating type I interferon signaling in innate immunity, regulates T cell activation and T helper 17 (Th17) cell differentiation by deubiquitinating the TAK1âTAB1 complex. USP18-deficient T cells are defective in Th17 differentiation and Usp18(â/â) mice are resistant to experimental autoimmune encephalomyelitis (EAE). In response to T cell receptor engagement, USP18-deficient T cells exhibit hyperactivation of NF-ÎșB and NFAT and produce increased levels of IL-2 compared with the wild-type controls. Importantly, USP18 is associated with and deubiquitinates the TAK1âTAB1 complex, thereby restricting expression of IL-2. Our findings thus demonstrate a previously uncharacterized negative regulation of TAK1 activity during Th17 differentiation, suggesting that USP18 may be targeted to treat autoimmune diseases
Design, construction and operation of the ProtoDUNE-SP Liquid Argon TPC
The ProtoDUNE-SP detector is a single-phase liquid argon time projection chamber (LArTPC) that was constructed and operated in the CERN North Area at the end of the H4 beamline. This detector is a prototype for the first far detector module of the Deep Underground Neutrino Experiment (DUNE), which will be constructed at the Sandford Underground Research Facility (SURF) in Lead, South Dakota, U.S.A. The ProtoDUNE-SP detector incorporates full-size components as designed for DUNE and has an active volume of 7 Ă 6 Ă 7.2 m3. The H4 beam delivers incident particles with well-measured momenta and high-purity particle identification. ProtoDUNE-SP's successful operation between 2018 and 2020 demonstrates the effectiveness of the single-phase far detector design. This paper describes the design, construction, assembly and operation of the detector components
