Expression and purification of the immunogenically active fragment B of the Park Williams 8 Corynebacterium diphtheriae strain toxin
The construction of a hexahistidine-tagged version of the B fragment of diphtheria toxin (DTB) represents an important step in the study of the biological properties of DTB because it will permit the production of pure recombinant DTB (rDTB) in less time and with higher yields than currently available. In the present study, the genomic DNA of the Corynebacterium diphtheriae Park Williams 8 (PW8) vaccine strain was used as a template for PCR amplification of the dtb gene. After amplification, the dtb gene was cloned and expressed in competent Escherichia coli M15™ cells using the expression vector pQE-30™. The lysate obtained from transformed E. coli cells containing the rDTB PW8 was clarified by centrifugation and purified by affinity chromatography. The homogeneity of the purified rDTB PW8 was confirmed by immunoblotting using mouse polyclonal anti-diphtheria toxoid antibodies and the immune response induced in animals with rDTB PW8 was evaluated by ELISA and dermonecrotic neutralization assays. The main result of the present study was an alternative and accessible method for the expression and purification of immunogenically reactive rDTB PW8 using commercially available systems. Data also provided preliminary evidence that rabbits immunized with rDTB PW8 are able to mount a neutralizing response against the challenge with toxigenic C. diphtheriae
Misturas de forrageiras anuais e perenes para sucessão à soja em sistemas de integração lavoura‑pecuária
O objetivo deste trabalho foi avaliar a produção de forragem de espécies perenes e de suas misturas com espécies anuais, durante a estação seca, e a produtividade de grãos de soja cultivada em sucessão. Os experimentos foram realizados em 2007 e 2008, em Dourados e em São Gabriel do Oeste, MS. Em 2007, foram avaliadas as espécies Urochloa brizantha ('Xaraés') e Megathyrsus maximus ('Tanzânia'), em cultivos solteiros ou em misturas com Eleusine coracana (capim‑pé‑de‑galinha), Pennisetum glaucum (milheto) e Sorghum bicolor (sorgo forrageiro). Em 2008, foram avaliados U. decumbens e as cultivares Xaraés e Tanzânia em cultivos solteiros ou em misturas com milheto e sorgo forrageiro. As forrageiras foram avaliadas sob sucessivos cortes, durante a estação seca, e a soja no verão, no cultivo em sucessão. As misturas de espécies anuais e perenes não aumentam a produção de forragem, mas melhoram sua distribuição ao longo da estação seca. As forrageiras anuais têm maior participação na produção de forragem das misturas no início da estação seca, e as perenes no final dessa estação. A relação folha/colmo das forrageiras perenes individuais é maior do que a das misturas. Os tratamentos avaliados não têm efeito sobre a soja cultivada em sucessão
Anatomia foliar de Rudgea decipiens Müll. Arg. e R. macrophylla Benth. (Rubiaceae) Leaf anatomy of Rudgea decipiens Müll. Arg. and R. macrophylla Benth. [Rubiaceae]
São apresentados dados sobre a anatomia das folhas de Rudgea decipiens e R. macrophylla (Rubiaceae). Ambas ocorrem na Floresta de Tijuca (Mata Atlântica), Rio de Janeiro. As folhas são hipostomáticas, com estômatos do tipo paralelocítico, dotadas de mesofilo dorsiventral, feixes vasculares do tipo colateral, papilas na epiderme abaxial e cutícula estriada. Em R. macrophylla, destaca-se a ocorrência de estômatos peculiares.<br>Anatomical characteristics of Rudgea decipiens and R. macrophylla leaves were examined. The two species grow in Floresta da Tijuca (Mata Atlântica), Rio de Janeiro municipality. The leaves are hipostomatics, with parallelocytic stomata, dorsiventral mesophyll, colateral vascular bundles, papillae on the lower surface and striate cuticle. The stomata show peculiar aspects in R. macrophylla
Chemical and biochemical properties of Araucaria angustifolia (Bert.) Ktze. forest soils in the state of São Paulo
Araucaria angustifolia, commonly named Araucaria, is a Brazilian native species that is intensively exploited due to its timber quality. Therefore, Araucaria is on the list of species threatened by extinction. Despite the importance of soil for forest production, little is known about the soil properties of the highly fragmented Araucaria forests. This study was designed to investigate the use of chemical and biological properties as indicators of conservation and anthropogenic disturbance of Araucaria forests in different sampling periods. The research was carried out in two State parks of São Paulo: Parque Estadual Turístico do Alto do Ribeira and Parque Estadual de Campos de Jordão. The biochemical properties carbon and nitrogen in microbial biomass (MB-C and MB-N), basal respiration (BR), the metabolic quotient (qCO2) and the following enzyme activities: β-glucosidase, urease, and fluorescein diacetate hydrolysis (FDA) were evaluated. The sampling period (dry or rainy season) influenced the results of mainly MB-C, MB-N, BR, and qCO2. The chemical and biochemical properties, except K content, were sensitive indicators of differences in the conservation and anthropogenic disturbance stages of Araucaria forests. Although these forests differ in biochemical and chemical properties, they are efficient in energy use and conservation, which is shown by their low qCO2, suggesting an advanced stage of succession
