    Deer–vehicle collision trends at a suburban immunocontraception site

    Observations of extended estrus seasons in female white-tailed deer (Odocoileus virginianus) vaccinated with porcine zona pellucida (PZP) immunocontraceptives have led to speculation that management use of PZP would, perversely, increase the rate of deer–vehicle collisions. To test this hypothesis, we studied PZP-treated female deer at the National Institute of Standards and Technology, Gaithersburg, Maryland. PZP-treated female deer did not differ from untreated female deer in risk of death by vehicle collision, and the number of deer killed in vehicle collisions was uncorrelated with the number of deer treated with PZP, whether or not population size was controlled for. The reduction in deer population size observed during PZP applications was associated with a reduced frequency of deer–vehicle collisions. Our results suggest that any added collision risk caused by behavioral effects of PZP is unmeasurably small compared to the effects of population management and other mitigation actions

    Staurosporine Induces a Sequential Program of Mouse Keratinocyte Terminal Differentiation through Activation of PKC Isozymes

    Staurosporine (stsp) induces assembly of cornified envelopes in mouse keratinocyte cultures. To clarify whether this effect is the consequence of a coordinated differentiation program similar to that observed in epidermis, we assessed the expression of multiple differentiation-specific markers in stsp-treated keratinocytes. In medium containing 0.05mM Ca2+ in which the basal cell phenotype is normally maintained, stsp induced dose-dependent increases in keratin 1, epidermal and keratinocyte transglutaminases, SPR-1, loricrin, and profilaggrin mRNA. Based on nuclear run-on analysis, stsp-mediated marker expression was found to be due at least in part to increased transcription. Since protein kinase C (PKC) activation is required for keratinocyte differentiation, we tested whether stsp influenced this signaling pathway. Stsp induced the translocation of multiple PKC isoforms from the cytosol to membrane and/or cytoskeletal fractions, inducing isozyme downregulation within 24h. Moreover, AP-1 DNA binding activity was elevated in stsp-treated keratinocytes, consistent with the notion that this agent influences keratinocyte-specific gene expression via the PKC pathway. Stsp-mediated marker expression was inhibited by the PKC inhibitor GF 109203X. In cells pre-treated with bryostatin 1 to selectively down-modulate specific PKC isoforms, stsp-induced loricrin, filaggrin, and SPR-1 expression was suppressed when PKC α, ϵ, and/or δ were downregulated, suggesting that these isozymes may be necessary for marker expression in response to this agent. Thus, in addition to its effects on cornified envelope assembly, stsp induces a coordinate program of differentiation-specific keratinocyte gene expression that is mediated at least in part by the PKC signaling pathway

    Lysis inhibition (LIN) is a known feature of the T-even family of bacteriophages. Despite its historical role in the development of modern molecular genetics, many aspects of this phenomenon remain mostly unexplained. The key element of LIN is an interaction between two phage-encoded proteins, the T holin and the RI antiholin. This interaction is stabilized by RIII. In this report, we demonstrate the results of genetic experiments which suggest a synergistic action of two accessory proteins of bacteriophage T4, RI.-1, and RI.1 with RIII in the regulation of LIN

    24-h sheltering behaviour of individually kept horses during Swedish summer weather

    Provision of shelter for horses kept on summer pasture is rarely considered in welfare guidelines, perhaps because the benefits of shelter in warm conditions are poorly documented scientifically. For cattle, shade is a valued resource during summer and can mitigate the adverse effects of warm weather on well-being and performance. We found in a previous study that horses utilized shelters frequently in summer. A shelter with a roof and closed on three sides (shelter A) was preferred and can reduce insect pressure whereas a shelter with roof and open on three sides was not utilized. However, shelter A restricts the all-round view of a horse, which may be important for horses as flight animals. Therefore, we studied whether a shelter with roof, where only the upper half of the rear wall was closed (shelter B), would be utilized while maintaining insect protection properties and satisfying the horses’ sense for security. A third shelter was offered with walls but no roof (shelter C) to evaluate whether the roof itself is an important feature from the horse’s perspective. Eight Warmblood horses were tested each for 2 days, kept individually for 24 h in two paddocks with access to shelters A and B, or shelters A and C, respectively. Shelter use was recorded continuously during the night (1800–2400 h, 0200–0600 h) and the following day (0900–1600 h), and insect defensive behaviour (e.g., tail swish) in instantaneous scan samples at 5-min intervals during daytime

    A Nd Isotopic Study of Southern Sourced Waters and Indonesian Throughflow at Intermediate Depths in the Cenozoic Indian Ocean

    We present Nd isotopic data for fossil fish teeth recovered from the past 40 m.y. at ODP Site 757, currentlylocated at 1650m water depth on the Ninetyeast Ridge in the Indian Ocean. Although Site 757 sits in a regionstrongly influenced by weathering inputs from the Himalayas and volcanic inputs from the Indonesian arc, the pattern of Nd isotopic variations does not appear to respond to these potential sources of Nd. Instead, secular variations correlate to changes in the composition of intermediate to deep water masses bathing thesite and circulation patterns in the Indian Ocean. From ~40 to 10 Ma, eNd values and the pattern of change at Site 757 closely match those of ODP Site 1090, a deep water site in the Atlantic sector of the Southern Ocean. Comparison to data from several Fe-Mn crusts in the Indian Ocean suggests that intermediate to deep waterflow paths were similar to the modern distribution of Circumpolar DeepWater. At approximately 10 Ma, Ndisotopic values increase in a step function by 2 eNd units, suggesting that plate motions had carried Site 757into a region influenced by Indonesian Throughflow. Estimates of the vertical and horizontal position of thissite at 10 Ma imply that Indonesian Throughflow extended as far south as ~20S and to a depth of ~1500 m. From 10 to 0 Ma, Nd isotopic variations at Site 757 appear to record variations in Indonesian Throughflow. From 10 to 5.5 Ma, values at Site 757 overlap with those from crusts located in the southwest Pacific,indicating extensive flow through the Indonesian Seaway. From 5.5 to 3.4 Ma, eNd values become lessradiogenic at Site 757 and more radiogenic in the southwest Pacific, suggesting increasing closure of theseaway and concomitant rerouting of equatorial Pacific waters. Beginning at 3.4 Ma, eNd values becomemore radiogenic again at Site 757, which may be attributed to enhanced opening of the seaway or to a changein the source of Throughflow waters from a southern to a northern Pacific region

    Suppression of AP1 Transcription Factor Function in Keratinocyte Suppresses Differentiation

    Our previous study shows that inhibiting activator protein one (AP1) transcription factor function in murine epidermis, using dominant-negative c-jun (TAM67), increases cell proliferation and delays differentiation. To understand the mechanism of action, we compare TAM67 impact in mouse epidermis and in cultured normal human keratinocytes. We show that TAM67 localizes in the nucleus where it forms TAM67 homodimers that competitively interact with AP1 transcription factor DNA binding sites to reduce endogenous jun and fos factor binding. Involucrin is a marker of keratinocyte differentiation that is expressed in the suprabasal epidermis and this expression requires AP1 factor interaction at the AP1-5 site in the promoter. TAM67 interacts competitively at this site to reduce involucrin expression. TAM67 also reduces endogenous c-jun, junB and junD mRNA and protein level. Studies with c-jun promoter suggest that this is due to reduced transcription of the c-jun gene. We propose that TAM67 suppresses keratinocyte differentiation by interfering with endogenous AP1 factor binding to regulator elements in differentiation-associated target genes, and by reducing endogenous c-jun factor expression

    Evidence for intense REE scavenging at cold seeps from the Niger Delta margin

    International audienceFor many trace elements, continental margins are the location of intense exchange processes between sediment and seawater, which control their distribution in the water column, but have yet to be fully understood. In this study, we have investigated the impact of fluid seepage at cold seeps on the marine cycle of neodymium. We determined dissolved and total dissolvable (TD) concentrations for REE and well-established tracers of fluid seepage (CH4, TDFe, TDMn), and Nd isotopic compositions in seawater samples collected above cold seeps and a reference site (i.e. away from any fluid venting area) from the Niger Delta margin. We also analyzed cold seep authigenic phases and various core-top sediment fractions (pore water, detrital component, easily leachable phases, uncleaned foraminifera) recovered near the hydrocast stations. Methane, TDFe and TDMn concentrations clearly indicate active fluid venting at the studied seeps, with plumes rising up to about 100 m above the seafloor. Depth profiles show pronounced REE enrichments in the non-filtered samples (TD concentrations) within plumes, whereas filtered samples (dissolved concentrations) exhibit slight REE depletion in plumes relative to the overlying water column and display typical seawater REE patterns. These results suggest that the net flux of REE emitted into seawater at cold seeps is controlled by the presence of particulate phases, most probably Fe-Mn oxyhydroxides associated to resuspended sediments. At the reference site, however, our data reveal significant enrichment for dissolved REE in bottom waters, that clearly relates to diffusive benthic fluxes from surface sediments. Neodymium isotopic ratios measured in the water column range from εNd ~−15.7 to − 10.4. Evidence that the εNd values for Antarctic Intermediate waters (AAIW) differed from those reported for the same water mass at open ocean settings shows that sediment/water interactions take place in the Gulf of Guinea. At each site, however, the bottom water εNd signature generally differs from that for cold seep minerals, easily leachable sediment phases, and detrital fractions from local sediments, ruling out the possibility that seepage of methane-rich fluids and sediment dissolution act as a substantial source of dissolved Nd to seawater in the Gulf of Guinea. Taken together, our data hence suggest that co-precipitation of Fe-Mn oxyhydroxide phases in sub-surface sediments leads to quantitative scavenging of dissolved REE at cold seeps, preventing their emission into bottom waters. Most probably, it is likely that diffusion from suboxic surface sediments dominates the exchange processes affecting the marine Nd cycle at the Niger Delta margin

    Disruption of Spectrin-Like Cytoskeleton in Differentiating Keratinocytes by PKCδ Activation Is Associated with Phosphorylated Adducin

    Spectrin is a central component of the cytoskeletal protein network in a variety of erythroid and non-erythroid cells. In keratinocytes, this protein has been shown to be pericytoplasmic and plasma membrane associated, but its characteristics and function have not been established in these cells. Here we demonstrate that spectrin increases dramatically in amount and is assembled into the cytoskeleton during differentiation in mouse and human keratinocytes. The spectrin-like cytoskeleton was predominantly organized in the granular and cornified layers of the epidermis and disrupted by actin filament inhibitors, but not by anti-mitotic drugs. When the cytoskeleton was disrupted PKCδ was activated by phosphorylation on Thr505. Specific inhibition of PKCδ(Thr505) activation with rottlerin prevented disruption of the spectrin-like cytoskeleton and the associated morphological changes that accompany differentiation. Rottlerin also inhibited specific phosphorylation of the PKCδ substrate adducin, a cytoskeletal protein. Furthermore, knock-down of endogenous adducin affected not only expression of adducin, but also spectrin and PKCδ, and severely disrupted organization of the spectrin-like cytoskeleton and cytoskeletal distribution of both adducin and PKCδ. These results demonstrate that organization of a spectrin-like cytoskeleton is associated with keratinocytes differentiation, and disruption of this cytoskeleton is mediated by either PKCδ(Thr505) phosphorylation associated with phosphorylated adducin or due to reduction of endogenous adducin, which normally connects and stabilizes the spectrin-actin complex
