    Tensile behavior of an austenitic stainless steel subjected to multidirectional forging

    The mechanical behavior of a chromium -nickel austenitic stainless steel with submicrocrystalline structures produced by multidirectional forging (MDF) to a total strain of ∼ 4 at temperatures of 700 and 600° C was studied. This processing resulted in the formation of uniform ultrafine grained structure with an average crystallite size of 360 and 300 nm, respectively, and high dislocation density. The tensile tests were carried out in a wide temperature range 20 -650° C. At ambient temperature, the yield stress (YS) comprised 900 MPa and 730 MPa in the samples subjected to MDF at 600 and 700° C, respectively. It should be noted that this strength was achieved along with elongations of 16% and 22% in the samples subjected to MDF at 600 and 700° C. The YS decreased and elongati on-to-failure tends to increase with increasing test temperature and approaching 235 MPa and 51%, respectively, at 650° C.     Association of the EPAS1 gene G/A polymorphism with successful performance in a group of Russian wrestlers

    A large number of studies showed that the gene EPAS1 may serve as a possible predictor of success in sports because of its influence on the processes of oxygen transportation and consumption. However, data concerning the impact of EPAS1 polymorphisms on sports achievements in the modern research literature are very scarce and contradictory. The aim of the present paper was to study genetic selection in the polymorphic system of the EPAS1 gene (rs1867785) in a group of male sambo practitioners. 312 Russian males from 18 to 30 years of age were studied. Of them, 220 were professional athletes and 92 were non-athletes, who served as the control group. The genotype of a single nucleotide G/A polymorphic system of the EPAS1 gene was determined for each participant of the study. Analysis of genotype frequencies revealed statistically significant differences between the two groups. An increase of АА and AG genotype frequencies was revealed in the group of athletes (χ2 = 8.68, p = 0.01). Thus, for sambo practitioners, who reached high levels, the presence of the minor А-allele in the genotypes was typical. The odd ratio (OR) calculated for this group was 1.800 (95 % CI 1.227–2.641), demonstrating that the carriers of the А-allele of the EPAS1 gene had some advantages over the carriers of the G-allele. OR for the highest-rank wrestlers was even higher, 1.990 (95 % CI 1.195–3.313). These results suggest directed genetic selection in the А-allele carriers of the EPAS1 gene among sambo practitioners

    Objective: to study the genetic selection of the EPAS1 gene (G/А, rsl867785) in a group of athletes involved in Greco-Roman wrestling with different levels of sportsmanship. Materials and methods: the genetic examination of 79 male athletes (from 7 to 25 years old) involved in Greco-Roman wrestling and 92 controls (from 7 to 30 years old) was performed. Results: the frequency of A-allele associated with better adaptation to hypoxic conditions was higher in the group of wrestlers (EPAS1*GG - 27.8% EPAS1*AG - 67.1% and EPAS1*AA - 5.1% against EPAS1*GG - 56.5% EPAS1*AG - 35.9% and EPAS1 *AA - 7.6 %. x2 = 16.7 p = 0.0002). The frequency of A-allele carriers increased with increasing level of sportsmanship of studied athletes. The increase in the frequency of minor A-allele in the subgroup of Greco-Roman wrestlers testifies to their advantage in comparison with the carriers of G-allele.     Identification of cytoplasm types in accessions of the family Brassicaceae (Brassicaceae Burnett) with DNA markers

    Identification of cytoplasm types with the use of molecular markers enables to simplify the parent line pair selection for hybrid development based on CMS. Forty accessions of the family Brassicaceae (Brassicaceae Burnett) were analyzed with 22 pairs of PCR primers taken from previous research. Seven types of cytoplasm (Ogura, Ogu-NWSUAF, nap, pol, cam, rad, ole) were observed among breeding accessions using standard and multiplex PCR technique. The sequence of PCR products that differentiated different types of sterile cytoplasm confirmed the presence of three genes, orf138, orf222, and orf224, in mitochondrial DNA associated with exhibition of CMS. The accession of Nappa cabbage (Brassica rapa Pekinensis Group) had two genes, orf138 and orf222, which corresponded to cytoplasm Ogu-NWSUAF. All accessions carrying Ogura cytoplasm had the 417 bp fragment, which was 100 % identical to the mitochondrial orf138 gene sequence corres- pon­ding to Type А (formerly nine sequence Types, from A to I, had been identified). An exception was the accession of white head cabbage Tekila F1 with a new allelic variant of orf138 gene, which combined the 39-bp deletion typical of the Type F orf138 sequence and two nonsynonymous substitutions, A→T and G→A, at positions 95 and 99, respectively

    Methylotrophic yeast Pichia pastoris has proved to be especially useful for production of various heterologous proteins. In biotechnology it is very important to maintain the balance between high levels of heterologous gene expression and cell viability. Decisive understanding of gene regulation mechanisms is essential for reaching this goal. In this study, we investigated the effect of different nitrogen sources and phosphate concentration in media on methanol utilization. It was shown that expression levels of main genes, which are involved in methanol utilization (MUT genes) and in functioning of peroxisomes (PEX genes), are maximal when ammonium sulphate is used as a nitrogen source. Expression of these genes is decreased in media with poor nitrogen sources, such as proline. Addition of rapamycin to the media completely removed repression of AOX1 promoter in media with proline, which allows proposing that Tor-kinase is involved in establishing of nitrogen regulation of this gene. It was also shown that MUT genes expression levels get higher, when the phosphate concentration in media is increased

    The biotechnological methods enable to produce the ‘pure’ lines for the short period of time that intensify the breeding program. Particularly, the anther culture is widely used to obtain double haploid lines (DH lines) in plant breeding. The technology advantage is to fast up essentially the selection of homozygous lines with different morphological traits and high level of uniformity. Contemporary breeding programs are directed at F1 hybrid development; therefore DH lines are in need to serve as the parental form for heterotic hybrid production. Nevertheless, previous data has shown that DH plants have been noticed to produce few seeds on them. To introduce the lines into breeding program for hybrid production and to multiply them, the seed formation of DH lines is worth being studied. The result of assessment of seed generation of DH lines of Broccoli that have been produced from ‘Tonus’ and convariety ‘N1’ is presented in the article There is much difference in morphological traits and both ability to recognize their own pollen and quantity of seeds in the pod among plants produced. Nine DH lines of early maturity were obtained. The differences in seed formation among these lines and between generations were revealed. It was shown the influence of genotype on seed formation in DH lines. It was also noticed the much seed formation in case of geitonogamic pollination in DH lines obtained from cultivar ‘Tonus’.
