    Impact of Parental Substance Misuse on Attachment in Young Adults: A Qualitative Approach

    This study examined the impact of parental substance misuse on young adult development and relationships by interviewing young adults about their experience being raised by a parent who misused substances. A qualitative design based on constructivist grounded theory and informed by constructs from attachment theory was used. Participants consisted of 10 young adults, ages 18-26, who endorsed being raised by a parent who misused substances. The interview questions developed for the study were informed by a measure of adult attachment. Analysis of the data included identification of emergent categories/themes as well as a priori constructs from attachment theory (safe haven, secure base, reflective functioning, coregulation). Based on the analysis, the following emergent categories/themes were identified and described: behavior of the parent with substance misuse, feelings/experience of the offspring, acknowledgement of substance misuse, impact on the parent-child relationship, impact on relationships with others, and impact on mental health and identity. A priori attachment constructs were evident in the narratives and provided a useful frame for understanding the impact of parental substance misuse. This study demonstrated how constructs from attachment theory can be applied to better understand the relationship between a parent’s behavior and the impact on attachment security in the child when substance misuse is occurring. Parental absence, lack of attunement to the child’s needs, and inconsistent behavior may contribute to offspring feeling rejected, unknown, and confused, which may impact their sense of attachment security, feelings of worth, and perception of the reliability of others

    Global Acetylation Dynamics in the Heat Shock Response of Saccharomyces cerevisiae

    All organisms face a constant barrage of environmental stresses. Single-cell organisms such as Saccharomyces cerevisiae, or common Baker’s yeast, must rely solely on cellular responses in order to survive. This response must occur in a rapid and highly coordinated manner to quickly inhibit all unnecessary processes and shuttle all available resources to those necessary for survival. One method that cells utilize for rapid protein regulation is the use of post-translational modifications. Enzymes within the cell add or remove a variety of chemical modifications, thus altering the local chemical environment of a protein. This creates a conformational change in the protein that can increase, decrease, or completely change the activity of the protein, as well as target them for relocation or degradation. Examples of common post-translational modifications include phosphorylation, ubiquitination, and the focus of this dissertation, acetylation. That protein acetylation occurs has been known for decades, but it is only recently that advances in technology such as high-resolution mass spectrometry and immunoprecipitation have led to the recognition of thousands of acetylated proteins across all domains of life. The roles and regulation of this modification, however, are still widely unknown. One approach to better understand possible roles for acetylation is to look at its dynamics in response to environmental stress. In this dissertation, I examine global changes in protein acetylation in the response of Saccharomyces cerevisiae to a mild heat shock and the potential mechanisms regulating these changes. Following an introductory literature review, this dissertation will cover the results of a large time-scale profiling of acetylome dynamics in response to heat shock. Proteins identified in this experiment are enriched for many cellular processes, suggesting that acetylation may play a much wider regulatory role than previously believed. These proteins are also enriched for interactions with many lysine acetyltransferases and deacetylases, suggesting that the regulation of this modification is complex. The next chapter will then discuss possible mechanisms regulating this response. This includes the investigation into concentrations of metabolites known to affect acetylation and deacetylation, lysine acetyltransferase and deacetylase complex remodeling, and localization changes for those complexes within the cell

    Omega 3 fatty acids reduce myeloid progenitor cell frequency in the bone marrow of mice and promote progenitor cell differentiation

    <p>Abstract</p> <p>Background</p> <p>Omega 3 fatty acids have been found to inhibit proliferation, induce apoptosis, and promote differentiation in various cell types. The processes of cell survival, expansion, and differentiation are of key importance in the regulation of hematopoiesis. We investigated the role of omega 3 fatty acids in controlling the frequency of various myeloid progenitor cells in the bone marrow of mice. Increased progenitor cell frequency and blocked differentiation are characteristics of hematopoietic disorders of the myeloid lineage, such as myeloproliferative diseases and myeloid leukemias.</p> <p>Results</p> <p>We found that increasing the proportion of omega 3 fatty acids relative to the proportion of omega 6 fatty acids in the diet caused increased differentiation and reduced the frequency of myeloid progenitor cells in the bone marrow of mice. Furthermore, this had no adverse effect on peripheral white blood cell counts.</p> <p>Conclusion</p> <p>Our results indicate that omega 3 fatty acids impact hematopoietic differentiation by reducing myeloid progenitor cell frequency in the bone marrow and promoting progenitor cell differentiation. Further exploration of this discovery could lead to the use of omega 3 fatty acids as a therapeutic option for patients that have various disorders of hematopoiesis.</p

    Suppression of Implanted MDA-MB 231 Human Breast Cancer Growth in Nude Mice by Dietary Walnut

    Walnuts contain components that may slow cancer growth including omega 3 fatty acids, phytosterols, polyphenols, carotenoids, and melatonin. A pilot study was performed to determine whether consumption of walnuts could affect growth of MDA-MB 231 human breast cancers implanted into nude mice. Tumor cells were injected into nude mice that were consuming an AIN-76A diet slightly modified to contain 10% corn oil. After the tumors reached 3 to 5 mm diameter, the diet of one group of mice was changed to include ground walnuts, equivalent to 56 g (2 oz) per day in humans. The tumor growth rate from Day 10, when tumor sizes began to diverge, until the end of the study of the group that consumed walnuts (2.9 ± 1.1 mm3/day; mean ± standard error of the mean) was significantly less (P \u3e 0.05, t-test of the growth rates) than that of the group that did not consume walnuts (14.6 ± 1.3 mm 3 /day). The eicosapentaenoic and docosahexaenoic acid fractions of the livers of the group that consumed walnuts were significantly higher than that of the group that did not consume walnuts. Tumor cell proliferation was decreased, but apoptosis was not altered due to walnut consumption. Further work is merited to investigate applications to cancer in humans

    : We examine again the problem of the damping rate of a moving heavy fermion in a hot plasma within the resummed perturbative theory of Pisarski and Braaten. The ansatz for its evaluation which relates it to the imaginary part of the fermion propagator pole in the framework of a self-consistent approach is critically analyzed. As already pointed out by various authors, the only way to define the rate is through additional implementation of magnetic screening. We show in detail how the ansatz works in this case and where we disagree with other authors. We conclude that the self-consistent approach is not satisfactory.Comment: 17 page

Therefore, this research aims to design book of batik tulis Jarak Arum ex-Dolly with watercolor techniques in order to increase brand awareness. The design of the book batik tulis Jarak Arum ex-Dolly using qualitative data collection methods, namely by interviews, observation, literature study, existing studies and creative brief is very important to define the concept design study.     The role of source cloud spatial coherence in a Mach-Zehnder type atom interferometer is experimentally investigated. The visibility and contrast of a Bose-Einstein condensate (BEC) and three thermal sources with varying spatial coherence are compared as a function of interferometer time. At short times, the fringe visibility of a BEC source approaches 100 % nearly independent of pi pulse efficiency, while thermal sources have fringe visibilities limited to the mirror efficiency. More importantly for precision measurement systems, the BEC source maintains interference at interferometer times significantly beyond the thermal source

    80hk Momentum Separation with Bloch Oscillations in an Optically Guided Atom Interferometer

    We demonstrate phase sensitivity in a horizontally guided, acceleration-sensitive atom interferometer with a momentum separation of 80hk between its arms. A fringe visibility of 7% is observed. Our coherent pulse sequence accelerates the cold cloud in an optical waveguide, an inherently scalable route to large momentum separation and high sensitivity. We maintain coherence at high momentum separation due to both the transverse confinement provided by the guide, and our use of optical delta-kick cooling on our cold-atom cloud. We also construct a horizontal interferometric gradiometer to measure the longitudinal curvature of our optical waveguide.Comment: 6 pages, 6 figure
