Evaluation of a resistance training program for adults with or at risk of developing diabetes: an effectiveness study in a community setting
Background: To examine the effects of a community-based resistance training program (Lift for Life) on waist circumference and functional measures in adults with or at risk of developing type 2 diabetes.Methods: Lift for Life is a research-to-practice initiative designed to disseminate an evidence-based resistance training program for adults with or at risk of developing type 2 diabetes to existing health and fitness facilities in the Australian community. A retrospective assessment was undertaken on 86 participants who had accessed the program within 4 active providers in Melbourne, Australia. The primary goal of this longitudinal study was to assess the effectiveness of a community-based resistance training program, thereby precluding a randomized, controlled study design. Waist circumference, lower body (chair sit-to-stand) and upper body (arm curl test) strength, and agility (timed up-and-go) measures were collected at baseline and repeated at 2 months (n = 86) and again at 6 months (n = 32).Results: Relative to baseline, there was a significant decrease in mean waist circumference (-1.9 cm, 95% CI: -2.8 to -1.0) and the timed agility test (-0.8 secs, 95% CI: -1.0 to -0.6); and significant increases in lower body (number of repetitions: 2.2, 95% CI: 1.4-3.0) and upper body (number of repetitions: 3.8, 95% CI: 3.0-4.6) strength at the completion of 8 weeks. Significant differences remained at the 16 week assessment. Pooled time series regression analyses adjusted for age and sex in the 32 participants who had complete measures at baseline and 24-week follow-up revealed significant time effects for waist circumference and functional measures, with the greatest change from baseline observed at the 24-week assessment.Conclusions: These findings indicate that an evidence-based resistance training program administered in the community setting for those with or at risk of developing type 2 diabetes, can lead to favorable health benefits, including reductions in central obesity and improved physical function
Joint associations of multiple leisure-time sedentary behaviours and physical activity with obesity in Australian adults
BackgroundTelevision viewing and physical inactivity are independently associated with risk of obesity. However, how the combination of multiple leisure-time sedentary behaviours (LTSB) and physical activity (LTPA) may contribute to the risk of obesity is not well understood. We examined the joint associations of multiple sedentary behaviours and physical activity with the odds of being overweight or obese.MethodsA mail survey collected the following data from adults living in Adelaide, Australia (n = 2210): self-reported height, weight, six LTSB, LTPA and sociodemographic variables. Participants were categorised into four groups according to their level of LTSB (dichotomised into low and high levels around the median) and LTPA (sufficient: ≥ 2.5 hr/wk; insufficient: < 2.5 hr/wk). Logistic regression analysis examined the odds of being overweight or obese (body mass index ≥ 25 kg/m2) by the combined categories.ResultsThe odds of being overweight or obese relative to the reference category (low sedentary behaviour time and sufficient physical activity) were: 1.54 (95% confidence interval [CI]: 1.20–1.98) for the combination of low sedentary behaviour time and insufficient physical activity; 1.55 (95% CI: 1.20–2.02) for the combination of high sedentary behaviour time and sufficient physical activity; and 2.26 (95% CI: 1.75–2.92) for the combination of high sedentary behaviour time and insufficient physical activity.ConclusionThose who spent more time in sedentary behaviours (but were sufficiently physically active) and those who were insufficiently active (but spent less time in sedentary behaviour) had a similar risk of being overweight or obese. Reducing leisure-time sedentary behaviours may be as important as increasing leisure-time physical activity as a strategy to fight against obesity in adults.<br /
Growth and Feeding Studies on the Algal Feeding Stage of a Pfiesteria-like Dinoflagellate
The dinoflagellate Cryptoperidiniopsis sp. was isolated from sediment samples taken from Virginia estuaries, and established in culture for subsequent growth and feeding studies. The maximum abundance, or yield, of Cryptoperidiniopsis is exponentially related to the concentration of algal prey and is saturated at about 4.00 X 105 mL-1. Salinity from 10-20 ppt and temperature between 15-25 C have no effect on the yield of this form of Cryptoperidiniopsis. Light intensity has a secondary effect in that the algal prey reproduces more quickly in higher light as they are being grazed. Growth rates of Cryptoperidiniopsis were highest with a cryptophyte, Cryptomonas, as food, but growth was also demonstrated utilizing both diatoms and chlorophytes. Cryptoperidiniopsis sp. is similar to Pfiesteria in that it feeds myzocytotically with a peduncle, is similar in size and shape, has a complex life cycle, and is distinguished only by plates hidden under membranes

Physical activity, television viewing time and 12 year changes in waist circumference
PURPOSE: Both moderate-to-vigorous physical activity (MVPA) and sedentary behavior can be associated with adult adiposity. Much of the relevant evidence is from cross-sectional studies or from prospective studies with relevant exposure measures at a single time point prior to weight gain or incident obesity. This study examined whether changes in MVPA and television (TV) viewing time are associated with subsequent changes in waist circumference, using data from three separate observation points in a large population-based prospective study of Australian adults. METHODS: Data were obtained from the Australian Diabetes, Obesity and Lifestyle study collected in 1999-2000 (baseline), 2004-05 (Wave 2), and 2011-12 (Wave 3). The study sample consisted of adults aged 25 to 74 years at baseline who also attended site measurement at three time points (n=3261). Multilevel linear regression analysis examined associations of initial five-year changes in MVPA and TV viewing time (from baseline to Wave 2) with 12-year change in waist circumference (from baseline to Wave 3), adjusting for well-known confounders. RESULTS: As categorical predictors, increases in MVPA significantly attenuated increases in waist circumference (p for trend< 0.001). TV viewing time change was not significantly associated with changes in waist circumference (p for trend =0.06). Combined categories of MVPA and TV viewing time changes were predictive of waist circumference increases; compared to those who increased MVPA and reduced TV viewing time, those who reduced MVPA and increased TV viewing time had a 2cm greater increase in waist circumference (p=0.001). CONCLUSION: Decreasing MVPA emerged as a significant predictor of increases in waist circumference. Increasing TV viewing time was also influential, but its impact was much weaker than MVPA
Workplace sitting and height-adjustable workstations: a randomized controlled trial
Background Desk-based office employees sit for most of their working day. To address excessive sitting as a newly identified health risk, best practice frameworks suggest a multi-component approach. However, these approaches are resource intensive and knowledge about their impact is limited. Purpose To compare the efficacy of a multi-component intervention to reduce workplace sitting time, to a height-adjustable workstations-only intervention, and to a comparison group (usual practice). Design Three-arm quasi-randomized controlled trial in three separate administrative units of the University of Queensland, Brisbane, Australia. Data were collected between January and June 2012 and analyzed the same year. Setting/participants Desk-based office workers aged 20-65 (multi-component intervention, n=16; workstations-only, n=14; comparison, n=14). Intervention The multi-component intervention comprised installation of height-adjustable workstations and organizational-level (management consultation, staff education, manager e-mails to staff) and individual-level (face-to-face coaching, telephone support) elements. Main outcome measures Workplace sitting time (minutes/8-hour workday) assessed objectively via activPAL3 devices worn for 7 days at baseline and 3 months (end-of-intervention) . Results At baseline, the mean proportion of workplace sitting time was approximately 77% across all groups (multi-component group 366 minutes/8 hours [SD=49]; workstations-only group 373 minutes/8 hours [SD=36], comparison 365 minutes/8 hours [SD=54]). Does light-intensity physical activity moderate the relationship between sitting time and adiposity markers in adolescents?
Background
While the relationship between sedentary time and adiposity markers may be independent of moderate-to-vigorous intensity physical activity (MVPA) among adolescents, little is known about the role of light-intensity physical activity (LIPA) in this relationship. The aim of this cross-sectional study was to examine whether device-measured LIPA and MVPA moderate the associations between objectively measured sitting time and adiposity markers (body mass index (BMI)) and waist circumference (WC)) among adolescents.
Methods
This study included accelerometer and inclinometer data obtained from 219 adolescents (age = 14.9 ± 1.6 years, mean ± SD), collected during 2014 and 2015 in Melbourne, Australia. ActiGraph GT3X accelerometers were used to determine time spent in total-LIPA (101 counts/min to 3.99 metabolic equivalents (METs)) was dichotomized into low-LIPA (101–799 counts/min) and high LIPA (800 counts/min to 3.99 METs), and MVPA (≥ 4 METs). The average time spent sitting was obtained from activPAL inclinometers. Anthropometric measures were assessed by trained staff. Interactions between sitting and total-LIPA, low-LIPA, high-LIPA, and MVPA on BMI z-score (zBMI) and WC z-score (zWC), respectively, were examined using linear regression, adjusting for age and sex; and moderation by total-LIPA, low-LIPA, high-LIPA, and MVPA were examined by adding interaction terms. Significant interaction effects were probed by comparing associations at the mean and at 1 SD below and above the mean.
Results
Total-LIPA significantly moderated the association between sitting time and zBMI, and low-LIPA significantly moderated the association between sitting time and zBMI and zWC. No other associations were found for total-LIPA, high-LIPA, or MVPA. Specifically, at high levels of total-LIPA (+1 SD), there is a negative association between sitting time and zBMI. In addition, at high levels of low-LIPA (+1 SD), there is a negative association between sitting time and zBMI and zWC.
Conclusion
Associations between sitting and adiposity depended on time spent in total-LIPA and low-LIPA, but not high-LIPA or MVPA. Results suggest that increasing time spent in LIPA may provide protection from the deleterious effects of sitting on adiposity markers among adolescents. Experimental evidence is needed to support these conclusions
Computational Modeling of Interactions between Multiple Myeloma and the Bone Microenvironment
Multiple Myeloma (MM) is a B-cell malignancy that is characterized by osteolytic bone lesions. It has been postulated that positive feedback loops in the interactions between MM cells and the bone microenvironment form reinforcing ‘vicious cycles’, resulting in more bone resorption and MM cell population growth in the bone microenvironment. Despite many identified MM-bone interactions, the combined effect of these interactions and their relative importance are unknown. In this paper, we develop a computational model of MM-bone interactions and clarify whether the intercellular signaling mechanisms implemented in this model appropriately drive MM disease progression. This new computational model is based on the previous bone remodeling model of Pivonka et al. [1], and explicitly considers IL-6 and MM-BMSC (bone marrow stromal cell) adhesion related pathways, leading to formation of two positive feedback cycles in this model. The progression of MM disease is simulated numerically, from normal bone physiology to a well established MM disease state. Our simulations are consistent with known behaviors and data reported for both normal bone physiology and for MM disease. The model results suggest that the two positive feedback cycles identified for this model are sufficient to jointly drive the MM disease progression. Furthermore, quantitative analysis performed on the two positive feedback cycles clarifies the relative importance of the two positive feedback cycles, and identifies the dominant processes that govern the behavior of the two positive feedback cycles. Using our proposed quantitative criteria, we identify which of the positive feedback cycles in this model may be considered to be ‘vicious cycles’. Finally, key points at which to block the positive feedback cycles in MM-bone interactions are identified, suggesting potential drug targets
Sitting Less and Moving More: Implications for Hypertension.
Through a hypertension lens, this review focuses on the serious health implications and potential counter-measures for the high volumes of uninterrupted sitting that characterises modern lifestyles. Reducing and breaking up sitting may represent a viable clinical and population health strategy to reduce blood pressure and related cardiovascular risk factors, which in contrast to moderate-vigorous physical activity, may have significant potential for widespread uptake. This review synthesises findings on the specific relationships of sedentary behaviour with BP - which primarily are from observational and acute experimental studies - including a discussion on the relevant cardiovascular mechanisms. It also considers what will need to be better understood as a basis for evidence-based recommendations on sedentary time in the context of BP control, and identify evidence gaps for future research
Perceived neighbourhood environmental attributes and prospective changes in TV viewing time among older Australian adults
There has been a growing interest in environmental initiatives to reduce sedentary behaviour.
