Expression of Axolotl DAZL RNA, a Marker of Germ Plasm: Widespread Maternal RNA and Onset of Expression in Germ Cells Approaching the Gonad
AbstractHow germ cell specification occurs remains a fundamental question in embryogenesis. The embryos of several model organisms contain germ cell determinants (germ plasm) that segregate to germ cell precursors. In other animals, including mice, germ cells form in response to regulative mechanisms during development. To investigate germ cell determination in urodeles, where germ plasm has never been conclusively identified, we cloned a DAZ-like sequence from axolotls, Axdazl. Axdazl is homologous to Xdazl, a component of Xenopus germ plasm found in the vegetal pole of oocytes and eggs. Axdazl RNA is not localized in axolotl oocytes, and, furthermore, these oocytes do not contain the mitochondrial cloud that localizes Xdazl and other germ plasm components in Xenopus. Maternal Axdazl RNA is inherited in the animal cap and equatorial region of early embryos. At gastrula, neurula, and tailbud stages, Axdazl RNA is widely distributed. Axdazl first shows cell-specific expression in primordial germ cells (PGCs) approaching the gonad at stage 40, when nuage (germ plasm) appears in PGCs. These results suggest that, in axolotls, germ plasm components are insufficient to specify germ cells

Medical endoscopy is a growing industry, with a trend of chronic hand pain for gastroenterologists and flexible endoscope users. In order to address the lack of ergonomics in traditional gastrointestinal (GI) flexible endoscopes, the design team collaborated with a GI professional to design a more ergonomic endoscope interface, focusing on the more problematic controls. Two prototypes, as well as a mock-up were fabricated, which underwent verification and a user evaluation to assess the designâs benefits. As the number of endoscopies each year increase, the Ergoscope, offers an alternative to the traditional angulation controls and muchneeded relief for the issues that plague physiciansâespecially those with smaller handsâtoday

Resistance patterns are compared to those reported by the National Antimicrobial Resistance Monitoring System. Canada Geese may be vectors of antimicrobial resistance and resistance genes in agricultural environments
Superstructure based on β-CD self-assembly induced by a small guest molecule
The size, shape and surface chemistry of nanoparticles play an important role in cellular interaction. Thus, the main objective of the present study was the determination of the β-cyclodextrin (β-CD) self-assembly thermodynamic parameters and its structure, aiming to use these assemblies as a possible controlled drug release system. Light scattering measurements led us to obtain the β-CD's critical aggregation concentration (cac) values, and consequently the thermodynamic parameters of the β-CD spontaneous self-assembly in aqueous solution: Î[subscript agg]G[superscript o] = â16.31 kJ mol[superscript â1], Î[subscript agg]H[superscript o] = â26.48 kJ mol[superscript â1] and TÎ[subscript agg]S[superscript o] = â10.53 kJ mol[superscript â1] at 298.15 K. Size distribution of the self-assembled nanoparticles below and above cac was 1.5 nm and 60â120 nm, respectively. The number of β-CD molecules per cluster and the second virial coefficient were identified through Debye's plot and molecular dynamic simulations proposed the three-fold assembly for this system below cac. Ampicillin (AMP) was used as a drug model in order to investigate the key role of the guest molecule in the self-assembly process and the β-CD:AMP supramolecular system was studied in solution, aiming to determine the structure of the supramolecular aggregate. Results obtained in solution indicated that the β-CD's cac was not affected by adding AMP. Moreover, different complex stoichiometries were identified by nuclear magnetic resonance and isothermal titration calorimetry experiments.Brazil.
