    The Cytoplasmic Location of Chicken Mx Is Not the Determining Factor for Its Lack of Antiviral Activity

    Chicken Mx belongs to the Mx family of interferon-induced dynamin-like GTPases, which in some species possess potent antiviral properties. Conflicting data exist for the antiviral capability of chicken Mx. Reports of anti-influenza activity of alleles encoding an Asn631 polymorphism have not been supported by subsequent studies. The normal cytoplasmic localisation of chicken Mx may influence its antiviral capacity. Here we report further studies to determine the antiviral potential of chicken Mx against Newcastle disease virus (NDV), an economically important cytoplasmic RNA virus of chickens, and Thogoto virus, an orthomyxovirus known to be exquisitely sensitive to the cytoplasmic MxA protein from humans. We also report the consequences of re-locating chicken Mx to the nucleus.Chicken Mx was tested in virus infection assays using NDV. Neither the Asn631 nor Ser631 Mx alleles (when transfected into 293T cells) showed inhibition of virus-directed gene expression when the cells were subsequently infected with NDV. Human MxA however did show significant inhibition of NDV-directed gene expression. Chicken Mx failed to inhibit a Thogoto virus (THOV) minireplicon system in which the cytoplasmic human MxA protein showed potent and specific inhibition. Relocalisation of chicken Mx to the nucleus was achieved by inserting the Simian Virus 40 large T antigen nuclear localisation sequence (SV40 NLS) at the N-terminus of chicken Mx. Nuclear re-localised chicken Mx did not inhibit influenza (A/PR/8/34) gene expression during virus infection in cell culture or influenza polymerase activity in A/PR/8/34 or A/Turkey/50-92/91 minireplicon systems.The chicken Mx protein (Asn631) lacks inhibitory effects against THOV and NDV, and is unable to suppress influenza replication when artificially re-localised to the cell nucleus. Thus, the natural cytoplasmic localisation of the chicken Mx protein does not account for its lack of antiviral activity

    Prognostic impact of acute pulmonary triggers in patients with Takotsubo syndrome : new insights from the International Takotsubo Registry

This study aimed to investigate prevalence of acute pulmonary triggers in patients with TTS and their impact on outcomes. Methods and results: Patients with TTS were enrolled from the International Takotsubo Registry and screened for triggering factors and comorbidities. Patients were categorized into three groups (acute pulmonary trigger, chronic lung disease, and no lung disease) to compare clinical characteristics and outcomes. Of the 1670 included patients with TTS, 123 (7%) were identified with an acute pulmonary trigger, and 194 (12%) had a known history of chronic lung disease. The incidence of cardiogenic shock was highest in patients with an acute pulmonary trigger compared with those with chronic lung disease or without lung disease (17% vs. 10% vs. 9%, P = 0.017). In-hospital mortality was also higher in patients with an acute pulmonary trigger than in the other two groups, although not significantly (5.7% vs. 1.5% vs. 4.2%, P = 0.13). Survival analysis demonstrated that patients with an acute pulmonary trigger had the worst long-term outcome (P = 0.002). The presence of an acute pulmonary trigger was independently associated with worse long-term mortality (hazard ratio 2.12, 95% confidence interval 1.33-3.38; P = 0.002). Conclusions: The present study demonstrates that TTS is related to acute pulmonary triggers in 7% of all TTS patients, which accounts for 21% of patients with physical triggers. Japanese patients were more likely to experience cardiogenic shock during the acute phase (15.5 vs. 9.0%; p < 0.001) and had a higher in-hospital mortality (8.2 vs. 3.2%; p < 0.001). However, ethnicity itself did not appear to have an impact on in-hospital mortality. Machine learning approach revealed that the presence of physical stressors was the most important prognostic factor in both Japanese and European TTS patients. Conclusion: Differences in clinical characteristics and in-hospital outcomes between Japanese and European TTS patients exist. Ethnicity does not impact the outcome in TTS patients.     Worin unterscheidet sich eine rechte von einer linken Hand, trifft doch jede Eigenschaft der einen Hand auch auf die andere zu? Diese von Kant gestellte Frage und die von ihm gegebene Antwort waren immer wieder Anlass zu Kontroversen ĂŒber die Natur des Raumes. Einen klĂ€renden Beitrag lieferte der Mathematiker Reidemeister, dessen Aussagen in dieser Arbeit unter Verwendung eines in der Chemie wichtigen Begriffs interpretiert werden, nĂ€mlich der ChiralitĂ€t. Im Weiteren geht es um die Tragweite des aus dieser Deutung hervorgehenden PhĂ€nomens der DiachiralitĂ€t und um das Bilden von ChiralitĂ€tsklassen chiraler Objekte durch Orientierung. In einem zweiten Teil werden die besprochenen ChiralitĂ€tsaspekte auf konvexe Polytope angewendet, wobei u.a. ein Minimierungsverfahren und ein darauf basierender Algorithmus wesentlich sind

    Azimines. VIII. Contribution to the Preparation and Thermolysis of Alkyl (2Z)-2,3-Diisopropyl-azimine-1-carboxylates

    After improving the preparation of the azimines 9 (a = methyl, b = ethyl ester), we reinvestigated the mild thermolysis of 9 to 13 in CDCh: Unlike previously postulated, the azimines 9 first undergo a 1,4-H migration to the triazones 14 (step a), which suffer an elimination to yield the diazenes 16 (step b), which, in turn, are finally transformed by a 1,3-H migration to the carbazates 13 (step c). Step a is base catalyzed while step b is not (or less), so that 14, example of a relatively rare class of compounds, could be isolated after thermolysis of 9 in t-butylamine. During the thermolyses of pure 14 in CDCla, the diazenes 16 accumulated transiently (step b) before passing to 13. This accumulation of 16 was less (10%, as compared to 40%) when starting with 10% (as compared to 1%) solution of 14, which suggest that the conversion of 16 to 13 (step c) is catalyzed by 14. With pure 16, prepared in another way, step c occured in a 1% CDCh solution at 500C with ti" = - 21 hrs. Step c also proved to be catalyzed by silica
