    Background: From ancient times Siddha medicines have been proven to treat a wide range of communicable and noncommunicable diseases. In India, the number of people affected with diabetes is increasing drastically and it is considered as an epidemic. Purpose: To assess the anti-diabetic activity of the active compounds of the polyherbal formulation – Madhu Mukthi Kudineer Chooranum (MKC) by docking against potent enzymes involved in diabetes mellitus by in silico method. Methods: The major target enzymes Dipeptidyl peptidase, Glycogen synthase kinase - 3β, Alpha-glucosidase and Alpha-amylase were docked against the 10 active compounds present in the MKC using Auto Dock tools. Conclusion: The strong interactions between the target enzymes and the active compounds proved the anti-diabetic efficacy of the MKC formulation. Thus, the intake of MKC has potential benefits in the treatment of Type II Diabetes Mellitus
