    Impacts of Duck-Origin Parvovirus Infection on Cherry Valley Ducklings From the Perspective of Gut Microbiota

    Duck-origin goose parvovirus (D-GPV) is the causative agent of beak atrophy and dwarfism syndrome (BADS), characterized by growth retardation, skeletal dysplasia, and persistent diarrhea. However, the pathogenic mechanism of D-GPV remains undefined. Here, we first reported the gut microbiome diversity of D-GPV infected Cherry Valley ducks. In the investigation for the influence of D-GPV infection on gut microbiota through a period of infection, we found that D-GPV infection caused gut microbiota dysbiosis by reducing the prevalence of the dominant genera and decreasing microbial diversity. Furthermore, exfoliation of the intestinal epithelium, proliferation of lymphocytes, up-regulated mRNA expression of pro-inflammatory TNF-α, IL-1β, IL-6, IL-17A, and IL-22 and down-regulated mRNA expression of anti-inflammatory IL-10 and IL-4 occurred when D-GPV targeted in cecal epithelium. In addition, the content of short chain fatty acids (SCFAs) in cecal contents was significantly reduced after D-GPV infection. Importantly, the disorder of pro-inflammatory and anti-inflammatory cytokines was associated with the decrease of SCFAs-producing bacteria and the enrichment of opportunistic pathogens. Collectively, the decrease of SCFAs and the enrichment of pathogen-containing gut communities promoted intestinal inflammatory injury. These results may provide a new insight that target the gut microbiota to understand the progression of BADS disease and to research the pathogenic mechanism of D-GPV

    Methylprednisolone as Adjunct to Endovascular Thrombectomy for Large-Vessel Occlusion Stroke

    Importance It is uncertain whether intravenous methylprednisolone improves outcomes for patients with acute ischemic stroke due to large-vessel occlusion (LVO) undergoing endovascular thrombectomy. Objective To assess the efficacy and adverse events of adjunctive intravenous low-dose methylprednisolone to endovascular thrombectomy for acute ischemic stroke secondary to LVO. Design, Setting, and Participants This investigator-initiated, randomized, double-blind, placebo-controlled trial was implemented at 82 hospitals in China, enrolling 1680 patients with stroke and proximal intracranial LVO presenting within 24 hours of time last known to be well. Recruitment took place between February 9, 2022, and June 30, 2023, with a final follow-up on September 30, 2023.InterventionsEligible patients were randomly assigned to intravenous methylprednisolone (n = 839) at 2 mg/kg/d or placebo (n = 841) for 3 days adjunctive to endovascular thrombectomy. Main Outcomes and Measures The primary efficacy outcome was disability level at 90 days as measured by the overall distribution of the modified Rankin Scale scores (range, 0 [no symptoms] to 6 [death]). The primary safety outcomes included mortality at 90 days and the incidence of symptomatic intracranial hemorrhage within 48 hours. Results Among 1680 patients randomized (median age, 69 years; 727 female [43.3%]), 1673 (99.6%) completed the trial. The median 90-day modified Rankin Scale score was 3 (IQR, 1-5) in the methylprednisolone group vs 3 (IQR, 1-6) in the placebo group (adjusted generalized odds ratio for a lower level of disability, 1.10 [95% CI, 0.96-1.25]; P = .17). In the methylprednisolone group, there was a lower mortality rate (23.2% vs 28.5%; adjusted risk ratio, 0.84 [95% CI, 0.71-0.98]; P = .03) and a lower rate of symptomatic intracranial hemorrhage (8.6% vs 11.7%; adjusted risk ratio, 0.74 [95% CI, 0.55-0.99]; P = .04) compared with placebo. Conclusions and Relevance Among patients with acute ischemic stroke due to LVO undergoing endovascular thrombectomy, adjunctive methylprednisolone added to endovascular thrombectomy did not significantly improve the degree of overall disability.Trial RegistrationChiCTR.org.cn Identifier: ChiCTR210005172

    Elemental Mercury Adsorption by Cupric Chloride-Modified Mesoporous Carbon Aerogel

    Mesoporous carbon aerogels (MCA) synthesized via aqueous polymerization of resorcinol and formaldehyde were modified by copper chloride and applied for adsorption removal of elemental mercury (Hg0) at a low temperature. The sorbents were characterized by field emission scanning electron microscopy (FESEM), X-ray diffraction (XRD), nitrogen adsorption/desorption isotherms, and X-ray photoelectron spectroscopy (XPS) techniques. The results show that CuCl2-modified MCA exhibited good ability for Hg0 adsorption in the 40⁻160 °C temperature range, with Hg0 removal efficiency all above 95.0%. The Hg0 removal efficiency first increased and then decreased with the elevation of reaction temperature. It performed optimally at 80 °C with the highest Hg0 removal efficiency of 98.7%. XPS results indicate that covalent chlorines (C-Cl groups) play an important role in elemental mercury adsorption process. Hg0 is firstly captured in the form of oxidized mercury (Hg2+) and then reacts with C-Cl groups to form HgCl2

    Soy Isoflavones Improve the Spermatogenic Defects in Diet-Induced Obesity Rats through Nrf2/HO-1 Pathway

    Soy isoflavones (SIF) are biologically active compounds of non-steroidal and phenolic properties that are richly present in soybeans, which can reduce the body weight and blood lipids of obese animals. Recently, SIF have been reported to affect reproductive ability in obese male rats. However, the specific mechanism has not been well defined. The aim of the current study was to study the possible mechanisms for the effect of SIF administration on obesity induced spermatogenic defects. Obese rats model induced by high-fat diets were established and gavage treated with 0, 50,150 or 450 mg of SIF/kg body weight/day for 4 weeks. Here, our research shows that obesity resulted in spermatogenic degeneration, imbalance of reproductive hormone, testicular oxidative stress and germ cell apoptosis, whereas evidently recovery effects were observed at 150 and 450 mg/kg SIF. We also have discovered that 150 and 450 mg/kg SIF can activate Nrf2/HO-1 pathway in control of Bcl-2, BAX and cleaved caspase-3 expression with implications in antioxidant protection. Our study indicates the potential mechanism of SIF regulating spermatogenic function in obese rats, and provides a scientific experimental basis for the regulation of biological function of obese male reproductive system by SIF

    Spatio-temporal analysis and simulation on shallow rainfall-induced landslides in China using landslide susceptibility dynamics and rainfall I-D thresholds

    An empirical simulation method to simulate the possible position of shallow rainfall-induced landslides in China has been developed. This study shows that such a simulation may be operated in real-time to highlight those areas that are highly prone to rainfall-induced landslides on the basis of the landslide susceptibility index and the rainfall intensity-duration (I-D) thresholds. First, the study on landslide susceptibility in China is introduced. The entire territory has been classified into five categories, among which high-susceptibility regions (Zone 4- â\u80\u98Highâ\u80\u99 and 5-â\u80\u98Very highâ\u80\u99) account for 4.15% of the total extension of China. Second, rainfall is considered as an external triggering factor that may induce landslide initiation. Real-time satellite-based TMPA 3B42 products may provide real rainfall spatial and temporal patterns, which may be used to derive rainfall duration time and intensity. By using a historical record of 60 significant past landslides, the rainfall I-D equation has been calibrated. The rainfall duration time that may trigger a landslide has resulted between 3 hours and 45 hours. The combination of these two aspects can be exploited to simulate the spatiotemporal distribution of rainfall-induced landslide hazards when rainfall events exceed the rainfall I-D thresholds, where the susceptibility category is â\u80\u98highâ\u80\u99 or â\u80\u98very highâ\u80\u99. This study shows a useful tool to be part of a systematic landslide simulation methodology, potentially providing useful information for a theoretical basis and practical guide for landslide prediction and mitigation throughout China

    Evaluating three satellite-based precipitation products of different spatial resolutions in Shanghai based on upscaling of rain gauge

    Gridded precipitation products have been widely used in scientific literatures, e.g., weather forecasting, hydrological process modelling and disaster simulating. However, it is necessary to evaluate their accuracies before using them for practical purposes. In this study, three mainstream different-spatial-scale daily gridded precipitation data (GPD), including [Global Satellite Mapping of Precipitation–gauge adjusted (GSMAP_Gauge), TRMM 3B42 version-7 (TRMM 3B42V7) and Global Precipitation Climatology Project version 1.2 (GPCP-V1.2)] were systematically evaluated against local rain gauges for their ability to detect precipitation characteristics of Shanghai in 2008 based on upscaling of rain gauge. In general, the gridded precipitation products overestimate precipitation compared to upscaled ground observations (UGOs). However, all of the products have an obvious underestimation on extreme precipitation (>50 mm day −1 ). In contrast, the results show that GSMaP_Gauge has the highest correlation coefficient, TRMM 3B42V7 has the lowest RMSE, and Probability of Detection (POD) in GPCP-V1.2 is better than that in TRMM 3B42V7 and GSMaP_Gauge. As the spatial resolution decreases, the frequency and amount of extreme precipitation events decrease regardless of detection from UGOs and GPD. We also find that those heavy precipitation events detected from TRMM 3B42V7 at 0.25° and 1.0° are significantly underestimated. Overall, we believe that it is necessary to evaluate gridded precipitation products at the urban scale based on upscaling of rain gauge

    Improvement of Colonic Immune Function with Soy Isoflavones in High-Fat Diet-Induced Obese Rats

    Background: The damage to intestinal barrier function plays an important role in the development of obesity and associated diseases. Soy isoflavones are effective natural active components for controlling obesity and reducing the level of blood lipid. Here, we explored whether these effects of soy isoflavones were associated with the intestinal barrier function. Methods and Results: The obese rat models were established by high fat diet feeding. Then, those obese rats were supplemented with soy isoflavones at different doses for 4 weeks. Our results showed that obesity induced the expressions of pro-inflammatory cytokines, decreased the anti-inflammatory cytokine (IL-10) expression, elevated intestinal permeability, altered gut microbiota and exacerbated oxidative damages in colon. The administration of soy isoflavones reversed these changes in obese rats, presenting as the improvement of intestinal immune function and permeability, attenuation of oxidative damage, increase in the fraction of beneficial bacteria producing short-chain fatty acids and short-chain fatty acid production, and reduction in harmful bacteria. Furthermore, soy isoflavones blocked the expressions of TLR4 and NF-κB in the colons of the obese rats. Conclusions: Soy isoflavones could improve obesity through the attenuation of intestinal oxidative stress, recovery of immune and mucosal barrier, as well as re-balance of intestinal gut microbiota
