    Numerical simulation of material flow in AA6082 during friction stir spot welding

    Friction stir spot welding (FSSW) is a new solid state joining technology based on the linear friction stir welding which can be used to replace the conventional resistance spot welding as well as riveting. However, some key problems such as heat transfer and thermoplastic material flow have not yet been studied sufficiently and block the application of this advanced technology. This paper presents the coupled thermo-mechanical viscoplastic finite element formulation based on the character of FSSW. The model was calibrated by comparing temperature history between simulation results and experimental data and subsequently used to investigate the effective strain rate and material flow in joint. The temperature study showed that the simulation and experiment results coincided well with each other thus proofing the correctness of the model. The simulation results showed that the effective strain rate distribution were not uniform. The material close to the pin’s cylindrical surface had a higher effective strain rate than that of the other material. The materials in weld centre mainly experienced a compression process and the other material under the tool experienced both compression and shear process

    Polymorphisms of TGFB1 and VEGF genes and survival of patients with gastric cancer

    <p>Abstract</p> <p>Background</p> <p>Some <it>TGFB1 </it>and <it>VEGF </it>polymorphisms are believed to be functional. Given that these genes are involved in tumor growth and progression including angiogenesis, dissemination, and invasiveness, we hypothesized that these polymorphisms would be associated with survival in patients with gastric cancer.</p> <p>Methods</p> <p>We genotyped <it>TGFB1 </it>-509 C>T, +1869 T>C, and +915 G>C and <it>VEGF </it>-1498T>C, -634G>C, and +936C>T in 167 patients with gastric cancer. Using the Kaplan and Meier method, log-rank tests, and Cox proportional hazard models, we evaluated associations among <it>TGFB1 </it>and <it>VEGF </it>variants with overall, 1-year, and 2-year survival rates.</p> <p>Results</p> <p>Although there were no significant differences in overall survival rates among all polymorphisms tested, patients with <it>TGFB1</it>+915CG and CC genotypes had a poorer 2-year survival (adjusted hazard ratio (HR), 3.06; 95% confidence interval (CI), 1.09–8.62; <it>P </it>= 0.034) than patients with the GG genotype had. In addition, patients heterozygous for <it>VEGF </it>-634CG also had a poorer 1-year survival (adjusted HR, 2.08; 95% CI, 1.03–4.22; <it>P </it>= 0.042) than patients with the -634GG genotype.</p> <p>Conclusion</p> <p>Our study suggested that <it>TGFB1</it>+915CG/CC and <it>VEGF </it>-634CG genotypes may be associated with short-term survival in gastric cancer patients. However, larger studies are needed to verify these findings.</p

    Geochemical characteristics of surface dune sand in the Mu Us Desert, Inner Mongolia, and implications for reconstructing the paleoenvironment

    We present a new dataset from 28 active and 13 stabilizedesemi stabilized dunefields in the Mu Us Desert to report the geochemical macroscale properties as well as reveal the relationship between various chemical ratios and modern climate conditions among different types of Mu Us sandy landscapes. We find that several chemical-weathering indexes, such as CIA, CIW, CPA, and WIP, can be used for the reconstruction of the paleoclimate and servel conditions. One should be cautious in interpreting the weathering intensity using these chemical ratios at a given deposition site when the geochemical background is unknown. This preliminary geochemistry study shows that stabilizedesemi stabilized dunefields, which are influenced by Asian summer monsoon (ASM) precipitation, are analogous to buried paleosols, whereas active dunefields, which are controlled by Asian winter monsoon (AWM) wind, resemble paleo-dune sand. The comparison with the geochemical results from an excellent dune-paleosol succession implies that stronger ASM and AWM periods could have recurred 8-9 times in the Mu Us Desert during the early Holocene.     Study on SiCp/6063Al composites laser welding process with Al75Cu20Ti5 foil interlayer

    This experiment chooses 6063 aluminum matrix composites containing 55~75% SiC particle reinforcing phase as the parent metal and Al75Cu20Ti5 foil as the interlayer. The laser power, defocusing amounts and welding speed are discussed using three important parameters for the joint forming, microstructure and shear strength. The result shows that the weld width is mainly affected by laser power, and the weld penetration is almost determined by the defocusing amount. The welding speed has a certain influence on both weld width and weld penetration. When suitable laser heat input is adopted, superior weld appearance and satisfied shear strength could be obtained. The highest shear strength is 217MPa, with the laser power of 520W, the defocusing amounts of -1mm, and the welding speed of 1.5cm/s

    SATB2 shows different profiles between appendiceal adenocarcinomas ex goblet cell carcinoids and appendiceal/colorectal conventional adenocarcinomas: An immunohistochemical study with comparison to CDX2

    Background: Special AT-rich sequence-binding protein 2 (SATB2) is a novel marker for colorectal adenocarcinomas but little is known about its expression in appendiceal adenocarcinomas. We aim to investigate SATB2 in these tumors and colorectal adenocarcinomas with comparison to CDX2. Methods: Immunohistochemical stains for SATB2 and CDX2 were performed in 49 appendiceal adenocarcinomas (23 conventional, 26 adenocarcinoma ex goblet cell carcinoids (AdexGCCs)) and 57 colorectal adenocarcinomas. Their expression was correlated with tumor differentiation and growth patterns. Results: SATB2 staining was positive in 26/26 (100%) appendiceal AdexGCCs and 15/23 (65%) appendiceal conventional adenocarcinomas (P = 0.001). Their mean percentage of SATB2-positive cells was 93% and 34%, respectively (P \u3c 0.0001). CDX2 staining was seen in 26/26 (100%) AdexGCCs and 22/23 (96%) appendiceal conventional adenocarcinomas (P = 0.4694). SATB2 and CDX2 showed similar staining in AdexGCCs but CDX2 labeled more tumor cells than SATB2 in conventional adenocarcinomas (mean 84% vs. 34%, P \u3c 0.0001). SATB2 and CDX2 staining was seen in 82% (47/57) and 96% (55/57) colorectal adenocarcinomas, respectively (P = 0.01). The mean percentage of cells positive for SATB2 and CDX2 was 48% and 91%, respectively (P \u3c 0.00001). Decreased SATB2 immunoreactivity was associated with non-glandular differentiation particularly signet ring cells in colorectal (P = 0.001) and appendiceal conventional adenocarcinomas (P = 0.04) but not in appendiceal AdexGCCs. Conclusions: SATB2 is a highly sensitive marker for appendiceal AdexGCCs with similar sensitivity as CDX2. In colorectal and appendiceal conventional adenocarcinomas, SATB2 is not as sensitive as CDX2 and its immunoreactivity is dependent on tumor differentiation

    Comparative proteomic and clinicopathological analysis of breast adenoid cystic carcinoma and basal-like triple-negative breast cancer

    BackgroundAdenoid cystic carcinoma (ACC) is a rare type of triple-negative breast cancer that has an indolent clinical behavior. Given the substantial overlapping morphological, immunohistochemical, and molecular features with other basal-like triple-negative breast cancer (BL-TNBC), accurate diagnosis of ACC is crucial for effective clinical treatment. The integrative analysis of the proteome and clinicopathological characteristics may help to distinguish these two neoplasms and provide a deep understanding on biological behaviors and potential target therapy of ACC.MethodsWe applied mass spectrometry-based quantitative proteomics to analyze the protein expression in paired tumor and adjacent normal breast tissue of five ACC and five BL-TNBC. Bioinformatic analyses and the clinicopathological characteristics, including histological features, immunohistochemistry, and FISH results, were also collected to get comprehensive information.ResultsA total of 307 differentially expressed proteins (DEPs) were identified between ACC and BL-TNBC. Clustering analysis of DEPs clearly separated ACC from BL-TNBC. GSEA found downregulation of the immune response of ACC compared with BL-TNBC, which is consistent with the negative PD-L1 expression of ACC. Vesicle-mediated transport was also inhibited, while ECM organization was enriched in ACC. The top upregulated proteins in DEPs were ITGB4, VCAN, and DPT. Moreover, in comparison with normal breast tissue, ACC showed elevated ribosome biogenesis and RNA splicing activity.ConclusionThis study provides evidence that ACC presents a substantially different proteomic profile compared with BL-TNBC and promotes our understanding on the molecular mechanisms and biological processes of ACC, which might be useful for differential diagnosis and anticancer strategy

    Rapid environmental changes in the Lake Qinghai basin during the late Holocene

    The Lake Qinghai Basin is sensitive to global and regional climate change because of its unique geographical location. It is the hotspot for paleoclimate research in East Asia. In this study, we reconstructed the environmental evolution of the Lake Qinghai since ∼9 ka by using a high-resolution peat and fluvial-lacustrine record (Laoyinggou profile) obtained at the foot of Nanshan Mountain. Based on 8 AMS14C dates and lithology, loss on ignition (LOI), total organic matter (TOC), X-ray fluorescence (XRF) core-scanning measurements, ratio of total organic carbon to nitrogen (TOC/TN), and sediment particle sorting coefficients, we show that during the Middle Holocene (∼9–4.4 ka BP) this region was primarily dominated by the Asian summer monsoon, with a consistent, warm, and humid environment. By contrast, during the late Holocene (4.4 ka to present), the climatic context in this area fluctuated dramatically at the millennial scales. The low TOC content, lower TOC/TN ration and strong hydroclimatic indicate six rapid climate change events, which occurred at ∼4.0 ka, ∼3.6 ka, ∼3.2 ka, ∼2.8 ka, ∼2.1 ka, and ∼1.4 ka, all of which coincided to cold episodes in the North Atlantic Ocean

    The VEGF -634G>C promoter polymorphism is associated with risk of gastric cancer

    <p>Abstract</p> <p>Background</p> <p>Both TGF-β1 and VEGF play a critic role in the multiple-step process of tumorgenesis of gastric cancer. Single nucleotide polymorphisms (SNPs) of the <it>TGFB1 </it>and <it>VEGF </it>genes have been associated with risk and progression of many cancers. In this study, we investigated the association between potentially functional SNPs of these two genes and risk of gastric cancer in a US population.</p> <p>Methods</p> <p>The risk associated with genotypes and haplotypes of four <it>TGFB1 </it>SNPs and four <it>VEGF </it>SNPs were determined by multivariate logistic regression analysis in 171 patients with gastric cancer and 353 cancer-free controls frequency-matched by age, sex and ethnicity.</p> <p>Results</p> <p>Compared with the <it>VEGF</it>-634GG genotype, the -634CG genotype and the combined -634CG+CC genotypes were associated with a significantly elevated risk of gastric cancer (adjusted OR = 1.88, 95% CI = 1.24-2.86 and adjusted OR = 1.56, 95% CI = 1.07-2.27, respectively). However, none of other <it>TGFB1 </it>and <it>VEGF </it>SNPs was associated with risk of gastric cancer.</p> <p>Conclusion</p> <p>Our data suggested that the <it>VEGF</it>-634G>C SNP may be a marker for susceptibility to gastric cancer, and this finding needs to be validated in larger studies.</p
