    DNA damage and repair system in spinal cord ischemia

    AbstractBackground and Purpose: Spinal cord ischemia-reperfusion injury may be initiated by a number of mediators, including reactive oxygen species. Recent studies have shown that human MutY homologue (hMYH), human 8-oxo-7,8-dihydrodeoxyguanine (8-oxoG) glycosylase (hOGG1), and human MutS homologue 2 (hMSH2) are important DNA mismatch repair genes. We hypothesized that ischemia-reperfusion injury in spinal cord causes DNA damage manifested by 8-oxoG production and activates the DNA repair system involving hMYH, hOGG1, and hMSH2. Methods: Spinal cords of rabbits were removed at 1, 3, 6, 24, and 48 hours after 30 minutes of infrarenal aortic occlusion. DNA damage was determined with 8-oxoG staining. The expression and localization of DNA repair enzymes, such as hMYH, hOGG1, and hMSH2, were studied with Western blot analysis and immunohistochemical staining. The level of apoptosis was determined with TUNEL study. Activation of caspase-3, an enzyme induced by cellular injury that leads to apoptosis by degrading cellular structural proteins, was also studied. Results: DNA damage monitored with 8-oxoG level was significantly present from 1 hour to 6 hours after reperfusion in gray matter neurons of ischemic spinal cord. The levels of hMYH, hOGG1, and hMSH2 were markedly increased in gray matter neurons at 6 hours after reperfusion. Caspase-3 was also induced at 6 hours to 24 hours after reperfusion in ischemic spinal cord. However, the peak level of TUNEL reactivity was found at 48 hours after reperfusion in spinal cord neurons. Conclusion: This study has shown, for the first time, the rapid expression of DNA damage-repair processes associated with spinal cord ischemia and subsequent reperfusion.     Human ApoE ε2 promotes regulatory mechanisms of bioenergetic and synaptic function in female brain: a focus on V-type H+-ATPase

    Humans possess three major isoforms of the apolipoprotein E (ApoE) gene encoded by three alleles: ApoE ε2 (ApoE2), ApoE ε3 (ApoE3), and ApoE ε4 (ApoE4). It is established that the three ApoE isoforms confer differential susceptibility to Alzheimer’s disease (AD); however, an in-depth molecular understanding of the underlying mechanisms is currently unavailable. In this study, we examined the cortical proteome differences among the three ApoE isoforms using 6-month-old female, human ApoE2, ApoE3, and ApoE4 gene-targeted replacement mice and two-dimensional proteomic analyses. The results reveal that the three ApoE brains differ primarily in two areas: cellular bioenergetics and synaptic transmission. Of particular significance, we show for the first time that the three ApoE brains differentially express a key component of the catalytic domain of the V-type H+-ATPase (Atp6v), a proton pump that mediates the concentration of neurotransmitters into synaptic vesicles and thus is crucial in synaptic transmission. Specifically, our data demonstrate that ApoE2 brain exhibits significantly higher levels of the B subunit of Atp6v (Atp6v1B2) when compared to both ApoE3 and ApoE4 brains, with ApoE4 brain exhibiting the lowest expression. Our additional analyses show that Atp6v1B2 is significantly impacted by aging and AD pathology and the data suggest that Atp6v1B2 deficiency could play a role in the progressive loss of synaptic integrity during early development of AD. Collectively, our findings indicate that human ApoE isoforms differentially modulate regulatory mechanisms of bioenergetic and synaptic function in female brain. A more efficient and robust status in both areas could serve as a potential mechanism contributing to the neuroprotective and cognition-favoring properties associated with the ApoE2 genotype

    Operator radiation dose during trans-hepatic arterial chemoembolization: different patients’ positions via transradial or transfemoral access

    PURPOSEThis study aimed to compare the radiation dose received by the operator among different patients’ positions via transradial access (TRA) or transfemoral access (TFA) during transarterial chemoembolization (TACE) for hepatocellular carcinoma (HCC).METHODSA total of 120 patients with HCC undergoing TACE for the first time between January and November 2019 were randomized into 4 groups with 30 patients in each group. In group A, patients were placed in the foot-first position with the left upper arm abducted, and TACE was performed via the left radial artery. In group B, patients were placed in the conventional headfirst position with the left hand placed at the left groin, and TACE was performed via the left radial artery. In group C, patients were placed in the conventional head-first position, and TACE was performed via the right radial artery. In group D, patients were placed in the conventional head-first position, and TACE was performed via the right femoral artery. Before each procedure, thermoluminescent dosimeters were taped at 7 different body parts of the operator and the radiation dose was measured and collected after the procedure. The normalized radiation dose was also calculated. Procedural parameters included radiation dose, fluoroscopy time (FT), dose–area product (DAP), and air kerma (AK) were recorded. Patients’ demographics, tumor baseline characteristics, radiation dose, and procedural parameters were compared between groups.RESULTSNo significant differences were found in patients’ demographics, tumor baseline characteristics, as well as in total FT, DAP, and AK. However, significant differences were found in the total radiation dose received by the operator and the doses on the pelvic cavity and the right wrist (P < .05). In group C, the radiation doses received on the pelvic cavity, the right wrist, and the total radiation doses were relatively higher. Significant differences were also found in the normalized radiation doses received by the operator on the thyroid, chest, left wrist, right wrist, and pelvic cavity, and the total normalized doses (all P < .05). Similarly, the radiation doses received by the operator at the aforementioned parts in group C were higher, while those in group A were lower.CONCLUSIONNo statistically significant differences were observed in the FT, DAP, and AK in TACE via TRA when patients were placed in different positions. However, TACE via the left TRA, with patients in the feet-first position, reduced the radiation dose received by the operator, thereby reducing the radiation risk

    Performance Zoning of Asphalt Pavement and Performance Grade (PG) of Asphalt Binder in Karamay: A Case Study of Xinjiang, China

    Asphalt binder is a temperature-sensitive material with a performance that is greatly affected by changing climates. Improper selection of asphalt will cause a lot of damage and affect the durability of the road. The establishment of asphalt pavement performance zoning in Xinjiang, a vast area with great temperature differences, will provide a reference for the selection of asphalt suitability, the refinement of pavement design, and the sustainable development of road engineering. In this study, 11 years of temperature data in the Xinjiang region have been collected and analyzed, and 98% reliability of pavement design temperature has been used to draw a performance grading map of asphalt pavement in the Xinjiang region based on the ArcGIS platform. Finally, the Xinjiang region is divided into nine performance zones. At the same time, the performance grades (PG) of five kinds of asphalt binders in Karamay are explored. The result shows that there is little difference in continuous PG span between different matrix asphalt binders; the lower the penetration grade, the better the high-temperature performance, and the worse the low-temperature performance. After adding the SBS modifier, the continuous PG span can be about 20 °C higher than the matrix asphalt. The indoor test of asphalt mixture also shows that SBS-modified asphalt has better durability. All these provide the basis for a reasonable selection of asphalt binders in different areas of Xinjiang

    Evaluation of Comprehensive Development Quality of Port City from the Perspective of Flow Space

    As an important node of transportation network, it is the mainstream trend to improve the development quality of port city and build a high-quality port city. Based on the perspective of flow space, from the two dimensions of "dynamic" and "static" , this paper explored the characteristics and types of comprehensive development quality of 47 coastal port cities, and put forward countermeasures and suggestions. The results showed that: (1) The comprehensive development quality of each port city formed a relatively reasonable gradient distribution, but the driving role of the core port city and the ability of cooperation and mutual assistance among the port cities needed to be improved; (2) The comprehensive development quality of China's port cities can be divided into four types: very high, high, average and low. The number of low type port cities was much higher than that of very high type port cities, and the comprehensive development quality of China's port cities wa still not optimistic; (3) From the internal point of view of different port urban agglomerations, Bohai Rim, Yangtze River Delta and Pearl River Delta port urban agglomerations were concentrated, complete and homogeneous; (4) The key factors affecting the comprehensive development quality of port cities were technology flow, capital flow, logistics and container throughput

    Multi-Stream Convolutional Neural Networks for Rotating Machinery Fault Diagnosis under Noise and Trend Items

    In recent years, rotating machinery fault diagnosis methods based on convolutional neural network have achieved much success. However, in real industrial environments, interfering signals are unavoidable, which may reduce the accuracy of fault diagnosis seriously. Most of the current fault diagnosis methods are of single input type, which may lead to the information contained in the vibration signal not being fully utilized. In this study, theoretical analysis and comprehensive comparative experiments are completed to investigate the time domain input, frequency domain input, and two types of time&ndash;frequency domain input. Based on this, a new fault diagnosis model, named multi-stream convolutional neural network, is developed. The model takes the time domain, frequency domain, and time&ndash;frequency domain images as input, and it automatically fuses the information contained in different inputs. The proposed model is tested based on three public datasets. The experimental results suggested that the model achieved pretty high accuracy under noise and trend items without the help of signal separation algorithms. In addition, the positive implications of multiple inputs and information fusion are analyzed through the visualization of learned features

    Isolation and Characterization of <i>Mycoplasma ovipneumoniae</i> Infecting Goats with Pneumonia in Anhui Province, China

    Mycoplasma ovipneumoniae (M. ovipneumoniae) causes a fatal infection in goats, leading to significant economic losses in the small-ruminant industry worldwide. The present study aimed to characterize the strains of M. ovipneumoniae infecting goats with pneumonia in Anhui Province, China. From November 2021 to January 2023, among 20 flocks, a total of 1320 samples (600 samples of unvaccinated blood, 400 nasal swabs, 200 samples of pleural fluid, and 120 samples of lung tissue) were obtained from goats with typical signs of pneumonia, such as a low growth rate, appetite suppression, increased temperature, discharge from the nose, and a cough. Necropsied goats showed increased pleural fluid, fibrinous pleuropneumonia, and attached localized pleural adhesions. M. ovipneumoniae isolated from the samples were subjected to an indirect hemagglutination test (IHA), PCR amplicon sequencing, phylogenetic analysis, and biochemical identification tests. The overall positivity rate of M. ovipneumoniae was 27.50%. Mycoplasmas were obtained from 80 (20.0%) nasal swabs, 21 (10.5%) pleural fluid samples, and 15 (12.5%) lung samples. PCR amplicon (288 bp) sequencing identified eight strains of M. ovipneumoniae. In a phylogenetic tree, the isolated strains were homologous to the standard strain M. ovipneumoniae Y-98 and most similar to M. ovipneumoniae FJ-SM. Local strains of M. ovipneumoniae were isolated from goats in Anhui province. The identified genomic features and population structure will promote further study of M. ovipneumoniae pathogenesis and could form the basis for vaccine and therapy development

    Effect of In Vitro Culture of Long Shoot Tip on Variant Structure and Titer of Grapevine Viruses

    Shoot tip culture is a very effective approach for studying plant viruses. In this study, we evaluated the numbers, diversity, and titer of grapevine viruses in in vitro grapevine plants after long shoot tip culture. Six virus-infected grapevine cultivars (Cabernet Franc, Cabernet Gernischt, Cabernet Sauvignon, Wink, Victoria, and Merlot) collected from six regions of China were used as the research materials. Approximately 1.5 cm long shoot tips were used for meristem culture. The average survival rate of the six grapevine cultivars was 45.7%. Merlot collected from Beijing showed the highest survival rate (80.0%). Regeneration was not achieved in Cabernet Gernischt collected from Liaoning province and Cabernet Sauvignon from Tianjin due to bacterial and fungal contamination. Virus detection conducted in the surviving regenerated plants showed that the virus infection status, including the viral numbers and the species present in plants grown in vitro, was the same as that in corresponding in vivo plants. Moreover, the analysis of sequence diversity and the mutation frequency in grapevine viruses in vitro indicated that the structure of grapevine viruses was stable in long shoot tip culture after four sub-culture passages. Further, the relative viral titer of in vitro grapevine plants was much higher than that of in vivo plants. These results aid in the investigation of viruses in woody plants
