The Impact of Chronic Stress on Childhood Obesity and the Protective Effects of Parental Warmth
Mexican American children have the highest rates of obesity in the U.S. Mexican American children may experience chronic stress, which has been linked to child obesity. Hair cortisol accumulation has been implicated as a mechanism for this association. Highly sensitive parenting may buffer the harmful effects of chronic stress and hair cortisol on obesity. Thus, the aims of this project were to 1) identify the direct and indirect effects of chronic stress and hair cortisol on children\u27s zBMI and 2) examine the protective effects of parental warmth. This study examined hair cortisol levels, zBMI and chronic stress of children ages 6-10 from low-income Mexican-origin families. Parental warmth was assessed through video-taped family interactions and parent self-report. Chronic stress and hair cortisol levels were not associated, however, hair cortisol was positively related to child zBMI. Parental warmth moderated the relation between chronic stress and zBMI. Thin clients and computers' recycling
This document shows the achieved results in the installation and testing of thin clients' networks (TCN), arranged with obsolete computers. This research topic arises from previous investigations and it is framed in a wider project which includes the use and compatibility of free and/or open source software (FLOSS1). Role of early experience in the development of preference for low-quality food in sheep
Domestic ruminant selectivity induces floristic changes in pasturelands, risking sustainability and limiting the subsequent availability of susceptible plant species. Development of preferences for species of lower nutritional quality may help to overcome those problems. In this study, we tested the hypothesis that early experience of sheep with a low-quality food (LQF) in a nutritional enriched context increases preference for LQF in adulthood. We predicted a higher proportional consumption of LQF in experienced lambs (EL) than in inexperienced lambs (IL) in choice situations involving LQF and alternative foods. Additionally, we determined intake of LQF by EL and IL at different levels of high-quality food (HQF) availability. From 60 to 210 days of age, EL were fed in separated feed bunks mature oat hay (LQF) simultaneously with sunflower meal (SM) and corn grain (CG), whereas IL were fed alfalfa hay (HQF) simultaneously with SM and CG. After exposure, EL and IL were offered LQF in free choice situations involving alternative foods, and also at five levels of HQF availability (100%, 75%, 50%, 25% and 0% of ad libitum intake). Proportional consumption of LQF was lower or similar in EL than IL. Intake of LQF was also lower or similar in EL than IL at all levels of HQF availability, except when the LQF was the only food available. Our results did not support the hypothesis that early experience with a LQF in a nutritional enriched context increases preference for LQF in adulthood. On the contrary, experience with LQF diminished subsequent preference for LQF in adulthood. It is proposed that, in the conditions of our study, continuous comparison between the LQF and the high-quality supplements (CG and SM) during the early exposure period lead to devaluation of LQF by EL through a simultaneous negative contrast effect.Fil: Catanese, Francisco Hernan. Neuroprotection by Exogenous and Endogenous Neuregulin-1 in Mouse Models of Focal Ischemic Stroke.
Identifying novel neuroprotectants that can halt or reverse the neurological effects of stroke is of interest to both clinicians and scientists. We and others previously showed the pre-clinical neuroprotective efficacy of neuregulin-1 (NRG-1) in rats following focal brain ischemia. In this study, we examined neuroprotection by exogenous and endogenous NRG-1 using a mouse model of ischemic stroke. C57BL6 mice were subjected to middle cerebral artery occlusion (MCAO) followed by reperfusion. NRG-1 or vehicle was infused intra-arterially (i.a.) or intravenously (i.v.) after MCAO and before the onset of reperfusion. NRG-1 treatment (16 μg/kg; i.a.) reduced cerebral cortical infarct volume by 72% in mice when delivered post-ischemia. NRG-1 also inhibited neuronal injury as measured by Fluoro Jade B labeling and rescued NeuN immunoreactivity in neurons. Neuroprotection by NRG-1 was also observed in mice when administered i.v. (100 μg/kg) in both male and female mice. We investigated whether endogenous NRG-1 was neuroprotective using male and female heterozygous NRG-1 knockout mice (NRG-1+/-) compared with wild-type mice (WT) littermates. NRG-1+/- and WT mice were subjected to MCAO for 45 min, and infarct size was measured 24 h following MCAO. NRG-1+/- mice displayed a sixfold increase in cortical infarct size compared with WT mice. These results demonstrate that NRG-1 treatment mitigates neuronal damage following cerebral ischemia. We further showed that reduced endogenous NRG-1 results in exacerbated neuronal injury in vivo. These findings suggest that NRG-1 represents a promising therapy to treat stroke in human patients
Discontinuous roughage delivery on digestion, rumen metabolism, feed efficiency and liveweight gain of beef steers fed a concentrate diet
Two experiments were carried out to study the effect of feeding a total mixed ration (TMR) compared to feeding the roughage portion of the diet once every two days and separated of the daily delivered concentrate mixture on dry matter intake, nutrient digestibility, ruminal metabolism, feed efficiency and liveweight gain. In Trial 1, thirty beef steers (Braford and Braford × Criollo; initial BW = 259 ± 27 kg) were used in a 69-d feeding trial. Treatments were: total mixed ration (TMR), and the same proportion of ingredients for the ration but roughage offered once every 2-d and separated from the daily delivered concentrate portion of the diet (REOD). Treatments were arranged in a completely randomized design (three pens/ treatment). In both treatments, daily offered ration had on dry matter basis 90% concentrate and 10% grass hay (Setaria italica). Average daily gain (ADG) did not differ among treatment (1013 vs. 1080 g/d for TMR vs. REOD respectively; SEM = 95 g/d). Dry matter intake was greater in TMR compared to REOD (P < 0.01). Gain to feed ratio tended to be better for REOD than TMR (P = 0.07). In Trial 2, four rumen cannulated steers (Braford) were used in an experiment with a crossover design. Treatments were arranged as a 2*2 factorial design, where the first factor consisted of roughage level (RL): (R14) 14% roughage: 86% concentrate and (R7) 7% roughage: 93% concentrate. The second factor was roughage delivery system (RDS; as it was described for Trial 1): TMR and REOD. There were no RL*RDS interactions for intake and digestion (OM, CP, NDF and starch). Both RL were similar for intake and digestion. Roughage delivery system did not significantly affect intake and digestion of OM, CP, NDF, and starch measured by total fecal collection. Total organic acids (TOA), acetate to propionate ratio (A:P), pH, and rumen ammonia concentrations were not affected by RL and RDS. In conclusion, under the conditions of these trials, steers fed a separated roughage source once every 2-d had similar ADG, and tended to be more efficient compared with TMR. Total tract digestibility and rumen environment traits (pH, VFA, and ammonia) were not affected in response to discontinuous roughage delivery.Fil: Arroquy, Jose Ignacio. A dual function for Pex3p in peroxisome formation and inheritance
Saccharomyces cerevisiae Pex3p has been shown to act at the ER during de novo peroxisome formation. However, its steady state is at the peroxisomal membrane, where its role is debated. Here we show that Pex3p has a dual function: one in peroxisome formation and one in peroxisome segregation. We show that the peroxisome retention factor Inp1p interacts physically with Pex3p in vitro and in vivo, and split-GFP analysis shows that the site of interaction is the peroxisomal membrane. Furthermore, we have generated PEX3 alleles that support peroxisome formation but fail to support recruitment of Inp1p to peroxisomes, and as a consequence are affected in peroxisome segregation. We conclude that Pex3p functions as an anchor for Inp1p at the peroxisomal membrane, and that this function is independent of its role at the ER in peroxisome biogenesis
Proton radiation effects on carrier transport in diamond radiation detectors
Diamond, a highly radiation-resistant material, is considered a nearly ideal material for radiation detection, particularly in high-energy physics. In this study, radiation damage from high-energy proton beams was induced in diamond crystals to determine exposure lifetime in detectors made from this material; the effects were investigated using non-destructive x-ray techniques and through the FLUKA simulation package. Two diamond detectors were irradiated by an 800 MeV proton beam at different fluence rates, and the real-time current response was recorded to observe degradation in the signal over time. It was determined that the proton fluence rate had a significant effect on the device degradation. The detector performance from the irradiated detectors was characterized using x-ray beam-induced current measurements, and the mechanism of proton radiation damage to diamond sensors, especially the radiation effects on carrier transport, was studied. The vacancies generated from proton irradiation were considered the major source of detector degradation by trapping holes and inducing an internal electric field. Simulation results from the FLUKA package revealed an uneven distribution of the radiation-induced vacancies along the beam path, and the corresponding detector signals calculated from the simulation results displayed a good match to the experimental results
