Inbreeding effects on immune response in free-living song sparrows (Melospiza melodia)
The consequences of inbreeding for host immunity to parasitic infection have broad implications for the evolutionary and dynamical impacts of parasites on populations where inbreeding occurs. To rigorously assess the magnitude and the prevalence of inbreeding effects on immunity, multiple components of host immune response should be related to inbreeding coefficient (f) in free-living individuals. We used a pedigreed, free-living population of song sparrows (Melospiza melodia) to test whether individual responses to widely used experimental immune challenges varied consistently with f. The patagial swelling response to phytohaemagglutinin declined markedly with f in both females and males in both 2002 and 2003, although overall inbreeding depression was greater in males. The primary antibody response to tetanus toxoid declined with f in females but not in males in both 2004 and 2005. Primary antibody responses to diphtheria toxoid were low but tended to decline with f in 2004. Overall inbreeding depression did not solely reflect particularly strong immune responses in outbred offspring of immigrant–native pairings or weak responses in highly inbred individuals. These data indicate substantial and apparently sex-specific inbreeding effects on immune response, implying that inbred hosts may be relatively susceptible to parasitic infection to differing degrees in males and females
A Study of the Effectiveness of Water Resources Planning Groups A Final Report
Information and recommendations were developed pertaining to the organization of regional and river basin water planning studies, useful to river basin commissions and others responsible for such studies. The approach that was used included four basic components: (1) conceptualization of a planning model, based on an analysis of planning theory and organizational literature, (2) analysis and review of historical trends in water resources planned, (3) examination and comparative evaluation of 15 river basin planning studies, and (4) case studies of two planning efforts selected from the 15 examined in order to perform a more detailed analysis and to gain information about the informal aspects of the planning process that took place. In connection with the formulation of the planning model, a substantial amount of research effort was committed to the development of a computer simulation of decision-making in the planning or policy formulation process. This programmed simulation is designed to be a useful planning tool for gaining insights into the interrelationships among objectives, alternative courses of action, and political feasibility. The Luminosity Function of Galaxies in Compact Groups
From R-band images of 39 Hickson compact groups (HCGs), we use galaxy counts
to determine a luminosity function extending to M_R=-14.0, approximately two
magnitudes deeper than previous compact group luminosity functions. We find
that a single Schechter function is a poor fit to the data, so we fit a
composite function consisting of separate Schechter functions for the bright
and faint galaxies. The bright end is best fit with M^*=-21.6 and alpha=-0.52
and the faint end with M^*=-16.1 and alpha=-1.17. The decreasing bright end
slope implies a deficit of intermediate luminosity galaxies in our sample of
HCGs and the faint end slope is slightly steeper than that reported for earlier
HCG luminosity functions. Furthermore, luminosity functions of subsets of our
sample reveal more substantial dwarf populations for groups with x-ray halos,
groups with tidal dwarf candidates, and groups with a dominant elliptical or
lenticular galaxy. Collectively, these results support the hypothesis that
within compact groups, the initial dwarf galaxy population is replenished by
"subsequent generations" formed in the tidal debris of giant galaxy

A multicenter prospective phase 2 randomized study of extracorporeal photopheresis for treatment of chronic graft-versus-host disease
Abstract
Chronic graft-versus-host disease (cGVHD) is a major limitation of successful hematopoietic cell transplantation. The safety and efficacy of extracorporeal photopheresis (ECP) for 12 to 24 weeks together with standard therapy was compared with standard therapy alone in patients with cutaneous manifestations of cGVHD that could not be adequately controlled by corticosteroid treatment. The primary efficacy end point was a blinded quantitative comparison of percent change from baseline in Total Skin Score (TSS) of 10 body regions at week 12. Ninety-five patients were randomized to either ECP and standard therapy (n = 48) or standard therapy alone (n = 47). The median percentage improvement in TSS at week 12 was 14.5% for the ECP arm and 8.5% for the control arm (P = .48). The proportion of patients who had at least a 50% reduction in steroid dose and at least a 25% decrease from baseline in TSS was 8.3% in the ECP arm at week 12 and 0% in the control arm (P = .04). The nonblinded investigator assessment of skin complete or partial responses revealed a significant improvement in favor of ECP (P < .001). ECP was generally well tolerated. These results suggest that ECP may have a steroid-sparing effect in the treatment of cGVHD. Clinical trials registered at www.ClinicalTrials.gov as NCT00054613
Fifteen new risk loci for coronary artery disease highlight arterial-wall-specific mechanisms
Coronary artery disease (CAD) is a leading cause of morbidity and mortality worldwide. Although 58 genomic regions have been associated with CAD thus far, most of the heritability is unexplained, indicating that additional susceptibility loci await identification. An efficient discovery strategy may be larger-scale evaluation of promising associations suggested by genome-wide association studies (GWAS). Hence, we genotyped 56,309 participants using a targeted gene array derived from earlier GWAS results and performed meta-analysis of results with 194,427 participants previously genotyped, totaling 88,192 CAD cases and 162,544 controls. We identified 25 new SNP-CAD associations (P < 5 × 10(-8), in fixed-effects meta-analysis) from 15 genomic regions, including SNPs in or near genes involved in cellular adhesion, leukocyte migration and atherosclerosis (PECAM1, rs1867624), coagulation and inflammation (PROCR, rs867186 (p.Ser219Gly)) and vascular smooth muscle cell differentiation (LMOD1, rs2820315). Correlation of these regions with cell-type-specific gene expression and plasma protein levels sheds light on potential disease mechanisms

A high-resolution map of human evolutionary constraint using 29 mammals.
The comparison of related genomes has emerged as a powerful lens for genome interpretation. Here we report the sequencing and comparative analysis of 29 eutherian genomes. We confirm that at least 5.5% of the human genome has undergone purifying selection, and locate constrained elements covering ∼4.2% of the genome. We use evolutionary signatures and comparisons with experimental data sets to suggest candidate functions for ∼60% of constrained bases. These elements reveal a small number of new coding exons, candidate stop codon readthrough events and over 10,000 regions of overlapping synonymous constraint within protein-coding exons. We find 220 candidate RNA structural families, and nearly a million elements overlapping potential promoter, enhancer and insulator regions. We report specific amino acid residues that have undergone positive selection, 280,000 non-coding elements exapted from mobile elements and more than 1,000 primate- and human-accelerated elements. Overlap with disease-associated variants indicates that our findings will be relevant for studies of human biology, health and disease
