    A review of ocean color remote sensing methods and statistical techniques for the detection, mapping and analysis of phytoplankton blooms in coastal and open oceans

    The need for more effective environmental monitoring of the open and coastal ocean has recently led to notable advances in satellite ocean color technology and algorithm research. Satellite ocean color sensors' data are widely used for the detection, mapping and monitoring of phytoplankton blooms because earth observation provides a synoptic view of the ocean, both spatially and temporally. Algal blooms are indicators of marine ecosystem health; thus, their monitoring is a key component of effective management of coastal and oceanic resources. Since the late 1970s, a wide variety of operational ocean color satellite sensors and algorithms have been developed. The comprehensive review presented in this article captures the details of the progress and discusses the advantages and limitations of the algorithms used with the multi-spectral ocean color sensors CZCS, SeaWiFS, MODIS and MERIS. Present challenges include overcoming the severe limitation of these algorithms in coastal waters and refining detection limits in various oceanic and coastal environments. To understand the spatio-temporal patterns of algal blooms and their triggering factors, it is essential to consider the possible effects of environmental parameters, such as water temperature, turbidity, solar radiation and bathymetry. Hence, this review will also discuss the use of statistical techniques and additional datasets derived from ecosystem models or other satellite sensors to characterize further the factors triggering or limiting the development of algal blooms in coastal and open ocean waters

    Variations in the lithium abundances of turn off stars in the globular cluster 47 Tuc

    aims: Our aim is to determine Li abundances in TO stars of the Globular Cluster 47 Tuc and test theories about Li variations among TO stars. method: We make use of high resolution (R~ 43000), high signal-to-noise ratio (S/N=50--70) spectra of 4 turn off (TO) stars obtained with the UVES spectrograph at the 8.2m VLT Kueyen telescope. results: The four stars observed, span the range 1.6<~A(Li)} <~ 2.14, providing a mean A(Li) = 1.84 with a standard deviation of 0.25 dex. When coupled with data of other two TO stars of the cluster, available in the literature, the full range in Li abundances observed in this cluster is 1.6<~A(Li)<~ 2.3. The variation in A(Li) is at least 0.6 dex (0.7 dex considering also the data available in the literature) and the scatter is six times larger than what expected from the observational error. We claim that these variations are real. A(Li) seems to be anti-correlated with A(Na) exactly as observed in NGC 6752. No systematic error in our analysis could produce such an anti-correlation. conclusions: Na production through p captures on 22Ne at temperatures in excess of 3x10^7 K and the contemporary Li destruction could result in this anti-correlation. However such nuclear processing cannot have taken place in the stars themselves, which do not reach such high temperatures, even at their centre. This points towards the processing in a previous generation of stars. The low N/O ratios in the observed stars and the apparent lack of correlation between N an Li abundances, place a strong constraint on the properties of this previous generation. Our results indicate a different behaviour among the Globular Clusters so far studied as far as the abundance patterns are concerned.Comment: recommended for publication in A&A by the managing associate Edito

    A pathway for mitotic chromosome formation

    Mitotic chromosomes fold as compact arrays of chromatin loops. To identify the pathway of mitotic chromosome formation, we combined imaging and Hi-C analysis of synchronous DT40 cell cultures with polymer simulations. Here we show that in prophase, the interphase organization is rapidly lost in a condensin-dependent manner, and arrays of consecutive 60-kilobase (kb) loops are formed. During prometaphase, ~80-kb inner loops are nested within ~400-kb outer loops. The loop array acquires a helical arrangement with consecutive loops emanating from a central spiral staircase condensin scaffold. The size of helical turns progressively increases to ~12 megabases during prometaphase. Acute depletion of condensin I or II shows that nested loops form by differential action of the two condensins, whereas condensin II is required for helical winding

    Seeing is believing: the nocturnal malarial mosquito Anopheles coluzzii responds to visual host-cues when odour indicates a host is nearby

    Background: The immediate aim of our study was to analyse the behaviour of the malarial mosquito Anopheles coluzzii (An. gambiae species complex) near a human host with the ultimate aim of contributing to our fundamental understanding of mosquito host-seeking behaviour and the overall aim of identifying behaviours that could be exploited to enhance sampling and control strategies. Results: Based on 3D video recordings of individual host-seeking females in a laboratory wind-tunnel, we found that despite being a nocturnal species, An. coluzzii is highly responsive to a visually conspicuous object, but only in the presence of host-odour. Female mosquitoes approached and abruptly veered away from a dark object, which suggests attraction to visual cues plays a role in bringing mosquitoes to the source of host odour. It is worth noting that the majority of our recorded flight tracks consisted of highly stereotyped ‘dipping’ sequences near the ground, which have been mentioned in the literature, but never before quantified. Conclusions: Our quantitative analysis of female mosquito flight patterns within ~1.5 m of a host has revealed highly relevant information about responsiveness to visual objects and flight height that could revolutionise the efficacy of sampling traps; the capturing device of a trap should be visually conspicuous and positioned near the ground where the density of host-seeking mosquitoes would be greatest. These characteristics are not universally present in current traps for malarial mosquitoes. The characterisation of a new type of flight pattern that is prevalent in mosquitoes suggests that there is still much that is not fully understood about mosquito flight behaviour

    An exploration of parents’ preferences for foot care in juvenile idiopathic arthritis: a possible role for the discrete choice experiment

    Background: An increased awareness of patients’ and parents’ care preferences regarding foot care is desirable from a clinical perspective as such information may be utilised to optimise care delivery. The aim of this study was to examine parents’ preferences for, and valuations of foot care and foot-related outcomes in juvenile idiopathic arthritis (JIA).&lt;p&gt;&lt;/p&gt; Methods: A discrete choice experiment (DCE) incorporating willingness-to-pay (WTP) questions was conducted by surveying 42 parents of children with JIA who were enrolled in a randomised-controlled trial of multidisciplinary foot care at a single UK paediatric rheumatology outpatients department. Attributes explored were: levels of pain; mobility; ability to perform activities of daily living (ADL); waiting time; referral route; and footwear. The DCE was administered at trial baseline. DCE data were analysed using a multinomial-logit-regression model to estimate preferences and relative importance of attributes of foot care. A stated-preference WTP question was presented to estimate parents’ monetary valuation of health and service improvements.&lt;p&gt;&lt;/p&gt; Results: Every attribute in the DCE was statistically significant (p &#60; 0.01) except that of cost (p = 0.118), suggesting that all attributes, except cost, have an impact on parents’ preferences for foot care for their child. The magnitudes of the coefficients indicate that the strength of preference for each attribute was (in descending order): improved ability to perform ADL, reductions in foot pain, improved mobility, improved ability to wear desired footwear, multidisciplinary foot care route, and reduced waiting time. Parents’ estimated mean annual WTP for a multidisciplinary foot care service was £1,119.05.&lt;p&gt;&lt;/p&gt; Conclusions: In terms of foot care service provision for children with JIA, parents appear to prefer improvements in health outcomes over non-health outcomes and service process attributes. Cost was relatively less important than other attributes suggesting that it does not appear to impact on parents’ preferences.&lt;p&gt;&lt;/p&gt

    Determining sensitivity and specificity of HER2 testing in breast cancer using a tissue micro-array approach

    INTRODUCTION: Overexpression of the human epidermal growth factor receptor 2 (HER2) as a result of HER2 gene amplification is associated with a relatively poor prognosis in breast cancer and is predictive of HER2-targeting therapy response. False-positive rates of up to 20% for HER2 testing have been described. HER2-testing laboratories are therefore encouraged to participate in external quality control schemes in order to improve HER2-testing standardization. METHODS: This study investigated the feasibility of retesting large numbers of invasive breast cancers for HER2 status on tissue micro-array (TMA) as part of a quality control scheme. For this assessment different HER2 testing methods were used including HER2 detecting antibodies SP3, 4B5, Herceptest and mono color silver in situ hybridization (SISH) and dual color SISH. Final HER2 status for each tumor on the TMA was compared to the local testing result for the same tumor. Discordances between these two results were investigated further by staining whole tumor sections. RESULTS: For this study, 1,210 invasive breast carcinomas of patients treated in six hospitals between 2006 and 2008 were evaluated. Results from the three immunohistochemistry (IHC) and two in situ hybridization (ISH) assays performed on the TMAs were compared. The final HER2 status on TMA was determined with SP3, 4B5 and mono color SISH. Concordance between local HER2 test results and TMA retesting was 98.0%. Discordant results between local and TMA retesting were found in 20 tumors (2.0%). False positive HER2 IHC results were identified in 13 (1.3%) tumors; false negative IHC results in seven (0.7%) tumors. CONCLUSIONS: Retesting large volumes of HER2 classified breast carcinomas was found to be feasible and can be reliably performed by staining TMAs with SP3, 4B5 and mono color SISH in combination with full-sized slides for discordant cases. The frequency of false-positive results was lower than previously reported in the literature. This method is now offered to other HER2-testing laboratories
