    Attenuation of Cytotoxic Natural Product DNA Intercalating Agents by Caffeine

    Many anti-tumor drugs function by intercalating into DNA. The xanthine alkaloid caffeine can also intercalate into DNA as well as form π-π molecular complexes with other planar alkaloids and anti-tumor drugs. The presence of caffeine could interfere with the intercalating anti-tumor drug by forming π-π molecular complexes with the drug, thereby blocking the planar aromatic drugs from intercalating into the DNA and ultimately lowering the toxicity of the drug to the cancer cells. The cytotoxic activities of several known DNA intercalators (berberine, camptothecin, chelerythrine, doxorubicin, ellipticine, and sanguinarine) on MCF-7 breast cancer cells, both with and without caffeine present (200 μg/mL) were determined. Significant attenuation of the cytotoxicities by caffeine was found. Computational molecular modeling studies involving the intercalating anti-tumor drugs with caffeine were also carried out using density functional theory (DFT) and the recently developed M06 functional. Relatively strong π–π interaction energies between caffeine and the intercalators were found, suggesting an “interceptor” role of caffeine protecting the DNA from intercalation

    Marketplace Experiences of Individuals with Visual Impairments: Beyond the Americans with Disabilities Act

    The authors explore how consumers with a variety of visual impairments handle day-to-day interactions with service providers, products, and services. Interviews with 21 people with visual impairments reveal that there are considerable individual differences in adaptation strategies, including the degree of independence desired and achieved. Interview themes are explicated by means of a conceptual model of the contexts and interplay of dependence and independence in the lives of people with visual impairments. The model suggests that independence and dependence are not mere opposites on a single dimension; rather, they are domain-specific and complex and are determined by both environmental factors and personal characteristics. It further suggests that some forms of dependency may be as adaptive for many people as is the striving for independence by others. The authors conclude with a discussion of implications for the Americans with Disabilities Act and marketing practice as well as the larger area of consumer vulnerability

    Co-located Retail Clinics and Pharmacies: An Opportunity to Provide More Primary Care.

    This paper proposes that co-located retail clinics (RCs) and community pharmacies can increase opportunities to provide more accessible, affordable, and patient-friendly primary care services in the United States. RCs are small businesses of about 150-250 square feet with a clientele of about 10-30 patients each day and most frequently staffed by nurse practitioners (NPs). Community pharmacies in the U.S. at ~67,000 far outnumber RCs at ~2800, thereby opening substantial opportunity for growth. Community pharmacies and pharmacists have been working to increase on-site clinical services, but progress has been slowed by the relative isolation from other practitioners. An ideal merged facility based on an integrated platform is proposed. NPs and pharmacists could share functions that fulfill documented consumer preferences and still maintain separate practice domains. Potential benefits include a broader inventory of clinical services including laboratory tests, immunizations, patient education, and physical assessment, as well as better patient access, interprofessional training opportunities, and economies related to the use of resources, day-to-day operations, and performance metrics. Challenges include the availability of sufficient, appropriately trained staff; limitations imposed by scope of practice and other laws; forging of collaborative relationships between NPs and pharmacists; and evidence that the merged operations provide economic benefits beyond those of separate enterprises

    Evaluating Utah\u27s Rural Online Initiative: Empowering Rural Communities Through Remote Work

    Utah\u27s rural counties have experienced high levels of unemployment compared to the state\u27s urban counties. Utah State University Extension developed a remote work educational program intended to reduce rural unemployment. We conducted a descriptive study to gather data from May–August 2019 participants (N = 1,025). Our results indicate that short-term outcomes from the course were realized. On average, participants experienced increases in knowledge, improved skills, and positive intentions toward seeking remote employment. We recommended formative evaluation for continuous course improvement and follow-up procedures to measure participants\u27 success in securing remote employment. Extension professionals can plan and evaluate their programs using the framework presented in this article

    A Descriptive Analysis of Remote Work Adoption in the United States

    Advances in information and communications technology have enabled organizations to shift traditional work functions away from place or where work is accomplished to how work is accomplished (i.e., task facilitation). With the rise in remote work, there was a need to describe the adoption process by organizations in the United States. Given that the practice of remote work is considered an innovation, this quantitative study was guided by the theory of Diffusion of Innovations and followed a nonexperimental design with a correlational analysis, collecting cross-sectional data from a sample of organizational leaders in the United States (N = 1,259). Results describe where organizations range in the innovation-decision process of remote work adoption and categorize organizations based on innovativeness. This research demonstrates the role of COVID-19 in precipitating organizations’ rapid implementation of remote work during a pandemic. Findings hold implications for leaders deciding whether to adopt remote work as a formal workplace practice and can assist them in making informed operational decisions. Findings also provide Extension professionals with insights into responding to the social and economic consequences of the widespread adoption of remote work with relevant, research-based educational programming in their local communities

    ABERRANT TESTA SHAPE encodes a KANADI family member, linking polarity determination to separation and growth of Arabidopsis ovule integuments

    The Arabidopsis aberrant testa shape (ats) mutant produces a single integument instead of the two integuments seen in wild-type ovules. Cellular anatomy and patterns of marker gene expression indicate that the single integument results from congenital fusion of the two integuments of the wild type. Isolation of the ATS locus showed it to encode a member of the KANADI (KAN) family of putative transcription factors, previously referred to as KAN4. ATS was expressed at the border between the two integuments at the time of their initiation, with expression later confined to the abaxial layer of the inner integument. In an inner no outer (ino) mutant background, where an outer integument does not form, the ats mutation led to amorphous inner integument growth. The kan1 kan2 double mutant exhibits a similar amorphous growth of the outer integument without affecting inner integument growth. We hypothesize that ATS and KAN1/KAN2 play similar roles in the specification of polarity in the inner and outer integuments, respectively, that parallel the known roles of KAN proteins in promoting abaxial identity during leaf development. INO and other members of the YABBY gene family have been hypothesized to have similar parallel roles in outer integument and leaf development. Together, these two hypotheses lead us to propose a model for normal integument growth that also explains the described mutant phenotypes

    Compared to urban counties, Utah\u27s rural counties experienced high levels of unemployment. Informed by a statewide needs assessment, Utah State University Extension developed a remote work leadership course to equip business leaders with knowledge and skills to create remote jobs as a solution to rural unemployment. This descriptive evaluation study collected data from course participants (N = 62). Findings showed short-term outcomes were achieved; participants experienced increases in knowledge and skills and had more positive intentions toward creating remote jobs and hiring employees from rural counties. Extension professionals can design and evaluate their programs using the framework in this study
