Some Aspects of the Prospective Experimental Use of the Stanford Two-Mile Accelerator
Eleven papers dealing with photon beams from the accelerator, use of hydrogen bubble chambers and spark chambers, a storage ring for 10-Bev muons, muon beams and -p scattering experiments, mass analysis of highenergy accelerator beams, the search for intermediate bosons and heavy leptons, particle yields arising from decay of short-lived intermediate particles, and conjectures on the effects of Regge poles on Drell processes are included. Separate abstracts were prepared for the eleven papers. (D.C.W.
Primakoff effect in eta-photoproduction off protons
We analyse data on forward eta-meson photoproduction off a proton target and
extract the eta to gamma gamma decay width utilizing the Primakoff effect. The
hadronic amplitude that enters into our analysis is strongly constrained
because it is fixed from a global fit to available gamma p to p eta data for
differential cross sections and polarizations. We compare our results with
present information on the two-photon eta-decay from the literature. We provide
predictions for future PrimEx experiments at Jefferson Laboratory in order to
motivate further studies.Comment: 5 pages, 6 figures, gamma-gamma*-eta form factor included, version to

Vaccine protection against simian immunodeficiency virus in monkeys using recombinant gamma-2 herpesvirus
Recombinant strains of replication-competent rhesus monkey rhadinovirus (RRV) were constructed in which strong promoter/enhancer elements were used to drive expression of simian immunodeficiency virus (SIV) Env or Gag or a Rev-Tat-Nef fusion protein. Cultured rhesus monkey fibroblasts infected with each recombinant strain were shown to express the expected protein. Three RRV-negative and two RRV-positive rhesus monkeys were inoculated intravenously with a mixture of these three recombinant RRVs. Expression of SIV Gag was readily detected in lymph node biopsy specimens taken at 3 weeks postimmunization. Impressive anti-SIV cellular immune responses were elicited on the basis of major histocompatibility complex (MHC) tetramer staining and gamma interferon enzyme-linked immunospot (ELISPOT) assays. Responses were much greater in magnitude in the monkeys that were initially RRV negative but were still readily detected in the two monkeys that were naturally infected with RRV at the time of immunization. By 3 weeks postimmunization, responses measured by MHC tetramer staining in the two Mamu-A*01(+) RRV-negative monkeys reached 9.3% and 13.1% of all CD8(+) T cells in peripheral blood to the Gag CM9 epitope and 2.3% and 7.3% of all CD8(+) T cells in peripheral blood to the Tat SL8 epitope. Virus-specific CD8(+) T cell responses persisted at high levels up to the time of challenge at 18 weeks postimmunization, and responding cells maintained an effector memory phenotype. Despite the ability of the RRVenv recombinant to express high levels of Env in cultured cells, and despite the appearance of strong anti-RRV antibody responses in immunized monkeys, anti-Env antibody responses were below our ability to detect them. Immunized monkeys, together with three unimmunized controls, were challenged intravenously with 10 monkey infectious doses of SIVmac239. All five immunized monkeys and all three controls became infected with SIV, but peak viral loads were 1.2 to 3.0 log(10) units lower and chronic-phase viral loads were 1.0 to 3.0 log(10) units lower in immunized animals than the geometric mean of unimmunized controls. These differences were statistically significant. Anti-Env antibody responses following challenge indicated an anamnestic response in the vaccinated monkeys. These findings further demonstrate the potential of recombinant herpesviruses as preventive vaccines for AIDS. We hypothesize that this live, replication-competent, persistent herpesvirus vector could match, or come close to matching, live attenuated strains of SIV in the degree of protection if the difficulty with elicitation of anti-Env antibody responses can be overcome
Analysis of recent eta photoproduction data
Recent data on eta-meson photoproduction off a proton target in the energy
range 2 < sqrt{s} < 3 GeV are analyzed with regard to their overall
consistency. Results from the ELSA and CLAS measurements are compared with
predictions of a Regge model whose reaction amplitude was fixed via a global
fit to pre-2000 measurements of differential cross sections and polarization
observables for gamma p -> eta p at higher energies. We find that all recent
experimental results on differential cross sections for eta-meson
photoproduction are in good agreement with each other, except for the CLAS data
from 2009. However, the latter can be made consistent with the other data at
the expense of introducing an energy-dependent renormalization factor. We point
out that there indications in the data for a possible excitation of baryon
resonances with masses around 2.1 and 2.4 GeV.Comment: 13 pages, 13 figures; small modifications in the text, some figures

A tudor domain protein SPINDLIN1 interacts with the mRNA-binding protein SERBP1 and is involved in mouse oocyte meiotic resumption
Mammalian oocytes are arrested at prophase I of meiosis, and resume meiosis prior to ovulation. Coordination of meiotic arrest and resumption is partly dependent on the post-transcriptional regulation of maternal transcripts. Here, we report that, SPINDLIN1 (SPIN1), a maternal protein containing Tudor-like domains, interacts with a known mRNA-binding protein SERBP1, and is involved in regulating maternal transcripts to control meiotic resumption. Mouse oocytes deficient for Spin1 undergo normal folliculogenesis, but are defective in resuming meiosis. SPIN1, via its Tudor-like domain, forms a ribonucleoprotein complex with SERBP1, and regulating mRNA stability and/or translation. The mRNA for the cAMP-degrading enzyme, PDE3A, is reduced in Spin1 mutant oocytes, possibly contributing to meiotic arrest. Our study demonstrates that Spin1 regulates maternal transcripts post-transcriptionally and is involved in meiotic resumption.Ting Gang Chew, Anne Peaston, Ai Khim Lim, Chanchao Lorthongpanich, Barbara B. Endocytosis of the Anthrax Toxin Is Mediated by Clathrin, Actin and Unconventional Adaptors
The anthrax toxin is a tripartite toxin, where the two enzymatic subunits require the third subunit, the protective antigen (PA), to interact with cells and be escorted to their cytoplasmic targets. PA binds to cells via one of two receptors, TEM8 and CMG2. Interestingly, the toxin times and triggers its own endocytosis, in particular through the heptamerization of PA. Here we show that PA triggers the ubiquitination of its receptors in a β-arrestin-dependent manner and that this step is required for clathrin-mediated endocytosis. In addition, we find that endocytosis is dependent on the heterotetrameric adaptor AP-1 but not the more conventional AP-2. Finally, we show that endocytosis of PA is strongly dependent on actin. Unexpectedly, actin was also found to be essential for efficient heptamerization of PA, but only when bound to one of its 2 receptors, TEM8, due to the active organization of TEM8 into actin-dependent domains. Endocytic pathways are highly modular systems. Here we identify some of the key players that allow efficient heptamerization of PA and subsequent ubiquitin-dependent, clathrin-mediated endocytosis of the anthrax toxin
