    Cholesterol in condensed and fluid phosphatidylcholine monolayers studied by epifluorescence microscopy

    Epifluorescence microscopy was used to investigate the effect of cholesterol on monolayers of dipalmitoylphosphatidylcholine (DPPC) and 1 -palmitoyl-2-oleoyl phosphatidylcholine (POPC) at 21 +/- 2 degrees C using 1 mol% 1-palmitoyl-2-[12-[(7-nitro-2–1, 3-benzoxadizole-4-yl)amino]dodecanoyl]phosphatidylcholine (NBD-PC) as a fluorophore. Up to 30 mol% cholesterol in DPPC monolayers decreased the amounts of probe-excluded liquid-condensed (LC) phase at all surface pressures (pi), but did not effect the monolayers of POPC, which remained in the liquid-expanded (LE) phase at all pi. At low pi (2–5 mN/m), 10 mol% or more cholesterol in DPPC induced a lateral phase separation into dark probe-excluded and light probe-rich regions. In POPC monolayers, phase separation was observed at low pi when > or =40 mol% or more cholesterol was present. The lateral phase separation observed with increased cholesterol concentrations in these lipid monolayers may be a result of the segregation of cholesterol-rich domains in ordered fluid phases that preferentially exclude the fluorescent probe. With increasing pi, monolayers could be transformed from a heterogeneous dark and light appearance into a homogeneous fluorescent phase, in a manner that was dependent on pi and cholesterol content. The packing density of the acyl chains may be a determinant in the interaction of cholesterol with phosphatidylcholine (PC), because the transformations in monolayer surface texture were observed in phospholipid (PL)/sterol mixtures having similar molecular areas. At high pi (41 mN/m), elongated crystal-like structures were observed in monolayers containing 80–100 mol% cholesterol, and these structures grew in size when the monolayers were compressed after collapse. As a significant next step, the University has taken the strategic decision to develop a repository for educational materials. In developing EdShare at Southampton, we are promoting a cultural shift to a more open and collaborative approach to scholarship as well as research.Successful implementation in such a context requires a lightweight and very simple approach to sharing content facilitated by web 2.0 functionality.<br/

    Condensing Momentum Modes in 2-d 0A String Theory with Flux

    We use a combination of conformal perturbation theory techniques and matrix model results to study the effects of perturbing by momentum modes two dimensional type 0A strings with non-vanishing Ramond-Ramond (RR) flux. In the limit of large RR flux (equivalently, mu=0) we find an explicit analytic form of the genus zero partition function in terms of the RR flux qq and the momentum modes coupling constant alpha. The analyticity of the partition function enables us to go beyond the perturbative regime and, for alpha>> q, obtain the partition function in a background corresponding to the momentum modes condensation. For momenta such that 0<p<2 we find no obstruction to condensing the momentum modes in the phase diagram of the partition function.Comment: 22 page

    Integrable Deformations of c^=1\hat{c}=1 Strings in Flux Backgrounds

    We study d=2 0A string theory perturbed by tachyon momentum modes in backgrounds with non-trivial tachyon condensate and Ramond-Ramond (RR) flux. In the matrix model description, we uncover a complexified Toda lattice hierarchy constrained by a pair of novel holomorphic string equations. We solve these constraints in the classical limit for general RR flux and tachyon condensate. Due to the non-holomorphic nature of the tachyon perturbations, the transcendental equations which we derive for the string susceptibility are manifestly non-holomorphic.     Primary ciliary dyskinesia: Recent advances in diagnostics, genetics, and characterization of clinical disease

    Primary ciliary dyskinesia (PCD) is a genetically heterogeneous recessive disorder of motile cilia that leads to oto-sino-pulmonary diseases and organ laterality defects in approximately 50% of cases. The estimated incidence of PCD is approximately 1 per 15,000 births, but the prevalence of PCD is difficult to determine, primarily because of limitations in diagnostic methods that focus on testing ciliary ultrastructure and function. Diagnostic capabilities have recently benefitted from (1) documentation of low nasal nitric oxide production in PCD and (2) discovery of biallelic mutations in multiple PCD-causing genes. The use of these complementary diagnostic approaches shows that at least 30% of patients with PCD have normal ciliary ultrastructure. More accurate identification of patients with PCD has also allowed definition of a strong clinical phenotype, which includes neonatal respiratory distress in >80% of cases, daily nasal congestion and wet cough starting soon after birth, and early development of recurrent/chronic middle-ear and sinus disease. Recent studies, using advanced imaging and pulmonary physiologic assessments, clearly demonstrate early onset of lung disease in PCD, with abnormal air flow mechanics by age 6-8 years that is similar to cystic fibrosis, and age-dependent onset of bronchiectasis. The treatment ofPCDis not standardized, and there are no validated PCD-specific therapies. Most patients with PCD receive suboptimal management, which should include airway clearance, regular surveillance of pulmonary function and respiratory microbiology, and use of antibiotics targeted to pathogens. The PCD Foundation is developing a network of clinical centers, which should improve diagnosis and management of PCD

    Effects of old landfills on groundwater quality. Phase 2, investigation of the Thriplow landfill 1996–1997

    Disused sand and gravel excavations overlying the major Chalk aquifer at Thriplow in Cambridgeshire have been filled with domestic waste in two phases. One area (Phase 1) was filled between 1957–77 with little compaction of the refuse and was left uncapped, while Phase 2 was deposited between 1981–87 and capped with clay. Aerial photography and surface resistivity surveys indicate that the site geometry is complex, with several phases of landfilling into excavations of differing depths. Drilling through the waste indicates that leachate production and waste stabilisation proceed at different rates in capped and uncapped landfills. Analysis of leachate obtained by centrifugation or squeezing appears to give more insight into the pollution potential than do leach tests with distilled water. The Biological Methane Potential (BMP) of the waste appears to be related to the quantity of decomposable material but the chemical oxygen demand (COD) values are distorted by the presence of reduced metals. Too few boreholes have been drilled to define the leachate source in terms of its spatial distribution and little is known of how its composition has changed with time. However, hydraulic conductivity measurements on the landfill caps suggest that it is sufficiently permeable for all rainfall to potentially infiltrate the waste. Boreholes outside the landfill penetrate the Upper and Lower Chalk, and identify the Melbourn Rock and underlying Plenus Marls at the junction of the two formations about 20 m below ground level (bgl). Surface resistivity surveys using the BGS RESCAN system, confirm aerial photographs of the extent of the landfill and also suggest that leachate has migrated beyond the base of the landfill. Evidence of leachate migration in pre-existing screened boreholes completed above and below the Plenus Marls suggests that leachate is flowing above the Plenus Marls. Hydraulic head measurements whilst drilling a borehole to the base of the lower Chalk approx. 70 m bgl revealed the potential for upward groundwater flow through the Plenus Marls. Thus, previously-drilled boreholes penetrating the Plenus Marls are expected to recharge upwards into the shallow aquifer above the Plenus Marls diluting any leachate in the upper aquifer and distorting the flow regime. Several of these boreholes have subsequently been modified to stem the flow across the Plenus Marls. One borehole down-gradient to the west of the site revealed a large thickness of drift composed of both sand and clay rich material. This suggests the existence of a buried channel, the hydrogeological significance of which has yet to be assessed. Groundwater chemistry appears to be influenced by three major factors. (a) the landfill leachate (b) the composition of shallow groundwater in the top 10 m of the Chalk, and (c) the composition of water from the Lower Chalk. Limited groundwater monitoring data appear to display a cyclic variation in chloride concentration. The origin for this is not clear but it may correlate with cyclic variations in groundwater levels when the water table rises into the waste. Cyclic flushing of the landfill may release leachate into the aquifer giving rise to pulses of chloride. Alternatively changes in chloride may arise by the changing direction of groundwater flow which as yet has not been assessed. A conceptual hydrogeological model in which flow is limited to above the Plenus Marls has been used to develop a more appropriate groundwater flow and solute transport model. However, the model lacks data on aquifer properties, on contaminant inputs concentrations, fluxes and spatial variations, and there is a paucity of monitoring data for calibration. Nonetheless preliminary transport modelling using an equivalent porous medium approach shows that an effective porosity of about 5% best fits the regional data. Since this is much less than the total porosity of about 40% for the Chalk, it would appear that only part of the Chalk is available for flow but that matrix diffusion could play an important role in leachate attenuation. Discrete fracture modelling using the FRACTRAN code has allowed some scoping to be made of the hydraulic properties of the aquifer by comparison with chloride hydrographs, but these again need to be better conditioned by in-situ measurement of fracture distributions and transmissivities. A number of additional activities are required to improve the understanding of flow and contaminant transport at the site. These include better spatial definition of the waste distribution, improved data on the hydraulic properties of the Chalk aquifer, and the use of automatic monitoring to record temporal changes in groundwater chemistry and groundwater levels

    Starting, building and sustaining a program of research in emergency medicine in Canada

    Objective: To develop pragmatic recommendations for starting, building and sustaining a program of research in emergency medicine (EM) in Canada at sites with limited infrastructure and/or prior research experience. Methods: At the direction of the Canadian Association of Emergency Physicians (CAEP) academic section, we assembled an expert panel of 10 EM researchers with experience building programs of research. Using a modified Delphi approach, our panel developed initial recommendations for (1) starting, (2) building, and (3) sustaining a program of research in EM. These recommendations were peer-reviewed by emergency physicians and researchers from each of the panelist’s home institutions and tested for face and construct validity, as well as ease of comprehension. The recommendations were then iteratively revised based on feedback and suggestions from peer review and amended again after being presented at the 2020 CAEP academic symposium. Results: Our panel created 15 pragmatic recommendations for those intending to start (formal research training, find mentors, local support, develop a niche, start small), build (funding, build a team, collaborate, publish, expect failure) and sustain (become a mentor, obtain leadership roles, lead national studies, gain influence, prioritize wellness) a program of EM research in centers without an established research culture. Additionally, we suggest four recommendations for department leads aiming to foster a program of research within their departments. Conclusion: These recommendations serve as guidance for centres wanting to establish a program of research in EM

    Lagrangian evolution of global strings

    We establish a method to trace the Lagrangian evolution of extended objects consisting of a multicomponent scalar field in terms of a numerical calculation of field equations in three dimensional Eulerian meshes. We apply our method to the cosmological evolution of global strings and evaluate the energy density, peculiar velocity, Lorentz factor, formation rate of loops, and emission rate of Nambu-Goldstone (NG) bosons. We confirm the scaling behavior with a number of long strings per horizon volume smaller than the case of local strings by a factor of ∼\sim 10. The strategy and the method established here are applicable to a variety of fields in physics.Comment: 5 pages, 2 figure
